HyPerforma G3Lite Controller

The Thermo Scientific™ HyPerforma™ G3Lite™ Controller is an open architecture control system that can be integrated with any single-use bioreactor such as the Thermo Scientific™ HyPerforma™ Single-Use Bioreactor (S.U.B.) (Available in sizes ranging from 50 L to 2,000 L). The system consists of a control tower that leverages intelligent transmitters, mass flow controllers (MFCs), pumps, sensors, and Thermo Scientific™ TruBio™ software that facilitates easy, reliable, and repeatable process development and commercial cell culture processes. HyPerforma G3Lite Controllers are fully self-contained, movable units that can be operated as stand-alone (for 1 vessel) or networked for multiple vessels. They are engineered to optimize capital cost for use in non-GMP and cGMP-certified production facilities.

Thermo Scientifi c™ TruBio™ confi gurable upstream software provides easy-to-use process control. It can be confi gured by the user through a visual interface, eliminating the need to know process control programming. TruBio software is powered by an Emerson™ DeltaV™ system, has been developed according to GAMP™ 5 methods, and conforms to regulatory requirements for use in cGMP-compliant processes. Developed for use with HyPerforma controllers, TruBio software is designed to support easy-scaling tech transfer, reducing the time required for validation services. It enables the user to build sophisticated process control strategies without knowledge of DeltaV programming. It also provides the fl exibility to incorporate a wide range of cell culture, fermentor, or mixing vessels and manage multiple data streams from several unit operations.

The HyPerforma S.U.B. is one of the most widely used S.U.B.s designed for mammalian cell culture. We fi rst launched the HyPerforma S.U.B. in 2006, and updated the design in 2013, while maintaining the robust, top-down, stirred-tank mixing system.

The S.U.B. has been widely adopted in process development and clinical trials as well as in bioproduction of cGMP cell culture. A superior solution, our bioreactors can be easily integrated withany controller available in the market today.

The Thermo Scientifi c™ TruFlow™ gas MFC is designed to work with all of the HyPerforma control systems. Its compact assembly provides up to six standard mass fl ow controllers, three associated solenoid valves, and three gas outlet bulkheads from which to direct the gases. When connected, the TruFlow gas MFC is instantly recognized by TruBio software to provide precise control of gas fl ow, without requiring any confi guration, even at extremely low fl ow rates.

A range of single-use sensors have been designed specifically to optimize cell growth in single-use bioreactors. Thermo Scientific™ TruFluor™ and TruTorr™ sensors measure the four fundamental process parameters: pH, dissolved oxygen, temperature, and headspace pressure. Thermo Scientific sensors seal reliably into an insertion port, so that process sterility is guaranteed.

Thermo Scientific single-use sensors implement a patented optical design using only USP Class VI materials to optimize performance while maximizing reliability. All sensors are precalibrated and provide full traceability information using a gamma irradiation–resistant chip. The chip includes all calibration information, which is transferred to the transmitter when connected with a cable.

Using a HyPerforma G3 controller with TruFluor single-use sensors provides accurate measurements that enable the bioreactors to create a suitable cell culture environment.

Single-use sensorsMinimize contamination riskOur single-use sensors can be inserted in the S.U.B. BPC ports and irradiated with the bag in order to both preserve and guarantee the sterile barrier.

Biocompatibility test and gamma irradiation qualificationAll of our single-use sensor materials have undergone USP Class VI and gamma irradiation (up to 50 kGy) testing. The results are available by request for auditing purposes. Certificates are also available for all wetted materials.

Blade transmittersThermo Scientifi c™ TruFluor pH and DO blades are panel-mounted instruments that have plug-and-play capabilities with any TruFluor pH or DO reader. The blades provide the output from the sensors to the controller in a digital format.

TruFluor and TruTorr blades are rail mounted in a controller and do not have a display or keypad. All blades require a 2-point calibration for accuracy specs and provide data from sheaths containing a chip, and detailed diagnostics to the bioreactor control system.

A TruFluor loop consists of a disposable sheath welded in the BPC, an optical reader, a cable, and a transmitter (blade).

Readers minimize drift and photodegradationThermo Scientifi c™ TruFluor pH and TruFluor DO optical smart readers utilize a patented design that minimizes photodegradation of the active sensing element. The design has been optimized to provide accurate in situmeasurement of pH and dissolved oxygen using phase fl uorometric detection in real time. The temperature measurement leverages a 316L stainless steel thermal plate embedded in the sheath, and provides accurate results that can also be used as a process variable. TruFluor DO sensors are based on oxygen quenching of fl uorescence, whereas TruFluor pH sensors are based on dual lifetime referencing. There is no detectable photoinduced drift for either of these sensors in a 3-week period of sampling at 2-second intervals.
