ILAB UTRECHT
Co-creationiLab (innovation Lab) Utrecht is a fully equipped andstaffed laboratory in which companies have accessto research infrastructure, students and scientists ina co-creation setting. The first iLab Utrecht facilitywill be accommodated by the University of AppliedSciences; we expect to open multiple iLabs in otherlocations at the Utrecht Science Park that provideadditional facilities and fields of expertise. Thecollaboration with research and education institutesenables companies to work in a researchenvironment without the need for heavyinvestments to establish their own labinfrastructure. Furthermore, the access to studentsenables companies to identify talented studentsand offer an internship or even an employmentcontract. For startup companies that are still in theprocess of attracting financing for their venture, wecan develop tailor-made business models forpublic-private partnerships. iLab Utrecht is part ofan ecosystem strategy that will organize not only
access to infrastructure and human capital but alsoaims to facilitate start-ups in other ways, byproviding incubation and acceleration programs.
National networkiLab Utrecht in its current form has expertise andfacilities in specialized chemical analysis, chemicaltechnology, molecular & cell biology, microbiologyand alternative-to-animal-tests. However, iLabUtrecht is part of a national network of iLabs that isestablished by the Top Sector Chemistry. Currentlythere are 10 locations where an iLab is eitherestablished or under development. Hence, yourresearch question can be passed on to a wideexpert network in (bio)chemistry.

Life Sciences IncubatorThe newly build Life Sciences Incubator can hostinnovative startup and scale up companies that areactive in the life sciences and require both officeand lab space for their activities. The Life SciencesIncubator is hosted by Stichting Incubator, a legalentity fully owned by Utrecht University.
The facility comprises 4000 m2 effective floor areawith offices, laboratory spaces and general facilitiessuch as reception and meeting facilities. Thebuilding enables a community of life sciencesresearch companies that can benefit fromsubsidized rent and startup support. The building isdirectly connected to an accelerator building for lifesciences companies.
The Life Sciences Incubator is the first laboratorybuilding in The Netherlands with a sustainabilityrating of BREEAM Outstanding*. The Life SciencesIncubatoris supported by the European RegionalDevelopment Fund.
Accelerator buildingResearch companies and corporate researchorganizations that require larger volumes of officeand lab space can be hosted in the separateaccelerator building. In addition, Utrecht SciencePark is preparing for the development of a newaccelerator building that can also host companiesrequiring larger volumes.

NMR LARGE SCALE FACILITY
Novel research concepts are being developed andapplied to complex biomolecular systems includingprotein-protein and protein-nucleic acid complexesor membrane proteins. Additional research isconducted on heterogeneous systems such ashydrogels, biomaterials, liposomes and cellularmembranes. The facility is part of the Bijvoet Centerfor Biomolecular Research.
Specifications main equipmentAs a national and European large scale facility,state-of-the-art instrumentation for NMRexperiments on soluble molecules orheterogeneous preparations (solid-state NMR) isavailable. Currently, NMR fields range from 500 MHzto 950 MHz (solution NMR) and from 400 MHz to950 MHz (solid-state NMR). In addition, two solid-state spectrometers (400 and 800 MHz) areequipped with gyrotrons for DNP (dynamic nuclearpolarisation), which provides highly increased

With our experienced staff, state-of-the-artequipment and advanced software we are able tohandle a large variation of chemical compounds. Inacademic collaborations we contribute significantlyto the preparation of scientific publications.Examples of our structural studies include modelsystems that mimic catalytic sites in proteins orsynthetic catalysts to be used in the cleanproduction of desired pharmaceuticals. Forenantiopure compounds we are able to determinethe absolute configuration, even in the absence ofheavy atoms.
Our modern equipment for protein crystallographyenables us to perform screening experiments forprotein crystallization and to optimize the results.Co-crystallization and soaking of pharmaceuticalfragments is possible. The protein structuredetermination can be performed in-house.

