Structure­based subcellular pharmacokinetics – experimental and computational elucidation of factors determining the rates and equilibria of the trans­bilayer transport, creation of structure­ based models of subcellular pharmacokinetics containing trans­bilayer transport, metabolism, binding to major proteins (albumin, collagen, G­actin), hydrolysis and renal excretion.

Development of methods for structure­based prediction of binding affinities by a combination of molecular dynamics and quantum mechanics.

Development of methods for a comparison of flexible binding sites of related enzymes and receptors with the aim to discern the features of ligands leading to selectivity.

Development of selective inhibitors of matrix metalloproteinases with optimized distribution using the approaches of structure­based subcellular pharmacokinetics.

Development of web­based tools for the improvement of education in pharmacokinetics and drug design

I moved from Case Western Reserve University and joined NDSU on March 1, 2004. This transition resulted in a significant loss of manpower and time. J. Present Scholarly Interests and Activities

My research seeks to test the hypothesis that lipid­lipid interactions are by nature weak, cooperative, and transient so as to create an on/off switch for signaling events at the membrane surface. The use of lipids offers the possibility of cooperativity in the assembly of protein complexes on the membrane and of fine­tuning the combinations of proteins that will subsequently be assembled.

To both broach and answer this question, as to whether lipid diversity exists as a means to alter cellular outcomes by modulating a membrane­involving signaling event, requires merging of the disciplines of membrane biophysics, thermodynamics, protein purification and modification, statistical thermodynamics (binding partition functions), and pharmacology.

I seek to address questions regarding the underlying principles that dictate how membrane domains form, how the bilayers couple, how membrane domains are regulated, and application of the answers to systems of neuropharmacological importance (the synaptotagmins).

Last year I was team teaching in the Concept Pharmacy Laboratory, but this year will only be teaching in the lab occasionally. Job Description change effective July 2005 shifts teaching responsibilities to more experiential at an Adult Medicine Site in rural ND.
