Remarks

KATRINA must be connected to an electrical installation that conforms to all applicable safety standards, national or international prescriptions. In order to ensure user protection against electric shock, KATRINA protective earth conductor must be properly connected to the building earth. Do not operate a centrifuge until all necessary safety measures have been taken. KATRINA centrifuge is designed in such a way that its use does not expose the operator and his environment to any danger. This protection can only be guaranteed if the centrifuge is used in accordance with all the instructions in this manual. The rotors and buckets must be removed from use in the event of a trace of mechanical defect or corrosion. These elements have a lifetime, engraved on their visible part: To maintain good safety conditions, it is imperative to replace them when the recommended duration is reached. KATRINA can only be serviced by certified personal, informed and trained by AFI Centrifuge.

Warranty

AFI Centrifuge warrants this device does not have any assembly or material defects when it leaves the factory. In the event of a defect appearing during proper use, AFI Centrifuge undertakes to repair the device for a period of two years from the date of delivery, for malfunctions concerning the following parts:

- Motor - Electronic control center - Tachometer board. - Mains filter board - Interconnection board - Lid lock motor supply board

All parts claimed defective shall be returned, properly identified to the company at its factory, charges prepaid. Factory installed equipment or accessories are warranted only to the extent guaranteed by the original manufacturer, and this warranty shall not apply to any portion of the equipment modified by the user. Claims under this warranty should be directed to AFI Centrifuge. Setting forth in detail the nature of the defect, the date of the initial installation and the serial and model number of the equipment.

This warranty shall not apply to any AFI Centrifuge product or part thereof which has been subject to misuse, abuse, accident, shipping damage, improper installation or service, or damage by fire, or flood. If the serial number of this product is altered, removed or defaced as to be illegible, the Warranty shall be null and void in its entirety.

The warranty is for the sole benefit of the original purchaser and is not assignable or transferable. Prior to returning any item, for any reason, contact AFI Centrifuge for a Return Authorization Number. This number must accompany all returns. Any product shipped to AFI Centrifuge without this number will be returned refused shipment or collect freight. Please carefully read user manual instruction before using the centrifuge.

Forbbiden use

KATRINA is not designed for the following uses and environments: The following are considered prohibited: • Flammable materials, • Explosive substances, • Materials that can react causing a danger, • Toxic materials • Radioactive materials • Contaminated material not contained in an aerosol-tight container. The use of this type of container is understood to be in a biosecurity system in accordance with national and international indications, they must not constitute the only protection of workers and the environment during the handling of pathogenic microorganisms. (IEC / EN61010-2-020) See for example the "Laboratory biosafety manual" guide by the World Health Organization. • Rotors and rotating accessories having exceeded their maximum service life. • The rotors and rotating accessories not being maintained in accordance with the instructions in this manual, or showing signs of wear and / or corrosion. • Rotors and rotating accessories not validated by AFI Centrifuge • Explosive environments • Radioactive environments

Decontamination

Depending on its use, and for example if potentially pathogenic substances have been spilled inside the bowl, KATRINA can expose service personal to a biological danger. In this case, or in case of any doubt, cleaning and decontamination are necessary as a preventative measure before the centrifuge, rotors and other accessories are maintained, repaired or transferred. The manufacturers can provide a form allowing the users to document the realization of this treatment. (IEC / EN 61010-2-020) Before any intervention, in order to guarantee the operator's safety, please refer to the laboratory decontamination protocol, and carefully apply the decontamination prescriptions. During decontamination operations, the required personal protective equipment must be worn. Radioactive materials are prohibited and cannot be centrifuged by KATRINA.

1.9 Electronic Control Center / Block The electrical block, or Control Center (sometimes named “Ogive”), includes a touch screen, a Start/Stop button, a lid opening button, as well as the microcontroller electronic board, and the electrical power supply electronic board. There is no provision for changing the elements composing the Control Center. It is a complete separate part. In case of defective element, a standard exchange of the complete Control Center must be considered, which facilitates the restoration of the centrifuge.

Touch Screen

This display is based on a TFT type display its size is 5.7”. It offers 640x480 pixels, and 16 Million colors. It is also equipped with LED backlighting for bright background with low power consumption. Just in front of the display, a resistive touch pad sense for user finger press. It provides access to many features when using the interface. The display and display card unit are located in the upper part of the Control Center.