NPC PROTEINS AT WORKPROTEOMICS FACILITY
The Proteins At Work facilities of the NetherlandsProteomics Center provides access to state-of-the-artproteomics technology, equipment and expertise toresearchers in the Life Sciences from academia andindustry. The core facility at Utrecht University is anexpert center for state-of-the-art proteomics analysisthat develops and implements innovative massspectrometric methods for efficient and detailedcharacterization of biomolecules in relation to theirbiological function. The facility offers massspectrometry based proteomics technology forapplications ranging from single proteinidentifications to the analysis of complex, whole-celllysate, samples and determination ofposttranslational modifications.
Equipment and accessThe instrumentation of the facility consists of a widerange of HPLC coupled high-end mass spectro-meters, including state-of-the-art technologies such
as high-resolution Orbitrap mass spectrometers withCID, HCD and ETD fragmentation capability. For datastorage and analysis, the facility has dedicatedcomputational clusters. The facility offers its servicesthrough research collaborations with scientists in theNetherlands, Europe and world-wide.
NPC Out-stationThe proteomics facility at the University MedicalCenter Utrecht is an out-station of the Proteins atWork initiative of the Netherlands ProteomicsCenter. The location of the facility within theDepartment Molecular Cancer Research, part of theCenter for Molecular Medicine, ensures a strongbackground in biochemistry and molecular biology.Together with the high-end equipment, this allowsfor tailor made innovative approaches to aidfundamental, translational and clinical research intoproteins and their modifications relevant for healthand disease. Questions that are addressed rangefrom single protein identification to proteomequantification. The facility offers its services primarilyto researchers from the UMCU and its partners.

ABLAB NATIVE PROTEINMASS SPECTROMETRYSERVICES
AbLab is the service unit that operates within theUtrecht University, providing state of the art nativemass spectrometry services.
Native MSNative mass spectrometry (native MS) allows themass spectrometric analysis of intact proteins andprotein complexes under their native folded stateproviding a wide range of information, such asstructural information, complex stoichiometry andsample composition and purity. The key feature thatmakes native MS unique is the retention ofmolecular non-covalent interactions during theionization process and the subsequent massanalysis. Over the years, the progressivedevelopment of this analytical technique has beenpushing the limits of this technique, today allowingthe measurement of large, intact protein complexes.
EquipmentAblab can provide access to analysis of proteinsamples using dedicated, modified (Q)-ToF massspectrometers analysis, all providing information onthe structure and composition of the native proteincomplexes. The presence of an Orbitrap analyzerspecifically developed for intact protein analysisenables in-depth analyses of post-translationalmodifications (PTMs) directly at the intact proteinlevel.

LIPIDOMICS FACILITY
The Lipidomics Facility houses chromatographic-and mass spectrometric equipment that isspecialized in the analysis of lipids. It providescomplete molecular analyses of a broad spectrumof (phospho)lipids (lipid fingerprinting) in thepicomole range as well as analysis of unusual lipids(lipid biomarkers).
High-throughputHigh throughput technologies have been developedallowing automated lipid analysis of 300samples/day. In combination with molecularimaging and modern information technology thiswill revolutionize our understanding of the complexinteraction networks in a functioning cell and howlipids together with genes and proteins determinecellular functions in health and disease.
EquipmentThe available technologies include MS basedlipidomics techniques (three mass spectrometers,all equipped with ion sources (turbo-)ESI, nano-ESI,and APCI, autosamplers, micropumps, UV detectors,UPLC and variable wavelength Fluorescencedetector), assays for lipid metabolism and lipid fluxusing heavy isotope labeled precursors, lipidisolation and separation techniques.

ServicesWe are committed to support your Next GenerationSequencing (NGS)-based research by providingexpertise input for your experimental design, choiceof sequencing platform, advice on samplepreparation protocols, and help with data analysis.For experienced users, we provide sequencingcapacity and routine bioinformatic data analysis.The Utrecht Sequencing Facility (USF) is embeddedwithin an active research environment aiming at theuse of NGS technology beyond the current state-of-the-art. Extensive expertise is available with wet-laband bioinformatic aspects of whole genomesequencing (WGS), whole exome sequencing (WES),targeted sequencing, RNA sequencing (RNA-seq),amplicon sequencing (Ampli-seq), ChromatinImmunoprecipitation sequencing (ChIP-seq), andother.

UTRECHT CENTRE FOR MEDICAL METABOLOMICS
MetabolomicsIn metabolomics, one of the platform sciences ofthe various ‘omics’, the small-molecule complement(metabolome) of a biological system is identifiedand quantified. These diverse molecules, calledmetabolites, underpin the ongoing biochemicalreactions in a living cell, tissue and organism.Measurement of some individual metabolites iscommon practice for diagnostic purposes. Progressin analytical science and informatics allows theanalysis of a large part of the metabolomesimultaneously, providing not only information onthe individual metabolites but on the whole offunctioning biochemical networks.
The Utrecht Centre for Medical Metabolomics(UCMM) is a joint initiative of the department ofMolecular Cancer Research) and the department of
Metabolic Diagnostics of the UMC Utrecht. It unitesmetabolic knowledge from both the diagnosticlaboratory and more fundamental metabolicresearch. The UCMM is specialized in the study ofmetabolites in individual cells, tissues, and bodyfluids in context of biological perturbations andnutritional status. The centre has a variety of massspectrometry setups and develops and validatesboth targeted and untargeted assays that willbecome accessible as a fee-for-service forresearchers. At this point several metabolite assaysfor biomedical research can be offered.

