Achievement: Branched ear wheat ideotype was observed to be suitable

for dry land conditions as tillering was suppressed and main shoot was affected less under such conditions.

Biomass, grain yield and harvest index showed significant positive correlation with pre-anthesis water use.

Nitrogen application was not beneficial under sever water stress but considerably helpful under mild water stress or limited water availability in improving yields in sorghum.

Technical advice to state governments for promoting plasticulture applications. To organize field demonstrations and training programmes. Survey of drip irrigation, installations in Delhi, Haryana and Punjab. Drip irrigation was found suitable for maximization (modification of leaf area) of

leaf area to maximize crop yield in potato and increased water use efficiency.Project 4. Physiological and molecular approaches for crop improvement (1999-2003)Objective: Role of Nitrogen and growth regulators in crop improvementAchievements: Benzyl- adenine maintained the chloroplast envelops which protect the loss of endogenous factors from thylakoid membrane to maintain the photosynthetic activity Sunflower and Bajra). In all the genotypes, higher doses of nitrogen resulted in the development of bigger chloroplasts with increased number of thylakoid membrane in stroma and starch granules, and early senescence. The higher activity of proteolytic enzyme due to nitrogen treatment may be the reason for early senescence. Treatment with BA delayed the appearance of symptoms of leaf senescence.Project 5. Improvement in abiotic stress tolerance in crop plants: Physiological approachesSub-project. Role of growth regulators in stress amelioration (2003 onward)Objective- Biochemical and Physiological basis of drought tolerance in wheat

Achievements: Water deficit led to a rise in abscisic acid levels and then to the induction of lignin synthesis as a survival mechanism in three different wheat cultivars grown under unfavorable environmental conditions. Photosynthets liberated through reduction of internode length were used in production of multiple layers of mesophyll tissue (thick leaf) and cell wall thickening. Plant growth reduction under drought was caused primarily by reduction in leaf area ratio. Cell sizes were reduced under higher temperature and late sown conditions due to the increased rate of cell division of the cells with different cell wall composition. Drought tolerant cultivar synthesized lignin during rehydration around vascular bundle in order to avoid cavitations. ABA enhanced programmed cell death under unfavorable growth conditions.

Project: 6. Physiological and Molecular Basis of Abiotic Stress Tolerance in Wheat, mustard and Chickpea (2009-2014)Sub- project: Plant growth regulators in amelioration of droughtObjective: Molecular, biochemical and physiological basis of drought (water stress) tolerance in wheat

Significant Achievements: From Final report of the project (5yrs)

The accumulation of the xanthophyll cycle carotenoids and zeaxanthin particularly indicated pH dependent energy dissipation (qE) a major component of NPQ. Neoxanthin and zeaxanthin served important functions as an antioxidant in the lipid phase of the membrane and are likely to act as key components in the memory of the chloroplast with respect to preceding photo-oxidative stress in next generation even under low light and normal temperature conditions. ABA accumulation was required for anthocyanin synthesis under November sown conditions (normal growth condition).

RUBP substrate remain available in C306 indicating reduced photorespiration under drought in tolerant cultivar compared to sensitive cultivar i.e. HD 2428.

A significant, positive, linear correlation was found between the expression of wheat 45–46 kDa RCA and plant productivity under heat-stress conditions. Results support the hypothesis that endogenous levels of RCA could play an important role in plant productivity under supraoptimal temperature.

Root length, root number, root mass, root tip thickness, resistance to water flow within the root, root/shoot completely modifies the plant architecture. ABA inhibited lateral root growth via suberization of cell wall. Wheat root growth is inhibited in horizontal direction due to endogenous ABA accumulation under normal as well as late sown conditions (high temperature).

ABA reduces the size of smaller vessels; reducing the efficiency of water flow in wheat plant at low temperature as well as high temperatures (inhibited cell expansion consequently reduced plant size and yield).

In the growing grass leaf, with a much looser mechanical association between mesophyll and epidermis, mechanical limitation of extension could be due particularly to stretching of protoxylem elements, while suberization of the bundle sheath or layers of small cells with low diffusivity for water may limit growth hydraulically. The xylem structure also affects the susceptibility of the root segment to cavitation, which generates transient drops of axial conductivity. These effects essentially affect the long-distance propagation of the transpiration-driven negative xylem tension.

Xylem development in flag leaf of wheat cultivars is co-limited by carbohydrate supply due to changes in PS II.

Plasmodesmata serve as directors of plant growth and development and may help to determine a program of cell differentiation, such as sealing off root and stem epidermal cells from the rest of the plant. We had recorded ectopic expression of lignin in wheat under the influence of ABA under favorable conditions for growth and development.

Membrane intrinsic protein TaPIP2.2 and AQP7 contributed in drought tolerance in wheat.

The increased expression in Ta PIP2.2 and AQP7 in leaves of C306 wheat that assisted leaf growth under high temperature and radiation stress occur primarily through post translational modifications, such as phosphorylation. We have identified conserved residues (indicated in bold type below) in a loop following the first NPA motif ARKVSLIR. A site with the sequence R-K-X-S-X-X-R is present at the internal loop in most PIPs. Sequences ELLSS at C- terminal indicate involvement of COP 1 protein in protein targeting to plasma membrane. Sequence DIE at N-terminal indicates that the protein was modified post translational after export from endoplasmic reticulum to golgibodies by adding amino sugar at sequence asparagine-X-serine i.e. NXS/T.

Amino sugar profile was changed in wheats under drought condition The higher levels of TCA intermediate suggests greater mitochondrial activity in

drought tolerant plants, which is directed towards generating more reductant and

ATP, but possibly also into mitochondrial fatty acids and for the provision of carbon skeletons for amino acid biosynthesis. Isileucine, leucine, valine, proline and arginine were detected in drought sensitive (HD 2428) and tolerant wheat cultivar (C 306). Arginine was detected in drought sensitive cultivar of wheat. Amino acid composition was dependent on development of flag leaf in wheat.
