How we measure size using Dynamic Light Scattering

Theprincipleofdynamiclightscatteringisthatfineparticlesandmoleculesthatareinconstantrandomthermalmotion,calledBrownianmotion,diffuseataspeedrelatedtotheirsize,smallerparticlesdiffusingfasterthanlargerparticles.ThespeedofBrownianmotionisalsodeterminedbythetemperature,thereforeprecisiontemperaturecontrolisessentialforaccuratesizemeasurement.

Tomeasurethediffusionspeed,thespecklepatternproducedbyilluminatingtheparticleswithalaserisobserved.Thescatteringintensityataspecificanglewillfluctuatewithtime,andthisisdetectedusingasensitiveavalanchephotodiodedetector(APD).Theintensitychangesareanalysedwithadigitalautocorrelatorwhichgeneratesacorrelationfunction.Thiscurvecanbeanalysedtogivethesizeandthesizedistribution.
