ARemotelyOperatedVehicle(ROV)isatetheredunderwaterrobotthatallowsthevehicle'soperatortoremaininasafeenvironmentwhiletheequipment(ROV)worksinthepotentiallyhazardousenvironmentbelow.Theopensource ROV,(OpenROV),offersaversatilemechanismformaritimesurveying.
TheanalysisofnucleicacidmaterialfrommarinesystemsiscurrentlycommonlyachievedviaNiskin samplingandpreservationofnucleicacidsisdelayeduntilarrivalatthesurface.ThisprojectaimstodevelopaninsitunucleicacidsamplerwhichpreservesthepayloadandthereforexpandsthecapabilityoftheOpenROV platform.
Focuswillbeonthecollectionofmarinebiomassfromseawaterandmetabolically‘freezing’theRNAprofileinsitu topreventdegradationandstress-induceddevianceuponbringingthesampletothesurface,withinasimpleandcheapdeviceattachedtoanROV.WewillaimtousethisdevicetomonitormicrobialcommunitiesinrealtimeusingbasicmicroscopylighttoselectivelytargetSpecificwaterbodiesforsampling.
ThenucleicacidpreservationdevicewillbeconstructedusingvariouscomponentssuchasArduinoandRaspberrypiforprogrammablecontrolandaperistalticpumpforsamplingcoupledwithsensorsmonitoringdepth,temperatureandsalinity.