Start/Stop Button

The control button placed under the screen allows for controlling the startup and interruption of a centrifugation cycle.

Lid Opening Button

The lid opening is controlled by the push button on the right side of the Control Center.

Electronic microcontroller Board

The electronic microcontroller board is located under the display card. The data for controlling the various parts are stored in this board, in the form of software, also marked “Microcontroller fw” The connection for the update of this software is below the Control Center, via the connection kit part no. 71199004. The procedure is described in the “Software programming annex” annex. This board communicates with: • The tachometer board • Llid position sensors for detecting the lid position, • Touchpad display module • The programming connector • The temperature probe • The Start / Stop control button on the membrane • The lid opening button on the right side of the Control Center.

Electronic Power Board

This board is directly connected to the mains and provides supply for: • The microcontroller board • The display card • The centrifuge motor • The refrigeration group • The lock motor It also transmits the power accumulated during rotor braking to the load shedding resistance.

Lid locking motor supply board

This board supplies 24Vdc to the lid lock moto reducer when opening or closing is required. It receives 230V at mains input, this voltage is provided by the Electronic Control Center only when required.

The board and especially the molded transformer are protected by a 2ATT 250V 5x20 fuse cartridge.

1.10 Transformer A toroid transformer is connected to the power board and provides 3 different voltages identified by the following colors:

1.11 Locking / unlocking system

Description :

1) Lid latches. The left one is equipped with 2 magnets, the right one is not.

2) Latch approach sensor (Magnetic switch). 3) Closed position sensor (Magnetic switch). It detects the

latch postion. 4) Crankshaft: This is the moving part driven by the motor

reducer. 5) Locked position sensor (microswitch contact). It detects

the deadbolt when the device is locked. 6) Motor reducerHook. They are part of the "welded shaft"

& deadbolt. This welded shaft is moved through the crankshaft, which moves in the oblong guide zone of the deadbolt.

7) Open position sensor. (microswitch contact). It detects when the device is unlocked.

8) Alignment marks of deadbolt and crankshaft. When aligned, the device is in locked position.

A safety system prevents the motoreducer from being energized if a speed signal is detected. This prevents the lid from

being open is rotor is rotating.

1.12 Motor Three phase induction motor, variable frequency U/F MLI driver. An integrated thermoswitch automatically stops the rotation cycle when windings temperature reaches 130°C, in this case “ERROR 04” is displayed.

1.13 Speed sensor KATRINA Speed sensor is a Hall Effect magnetic sensor, located under the motor. Six (6) magnets with alternating poles are arranged around a rotating disc. The sensor measures the frequency of passage of the magnets, and microcontroller determines the speed.

1.14 Tachometer board Speed sensor signal is filtered, shaped, and sent back to the Electronic Control Center, as well as the “rotating” signal. This “rotating” signal is activated when a speed signal is detected and used for the lid safety feature as described in §1.11 1) Connection to the tachometer sensor

1.15 Temperature probe Located on the lid internal panel, making it closed to the buckets once the lid is closed. Measurements are only taken during rotation. Sensor is an encapsulated PT500 class A element. During a centrifugation cycle, KATRINA displays the sample temperature, computed from the air temperature measurement, and assuming the samples enter the centrifuge at the program target temperature. Display accuracy is warranted only if samples enter the centrifuge at the target temperature. KATRINA is designed to maintain the desired temperature during a rotation cycle, this also can be achieved only if samples enters the centrifuge at the correct temperature.

1.16 Imbalance sensor In the case of an incorrect balancing of the buckets, oscillations and vibrations can reach an unacceptable level. Vibrations are permanently measured by an electronic accelerometer located inside the Electronic control center. If the measured level becomes higher than the acceptable threshold, centrifugation cycle is automatically interrupted and ERROR n°2 is displayed. The default detection threshold is: 50 gr This threshold is adjustable according to the media on which the centrifuge rests, in order for the imbalance sensor to always be triggered with a minimum imbalance of 50 gr see Section 3.3 for the procedure to adjust the imbalance detection threshold.