New Immunization methodWe offer a new and very efficient cellularimmunization method. With this method,antibodies can be raised against low immunogenicmembrane proteins, such as multispanningmembrane proteins, conformational epitopes andwe can generate agonistic and antagonistic MABs.On request we can make MABs against solubleproteins/peptides with Complete Freund’s adjuvant.

UTRECHT NANOBODY FACILITY
NanobodiesAnimals of the camelidae family have next to their repertoire of conventional antibodies also smaller antibodies,which are composed of heavy chain only and are devoid of light chains. These unusual small sized antibodies can be further reduced in size in absence of the Fc region. This structure is called single domain antibody (sdAb),variable domain of the heavy chain antibody (VHH) or nanobody. Nanobodies belong to the smallest functionalantigen binding fragments (around 15 kDa), which are only one tenth the size of a conventional antibody.
ServicesNanobodies that bind to any possible target can be selected using phage display technology. The facility isexperienced in further functionalization of the nanobodies using different conjugation strategies includingsortase tagging and click-chemistry. Examples exist for application of different nanobody conjugates forsuperresolution light microcopy, electron microcopy, and for development of novel diagnostic and therapeuticapproaches including targeted nanoparticles and photodynamic therapy.

ELECTRON MICROSCOPYSQUARE
ServicesWhile classically electron microscopy (EM) allows forthe analysis of structures in the nanometer tomicrometer range, very recent advances in detectortechnology have brought EM into the field ofanalyzing biomolecules and nanomaterials at theatomic level. The EM Square at Utrecht University, a joint initiative between the faculties of Science,Geosciences and the UMCU is devoted to thedevelopment and application of technologies for(cryo) specimen preparation, 3D(cryo) electronmicroscopy data collection, as well as imageanalysis for cell biology and material sciencesapplications.
EquipmentThe infrastructure includes state-of-the-artTransmission Electron Microscopes (TEMs) andScanning Electron Microscopes (SEM), with a full
range of possibilities such as STEM, EDX andenergy-filtering. In addition it houses a specializedfocused ion beam SEM. FLOW CYTOMETRY ANDCELL SORTING FACILITY
Flow cytometryFlow cytometry literally means the measurement ofcells in flow. In real life it means that any suspensionof particles within the size range of approximately 50micrometer to 200 nanometer can be measured.The measurements can be performed at high speed,on average up to 20.000 per second. In this way datacan be obtained from each individual particle andhigh numbers of particles can be measured in ashort period of time. This technique is extremelysuitable for measuring sub-populations (e.g. of cells)within a suspension. In addition, individual cells orparticles can be purified using cell sorting.
ServicesThe Flow Cytometry and Cell Sorting Facility at theFaculty of Veterinary Medicine has accumulatedspecific expertise on measurement and sorting ofcells since 1998. This expertise builds on theexperience accumulated with a wide variety of
analyses and purifications, for example with immunecells, stem cells, sperm cells, chromosomes, bacteria,hybridoma’s for monoclonal antibody production,single cell sorting for cloning and/or PCR analysis,cell selection based on fluorescent reporter geneproducts and rare event purifications. We are trainedto rethink seemingly impossible demands intosuccessful experiments. The facility is activelyinvolved in research related to improvement ofresolution and is for this purpose in close contactwith the manufacturers of the equipment.
- For analysis of suspensions we can accommodateexperiments requiring lasers with the followingwavelength: 405 nm, 488 nm and 640 nm.

Selection and purification of specific chemical orbiological materials are critical and often time-consuming aspects of fundamental and biomedicalresearch. Fluorescence-Activated Cell Sorting (FACS)provides a method for sorting specific cells from aheterogeneous mixture. Many objects are too largeor too sensitive for conventional FACS-based flowcytometry. The BioSorter-Pro (Union Biometrica®)has been developed for flow cytometry of objects ina wide range of sizes (10 to 1500 μm) with highspeed and high sensitivity.

The cell is a fundamental unit in Life Sciences, buttraditionally, many molecular and genetic assaysare based on material derived from thousands tomillions of cells, resulting in “average” values.However, no such thing as an average cell exists.Hence, genetic and transcriptome analysis of singlecells offers researchers a much deeperunderstanding of cell and tissue behavior, but istechnically challenging. EquipmentThe DMPC has a state-of-the-art facility forprocessing, staining and imaging of tissues fromexperimental animals. ServicesThe Pathology Department of the UMC Utrechtperforms diagnostic testing of body samples suchas tissues (histology) and cells (cytology). Furthermore, it enables a more closecollaboration with the Department of Radiology bycombining digital pathological images with digitalradiological images. This makes it easier to establisha link between imaging and tissue diagnosis, todetermine for example whether a tissue biopsy isrepresentative. This increases the quality ofdiagnosis and treatment, and thereby improvespatient safety.

