2.Poor Precision in ELISA experiment
ELISA failure can be attributed to many factors. Many unexpected results can be avoided if the manual is
read and fully understood before starting the experiment. On identifying assay failure, you can follow tips below.
1. Check the expiration dates of the kit, ensure that all the reagents have been stored as indicated in the
manual.
2. Check for signs of instability or deterioration in reagent solutions, such as precipitation or discoloration and
so on.
3. Substrate reagent should be colorless.
4. Use clean plastic disposable pipettes, tips, and containers for reagent preparation and storage. Avoid
cross-contamination of reagents by changing pipette tips between addition of each standard, sample and reagent.
5. Ensure that specified incubation times and temperatures have been adhered to.
6. Ensure that no substitution of kit reagents has occurred.
7. It is recommended that samples and standards be run in duplicate to improve accuracy.

useful in the event that strips become loose while decanting. Secondly, empty the liquid in the plate by decanting. Fill each well with the volume of Wash Buffer specified in the kit manual. Set a timer for the recommended time and let the Wash Buffer soak in the wells. Decant the wells by inverting the plate and rapping it against clean paper toweling. Repeat this process as directed in the kit manual. After the last decanting, remove any remaining Wash Buffer by inverting the plate and blotting it dry by rapping it firmly against clean paper toweling on a hard surface. Do not let the wells to sit dry. Proceed immediately to the next step in the kit manual.

Ensure that each cannulae is dispensing and aspirating properly. Set the volume of Wash Buffer in each well and soaking time for each washing. Then put the plate in. Ensure that no Wash Buffer is left in the wells after aspiration. Do not over-aspirate the wells by allowing the aspiration apparatus to sit in the wells after the aspiration.

12. Calculation of ELISA results
The key to repeated accuracy in pipetting is a consistent technique. Consistent speed and smoothness is necessary while pipetting. Avoid sudden motions when drawing or dispensing liquid.
1. Check to be sure that the pipette tip is fitted properly on the end of the pipette and seated with adequate pressure
2. Change pipette tips between each sample or reagent addition. 3. Do not pipette a lower volume than the manufacturer’s suggestion. This will decrease accuracy and
reproducibility.4. Some liquids will have a tendency to cling to the inside or outside surfaces of the pipette tips. To avoid
variations in the volumes drawn or dispensed caused by this surface tension, pre-rinse the pipette tip with the sample or reagent. This will improve accuracy.
5. If the reagent to be pipetted is viscous, wait a moment after drawing the liquid to allow the volume to reach equilibrium before removing the pipette tip from the reservoir.
6. After drawing the liquid into the tip, remove the tip from the solution and wipe the outside of the tip with a lint-free tissue.
7. If an air bubble appears in the tip while drawing the liquid, dispense the liquid back into the container and re-pipette the liquid at a slower rate. If the air bubble appears a second time, discard the pipette tip and replace with a new tip.
8. If foaming occurs in the tip while drawing up the liquid, it may be necessary to hold the pipette at a slight angle and draw the liquid slower.
9. Use separate reservoirs for each reagent.
1. ELISA samples are always tested in duplicate or in triplicate, then average the absorbance values for each set of standards and samples. Duplicates should be within 20% of the mean.
2. Subtract the average value of zero standard OD (optical density).3. Standard curve.Create a standard curve by plotting the mean absorbance for each standard on the x-axis against the
concentration on the y-axis and draw a best fit curve through the points on the graph. We suggest the suitable software should be used for this, such as CurveExpert 1.3. The software is recommended that various methods (e.g. linear, semi-log, log/log, 4 parameter logistics) be tried to see which curve best fits the data. One way to determine if the curve is to backfit the standard curve O.D. values. Treat standards as unknowns and interpolate the O.D. values from the standard curve. They should be close to the expected value (+/- 10%). Use the curve that gives the best correlation (r) value and backfit.
Step by step as shown:
Washing the plate too rapidly or too slowly, incomplete washing or aspirating, and letting the wells to sit dry are all factors that will affect the precision of the ELISA.