1.17 Gas spring A double gas cyclinder / spring system allows the opening of the lid when it is released by the lock. The spring allows a better relaxation at the opening start. The cylinder should open the lid without causing rebound at the end of travel.

1.18 Vibrations absorption / suspension KATRINA is designed with an optimized suspension system allowing vibrations and parasitic movement’s absorption in normal operational conditions, provided the rotor load is well balanced around the rotation axis. This suspension is made of 5 shock absorbers at the base of the motor ballast and 2 foam disks located at the top and bottom of the motor. A good balancing of the accessory is a key point to achieve satisfying separation quality.

1.19 Refrigeration System: KATRINA refrigeration system allows to maintain the samples temperature at their initial temperature while rotating. Temperature target can be set between -10°C and 40°C. Maximum performance is achieved for a 20 +/-2°C ambient temperature:

4°C can be maintained +/-2°C at 4300 rpm with windshield lid,

4°C can be maintained +/-2°c at 3800 rpm without windshield lid. These results can be achieved only if samples enter the centrifuge at the target temperature. The centrifuge air inlet / outlet must be free of any obstacle.

2 Proper installation

2.1 Centrifuge location

In order to install KATRINA, it is necessary to have a

free space of 1406 x 1560 mm.

The soil at this location must be able to withstand

twice the weight of the machine in use (about 350kg),

i.e. 700 kg,

and allow a vibration-less operation.These vibrations

could degrade the results of the centrifugation.

A free space of 30 cm all around the centrifuge must

absolutelybe provided, according to standard IEC

61010-2-020:

No potentially dangerous material, no person, nor any

object must be inside the free space during

centrifugation.

Leave a minimum space of 60 cm between 2 adjacent centrifuges. This space guarantees the required clearance area for

each machine and sufficient ventilation between them.

The reserved space must not be exposed to direct sunlight, nor placed near a heat source. The room will be well

ventilated to avoid overheating and electrical overconsumption.

2.2 Leveling Before being used, KATRINA must be properly leveled using its 4 adjustable feet. The centrifuge accessory must rotate in a perfectly horizontal plan, please see the leveling procedure available in the annex. A bad leveling can lead to:

drastically shorten lifetime of the motor and suspension system

poor centrifugation quality in reason of a bad behavior of the suspension system

2.3 Electrical installation

The centrifuge mains supply must be 230V / 50 Hz, single phase, in accordance with the manufacturer's plate on the

back of the device.

- 21Arms can be drawn for less than 1mn during centrifuges acceleration under heaviest load and with maximal

acceleration rate.

KATRINA must be directly connected to the distribution board and protected by its own protection device.

This device must be identified as the centrifuges' disconnector device and easily accessible in the immediate vicinity of

the centrifuge.

The protective earth conductor must imperatively be connected to the installation earth.

The centrifuge's power supply must be protected by a safety device ensuring the automatic cut-off in the event of an

3 Troubleshooting

3.1 Troubleshooting

Problem Cause Solution

No display: the screen remains black.

The lid has been opened with the manual opening key, Or centrifuge supply was turned off before the completion of locking / unlocking The lid motor supply fuse has blown

Press the open button, the locking motor starts in order to reset the system mechanical position. Once done, the lid can normally be closed. Turn Centrifuge OFF Check for any short cut between the board and the motor, check there is no mechanical blocking point preventing the locking or unlocking movement to complete. Once this points checked and eventually fixed, replace the fuse. (2A TT, 250V, 5x20mm)

Displayed speed value is decreasing, display is blinking.

After a power outage while centrifuging, the cycle is interrupted. The rotor slows down in free wheel.

The programmed temperature setpoint is inappropriate

Change speed / temperature pair of the program, which is not compatible with normal use.

The initial loading temperature is too high.

Place the samples before centrifugation in an environment between 4 and 37°C, for 1h.

The cooling unit no longer works.

Check the operation of the chiller with the menu “Auto-Diagnostic “ (Startup and performance)

The temperature probe is defective

In the probe calibration menu, the value displayed is different from the value measured by the probe, with a gap > 2°C.

The difference between the air temperature and the over temperature alarm set point is greater than the set value.