We have the facilities to grow bacteriological andviral strains and perform molecular analyses. We have specific expertise in the field ofantimicrobial resistance and perform both molecular and phenotypic assays to determineresistance to bacteria as well as viruses. We have several clinical strains of viruses andbacteria for research purposes. 3Rs-CENTRE UTRECHTLIFE SCIENCES
The Centre facilitates 3Rs-research in many ways, byproviding information and advice on 3Rs-models. Itmaintains the 3Rs-Database Program that aims tofacilitate researchers implementing the 3Rs. Thisprogram makes state-of-the-art knowledge on the3Rs available for researchers in the pharmaceuticalor chemical industry and research institutions,students and others involved in animalexperiments. Currently, the program has adoptedthe Interspecies Database and the HumaneEndpoints website, which help to respectivelyreduce and refine animal experiments.
Interspecies databaseThe Interspecies Database provides insight intophysiological, anatomical and biochemicalparameters of different animal species andhumans. By using the database, researchers cansmarter design the animal experiments in terms ofchoosing the best animal model in the translation
of the data towards another animal species ortowards humans. This could lead to a betterresearch quality and a reduction in the number ofexperimental animals. Some users have indicated areduction of their animal use by 40%, with anaverage of 20% by consulting the database. TheInterspecies Database is used by, on average, 5500unique visitors per month from all over the world.All available data originates from peer-reviewedliterature.
Human Endpoints databaseThe Humane Endpoints website gives informationon humane endpoints and teaches to recognizehumane endpoints in rodents. Interventions canthen be made at the ‘right’ moment by terminatingthe experiment. The website contributes torefinement: the prevention of unnecessarysuffering in laboratory animals. The information onthe website is offered in the form of texts,photographs, videos and training modules. Thewebsite is available in different languages and usedby, on average, 5000 unique visitors per monthfrom all over the world.

DatabaseDiagnostic algorithms and clinical research dependon proper identification and characterization ofpatient populations. The UPOD database is aunique relational database collecting andcombining clinical characteristics, medication andlaboratory measurements since 2000 andencompasses data from more than 400,000individuals and >90,000,000 laboratory results(including pediatric patients). ServicesThe Pharmacy Laboratory of the UMC Utrechtperforms analyses for clinical pharmacology andtoxicology for patient care and clinical research. There is a whole range ofroutine analyses that take place at least once a year.Most methods are based on LC-MS/MS technology.
ResearchThe central research theme of the PharmacyLaboratory is to explain, predict and improve thevariability of patient response to pharmacologicaltreatment through clinical pharmacology andclinical pharmaco-epidemiological approaches.

EquipmentThe following techniques are used for analysis ofpatient material: immunoassay, AAS, HPLC/UV-fluorescence, GC, LC-MS/MS, UV-spectrophoto-metry. For quality control of raw materials andpreparations additional techniques are usedincluding IR, titrimetry, TLC, osmolarity andchemical tests. The Pharmacy Laboratory at UMC Utrecht is CCKLaccredited.

MULTIPLEX CORE FACILITY
ServicesThe MultiPlex Core Facility of the UMC-Utrecht isstate-of-the-art and offers multiple immune-assaysbased on color-coded microspheres (xMAPtechnology), which is used for “tailor made” immunemonitoring in cohort studies. The MultiPlex CoreFacility is ISO9001:2008 accredited and measuresover 160 immune related mediators (cytokines,chemokines, adipokines, growth factors and matrixproteins) using less than 100 μl sample volume.Protocols are optimized for culture supernatant,plasma, serum, synovial fluid, liquor, ocular fluid,lysed tissue, urine, dried bloodspots and middle eareffusions. The assays are sensitive, precise,optimized and validated and are setup in a flexibleand open-architecture infrastructure which allowsrapid measurements of any customizedcombination.
EquipmentThe MultiPlex Core Facility operates two FlexMAP3Dsystems in combination with various 96 and 384well automated washers. Furthermore variousstatistical software packages are in place foranalyzing and handing ‘big data’.

