1 TCOT GENETIC SAMPLING DATA SHEET Please complete this data sheet whenever taking a genetic sample. Give the sample a unique code and write the sample code in pencil on a slip of paper. Insert the slip into the vial with the sample and mark the vial with sample code. Please store this master and send a copy of this sheet with the sample. While tissue samples in TCOT vials can be stored at room temperature, please store samples in a refrigerator when possible. SAMPLING PROTOCOL - Sample only from fresh specimens when the location and approximate date of capture is known. Live turtles - the biopsy punches should only be used on live turtles. Take skin samples from the soft skin around the limbs. Cut a shallow, circular incision with the punch and slice off the circular sample of the surface of the skin using a scalpel. Immediately place the sample in a vial. Safely discard the scalpel after use. Thoroughly boil and re-use the biopsy punch. Dead turtles - take muscle or skin samples of at least 1cm 2 using a sterile scalpel. Immediately place the sample in a vial. Safely discard the scalpel after use.