Check the value of the temperature alarm threshold. Change program settings: speed and/or temperature.

04 MOTOR OVERTEMPERATURE

The motor temperature is too high.

A stop in “freewheel” mode occurs. Wait 30 minutes (software reset) before opening the lid.

Activate the air conditioning system of the room to obtain a lower ambient temperature.

The motor is damaged Change the motor.

Motor temperature sensor connection malfunction

Change the motor.

05 ERROR MICROCONTACTOR ON LOCKING MECHANISM

One of the micro contacts / magnetic sensor is damaged or not working. There is a bad connection to the micro contacts.

Contact a certified technician.

06 MOTOR CONTACTOR INCONSISTENT STATE: NOT SEEN OPEN

The contactor does not work. The auxiliary contact is not working. The connection of the auxiliary micro contact is not good.

At centrifuge start, motor contactor is seen closed when it should be open. Contact a Certified technician.

07 MOTOR CONTACTOR INCONSISTENT STATE: NOT SEEN CLOSED

The motor contactor doesn’t work. The open position switch was released after the lid closed, The contactor coil is not energized. The coil of the micro contactor is not working properly.

At centrifuge start, motor contactor is seen open when it should be closed. Contact a certified technician.

09 NO SPEED SIGNAL AT START

The speed sensor is working improperly. The tachometer board is working improperly.

The speed signal was lost during more than 6s during rotation. Wait for centrifuge full stop, acknowledge the error, cycle power and try again. If Error 10 is still detected call a certified technician

4 Cleaning and lubrication

4.1 Cleaning After proper decontamination of the centrifuge, Open the lid, remove the buckets and place them on a stable support.

Buckets and liners

- For each bucket, clean the trunnions bearing area (rotation axis) with a dry cloth

- Clean the bottom of the bucket compartments

- Clean the bottom of the liner compartments and their structural grooves.

Bowl and bowl seal :

- Remove the rotor from the motor axis, using the provided wrench.

- Remove any liquid that could be present at the bottom of the centrifugation bowl, clean with water

and soap or an hydroalcoholic solution or a quaternary ammonium solution.

- NEVER USER ANY CHLORINATED PRODUCT (bleach, quaternary ammonium chloride…).

- Dry well after cleaning.

- Do not use any fluffy cloth.

- Reassemble the rotor on the motor shaft with the provided wrench.

4.2 Lubrication

Rotor

- Clean the 12 trunnions (bucket rotation axis)

- lay a thin layer of alimentary grease on the top of each trunnion

Buckets

- The trunnion bearing area does not require any lubrication, it is always lubricated by the trunnions

themselves.

Driving shaft:

- Remove the rotor from the motor axis, using the provided wrench.

- Clean any durt on the shaft and under the rotor.

- lay a thin layer of alimentary grease on the motor shaft

- Reassemble the rotor on the motor shaft with the provided wrench.

5.2 Setting the temperature probe Access the temperature probe calibration

menu.

The upper value “Probe Temp.” corresponds to the physical value measured by the temperature sensor, located in the lid. The lower value “Displayed Temp.” is the value displayed on the screen. This is the temperature value of the samples in the buckets, during a centrifugation cycle. A correction of + /- 2°C is possible in the case of deviation with the temperature of the sample in the buckets.

Press the “-” or “+” keys to adjust the value.

Confirm by pressing ok.

5.3 Unbalancing Sensitivity Detection The centrifuge has a reaction to the load balancing faults which may be different depending on the media on which the centrifuge is placed. A calibration of the imbalance sensitivity is therefore necessary during installation. Required equipment: Use the reference balancing kit 71122002.

Access the unbalance sensitivity calibration menu.

Equip a rotor swing-out and its 4 buckets in the centrifuge. Remove the inserts of the buckets.

Low threshold

1. Place the round 50 gram weight in one of the buckets and close the lid,

2. Press the button “50gr” 3. The rotor accelerates until it stabilizes. 4. The centrifuge shakes, then the rotor

slows down. 5. The low threshold is then determined. It

appears under the button “50 gr”. (e.g. 480)

6. Open the lid and remove the 50 gram weight.

High threshold

7. Place the 100 gram weight in the same bucket.

8. Proceed in the same way as for the 100 gram weight.

9. The low threshold is then determined. (e.g. “520”)

10. Open the lid and remove the 25 gram weight.

11. Press on the Next button to average the setting. (e.g. [480+520]/2 = 500 ) The average value is displayed. By default, the factory setting value is 500.