UTRECHT CENTER FORDIAGNOSTIC ADVANCES IN IMMUNOLOGY RESEARCH
The Utrecht center for Diagnostic Advances inImmunology Research (U-DAIR) provides an extensiveImmune Monitoring platform to study immunologicalparameters in patients with immune-mediateddiseases and to assess the effects of immunemodulatory treatment. Harmonized and standardizedapplication of diagnostic and state-of-the-art researchmethodologies enable validated biomarker researchand comparison of biomarker profiles between patientcohorts. Upon request we develop, optimize andvalidate analytical tests to facilitate research ondisease-specific parameters.
Core Competences- High throughput quantification of (adipo) cytokines,
chemokines and growth factors in multiplex assays(160+ analytes in 250 uL biological fluid).
- Extensive phenotyping of leukocytes (e.g. T, B andNK(T) cell lymphocyte subsets) using flow cytometry.
- Functional assays on cytotoxicity and assessment ofintracellular cytokine profiles in (un)stimulatedlymphocytes.
- Assessment of allergenicity and patient allergicphenotypes by Basophil ActivationTest (BAT).
- HLA-typing, SNP analyses, TCR- and IG-sequencingusing next generation sequencing.
- Pharmacological screening for immunologicalparameters and PK-PD modeling.
- Development and validation of novel immunoassays,including immunoassays for biologicals.
- Access to generic diagnostic services forimmunological parameters in Autoimmune diseases,Allergy, Transplant Immunology, Immunedeficiencies, Oncology, Hematological malignancies,and Acute & Chronic Inflammation.
- Access to certified laboratory analysis: ISO9001, EFI,CCKL.
U-DAIR is positioned within the diagnostic departmentof the Laboratory of Translational Immunology (LTI)and aims to accelerate and improve immunologyresearch and diagnostics.

HIGH-THROUGHPUTISO-CERTIFIED ELISA FACILITY
Biomarker research requires reliable analyses oflarge sample numbers in a high-throughputfashion. The ISO-certified ELISA facility is embeddedin the Department of Clinical Chemistry andHematology of the UMC Utrecht and is operated inclose collaboration with clinical chemistry experts.This ensures high-quality measurements of pre-clinical markers that can be combined withmeasuring clinical standard laboratory parametersin the routine lab.
ServicesThe ELISA facility has been set up to enable high-throughput measurements of a variety ofparameters. Additional analytes can be measuredor set up upon request. Prices depend on analytesand batch size but usually range between 3 and 4 Euro per sample including sample handling.
InfrastructureThe ELISA facility is specifically suited to handlebatch measurements. Samples are usuallymeasured in a 384 well format. Using this format,about 2000 samples can be handled and measuredper day.

MEDICAL DEVICE PROTOTYPING FACILITY
Our facility under ISO 13485 certificated offersMedTech startups and external researchers thepossibility to translate their concepts into functionaldemonstrators and even prototypes for clinical use.
Equipment and servicesThe infrastructure includes several traditionalmilling and turning machines as well as ComputerNumerical Control (CNC) machines. Also ComputerAided Manufacturing (CAM) software to translateComputer Aided Designs (CAD) into CNC codes isavailable and offered as a service. Other servicesinclude for example laser cutting and marking ofmaterials, cutting and bending of materials. Ourfacilities and dedicated instrument makers operateunder an EN ISO 13485:2012 and are certificated toensure safe product manufacturing and assembly,suitable for clinical investigation in our hospital andpartnering institutes.
Besides our own equipment we have a strongnetwork of companies specialized in dedicatedfabrication techniques, such as for example 3Dprinting, water jet cutting.
AccessWe offer this facility service to researchers andhealthcare workers of the UMC Utrecht, and also toMedTech companies. Together with Pontes Medicalwe assist (startup) companies to help them buildtheir demonstrator for preclinical investigation orbuild their medical device prototype for clinicalevaluation in the UMC Utrecht and/or with alliedpartners.

Engineering Group Engineering develops new equipment, electronics,instruments and tools. We give advice and realisedetailed designs in cooperation with our clients.Areas of expertise: 3D printing, fine mechanics,optics, (UHV) vacuum, thermodynamics,electromechanics, micro-controller design, dataacquisition (incl. for CERN), interfaces,programmable software and hardware (VHDL).Production, Service and Maintenance Production, Service and Maintenance processesmetal and plastics and also provides a full-range ofmaintenance services to all laboratory equipment.
Our machine park is capable of CNC milling andmachining up to five axes simultaneously,conventional machining and wire- and sparkmachining.
Glass workshop The Glass Workshop makes all laboratoryglassware. We use cold and warm processing onvarious types of glass and ceramics. Cold glass ismilled or machined ultrasonically using a DMG-DM-US50. We also have the equipment needed to saw,hone, strip, glue, grind, polish (flat and round),sinter at high temperature and vacuum-solder.