Check The following steps are compulsory. High threshold:

Do not adjust this threshold with a weight greater than 100 gr. This would cause a breach of warranty. The appliance has been dimensioned for a maximum threshold of 100gr.

12. Place the 100 gr weights in a bucket. 13. Start the cycle. 2000 rpm / 1 minute / 9

Acceleration / Braking 9 14. The centrifuge must stop during the

acceleration, and display the message ERROR1: The 100 gr imbalance is not tolerated. Repeat the High threshold check 3 times: ERROR1 must appear each time.

Low threshold:

15. Leave only a weight of 50 gr in a bucket. 16. Start the cycle. 2000 rpm / 1 minute / 9

Acceleration / Braking 9 17. The centrifuge should achieve the

desired speed and then slow down and stop without the following error message appearing: The 50 gr imbalance must be tolerated. Repeat the Low threshold check 3 times: Each cycle should take place normally.

5.4 Setting the lock Lid lock is an electromechanical system including moving parts. Keep your fingers / hand away from moving parts when operating it.

Locking steps : :

The lid is lowered manually. The left strike of the cover (1) is detected by the approach sensor (2). The motor reducer (6) starts. The open position sensor (8) changes to state 0.

The hooks (7) drive the 2 latches down. When the left strike is detected, the closed position sensor of the strike (3) goes to state 1.

The locked position sensor of the lock (5) goes to state 1. The motor reducer stops: The markings (9) of the crankshaft and the deadbolt are aligned: the lock is in the locked position. The lock is in closed position when the marks on the deadbolt and the crankshaft are aligned.

Unlocking steps : The steps are in the reverse order of the locking operation. The motor reducer rotates in the opposite direction.

Setting the Time Delay of the lock

Access the unbalance sensitivity calibration menu.

Locking timer: The motor reducer timer when closing. Adjust with the “-” and “+” buttons up to the position described above, in Section 3.4

Unlocking timer: Motor reducer time delay when closing. Adjust with the “-” and “+” buttons until the bolt is completely clear at the opening, in order to let the lid strike plate escape freely.

Close the lid. Access the Auto-Diagnostic menu Press “Contactor”: the button turns green and the motor contactor is energized. The return of information “contactor state” turns green meaning the contactor and its auxiliary contact work properly.

"Compressor" Manual start of the compressor and the chiller fan Performance: By closing the lid and activating the compressor (pressing on compressor), after 5 minutes, ice forms on the sides of the bowl. Coolant is always present in the circuit.

Lid mot. The locking system motor is energized. The mechanism runs without stopping. After this test

Operate the lock with the manual opening key: ¼ turn clockwise, then ¼ turn anti-clockwise”. When the “unlock” micro contact engages, change of state.

"Lid approach" Passes in green checkmark when the lid is lowered (magnet detection).

"Open button" Press the button to open lid: becomes green

"Start button" Press Start/Stop: Change of state.

"Motor Temp." Disconnect the motor over temperature sensor, (intermediate connector in front of the motor contactor) Check ths state change

5.6 Information update Caution: This information concerns the serial numbers of the electronic boards fitted on the centrifuge. In case of changing an electronic board, these values should be changed:

5.7 Counters modification The values are used to determine the age of the device and the rotating accessories. Changing values should ONLY be done in the case of an exchange of the control center. The values to be set must be identical to those recorded in the faulty control center. It is therefore necessary to record the values before removing the faulty control center. If the values are lost monitoring the life of the rotor will not be correct. A rotor may therefore be used beyond their maximum lifespan. This causes a DANGEROUS situation for users and machine. In case of inability to access data from the defective control center these values can be retrieved by the factory.

5.8 Overtemperature alarm menu This alarm can protect the samples subjected to excessive heating of the air with respect to the set temperature. In case of triggering of the alarm, the cycle is aborted. ERROR 3 message is displayed and buzzer starts.

Adjust the desired value. The default is + 10° C. The value is adjustable between + 5° C to + 20° C. Example: for a 10 ° C setpoint when the over temperature threshold is set at 5 ° C, the alarm will sound if the air temperature is> 15 ° C

Note: If the sample temperature exceeds 43 ° C, but also if the temperature of the air inside the bowl becomes higher than 50 ° C, then the cycle is aborted with the message ERROR 3.

5.9 Firmware update KATRINA centrifuge is controlled by 2 firmwares: • A firmware application called “Display firmware”, loaded into the screen board. It manages a HMI (Human Machine Interface), and visual screens. • A firmware application called “Microcontroller firmware”, loaded into the microcontroller board. It defines the behavior, and the interactions between the different parts of the centrifuge. It is necessary to use a specific programming cable, supplied with reference 71199004 “Software Programming Kit”. See detailed update process in annex.

5.10 Factory Reset After loading a new version of centrifuge firmware, it may be necessary to perform a factory reset, which returns the device to factory configuration. This deletes the following data: - Lock setting (a default is recorded) - The imbalance calibration - User programs - The error counters (reset to 0) - The serial numbers of the unit and electronic cards

5.11 Clearing counters The counters are used to determine the age of the device, and the rotating accessories. In case of resetting these counters, monitoring the life of the rotor will not be correct. A rotor may therefore be used beyond their maximum lifespan. This causes DANGER for users and machine. This menu is primarily used in the factory.

7 Functional checks

7.1 Mechanical Safety

Hinge: (lid) Check the tightness of the screws, and the integrity of the hinge by moving side-to-side. There should be no movement other than that of the hinge rotation.

Control Center: The Control Center must be secured by a nut located near the switch and fixed to the body of the

centrifuge. The lower protection (translucent plastic panel) must be screwed in place under the SUBD9 programing port.

Accessories: Check that the maximum cycle number marked on the free rotor is far superior to the value

recorded on the counter “SWING OUT CYCLES COUNT”

Conduct a visual inspection:

1) The surface of the rotors should be:

Circular in shape on the periphery of the trunions

7.2 Electrical installation – Protective earth In order to protect user against electric shock, KATRINA protective earth conductor must always be properly connected to the installation. This must be checked at least once a year, using a certified continuity tester.

7.3 Speed Check Required equipment: Calibrated tachometer and reflective adhesive optical tachometer. Place a reflective strip on the central part of the rotor. Remove the central porthole located in the middle of the lid. Close the lid. Start a cycle at the desired speed When the target speed is reached, place the speedometer on the central porthole. The speed displayed by the measurement device must be equal to that displayed on the screen, within a tolerance of ± 10 rpm If this is not the case, check that the reflective tape is well placed in the central part of the rotor, and firmly hold the optical tachometer, without movement during the measurement.

7.4 Checking the Timer Required equipment: Calibrated stopwatch. Define a centrifugation cycle of the desired duration (e.g.: 10 min, 3000 rpm, acceleration 9, braking 9). Start the cycle by pressing start. To start the centrifuge stopwatch, operate the stopwatch. Stop the stopwatch at the end of the programmed time. Check that the centrifuge starts to brake well.

7.5 Checking the Temperature This test must be done starting from ambient temperature for both centrifuge and accessory. If you are using a windshielded accessory, windshield lid shall not be used. - A reference thermometer is required. - If you are using RHD6 accessory, Tubes with water already cooled down to 4°C are required - If you are using RX6-1000 accessory, water already cooled to 4°C will be poured directly in the buckets - A test program (or routine) must be created with the following parameters

o Speed: 2700RPM for RX6-1000, 2300RPM with RHD6

o Temperature 4°

o Duration 15mn

1. Insert your accessory, close the centrifuge’s lid, and start the precool cycle. 2. Wait for the cycle’s end and full stop 3. Open the lid and, without waiting, insert the tubes already preconditioned to 4°C, close the lid, pour 200ml

of water in each buckets if you are using RX6-1000 accessory. 4. Start the previously defined test cycle 5. Wait for the end of the cycle and centrifuge’s full stop. 6. Open the lid and check the tubes water temperature, it must be 4°C +/-2°C
