with the current literature. Indeed, one of the great difficultiesin editing a book of this kind is the inevitable lag betweenwriting and publication. Most references are from the mid1990s, and few are as late as 2000. In the meantime, there hasbeen very substantial progress in the purification and crystal-lization of the most recalcitrant membrane proteins, thosewith multiple transmembrane domains such as pumps andchannels.
Spectacular success has recently come out of MacKinnon’slaboratory at the Rockefeller University, where novel strate-gies, including judicious choice of organism, have allowedthe structure of a voltage-dependent potassium channel to bedetermined at high resolution. At last, we can see a plausiblemechanism of how it works. Unfortunately, this advancecame too late for inclusion in this book. Such is the march ofscience.
If there is to be a second edition of this book, comparisonwith the current one would reveal that the pace is quickeningfor this class of difficult but fascinating and important pro-teins. We might expect a new edition to include greatlyincreased coverage of successful determination of the struc-ture of pumps and channels. Methods for approaching theimmense technical difficulties are becoming more reliable andfeasible. At long last, we are beginning to see how membraneproteins work at the atomic level. There is a growing sense ofsatisfaction from the improved understanding of normalphysiology. In the longer term, this should lead to moreeffective treatment of disease.

This is the fourth edition of Practical Immunology. The firstedition from 1976 graces my laboratory shelf and has beenused over the years by students and research assistants. Thecurrent edition has the same audience in mind. So often thebasic principles behind techniques are assumed, but here is atext that provides this information. I have no doubt that thisbook will still be useful in the laboratory despite the trend tosearch for everything on the Web. The authors have set up awebsite to maintain updates to methods and to be a forum forcomments from readers, although at the time of writing thisreview this site was not accessible.
This book contains 11 chapters covering a comprehensiverange of immunological techniques. The first five chapters aredevoted to antibodies, their isolation and characterization,monoclonal antibodies, antigen–antibody interactions, the useof antibodies as probes, and immunoassays. The next fivechapters deal with cells, their isolation, culture techniques,
measurement of effector functions and bioassays for cytokines.The last chapter deals with immunological studies
in vivo
.There are also three appendices: ‘Buffers and media’, ‘Basictechniques and useful data’, and ‘Equipment and manufacturersindex’.
The format of the book makes it easy to read. Each sectionbegins with a short paragraph introducing the method. This isfollowed by a list of the ‘Materials and equipment’ required,and then the ‘Method’ which is highlighted in a box, and thisis followed by a section on ‘Technical notes’. Each chapterconcludes with a list of articles entitled ‘Further reading’.
I think overall the book meets its objective. There areinstances where I thought a procedure was rather dated andunlikely to be used, such as the use of defibrinated blood forlymphocyte isolation. There are other instances where morerecent and commonly used procedures were not included,such as the use of cell surface biotinylation rather thanradioiodination for immunoprecipitation analysis. There havealso been tremendous advances in the use of fluorescentprobes for labelling lymphocytes for tracking cells
in vivo
andfor analysing cell division and these methods were notmentioned in the text. There was no mention of MHC tetramersto determine the frequency of antigen responsive cells
in vivo
,or of procedures for intracellular cytokine staining. Onefrustrating aspect was the inadequate cross-referencing toother sections of the book, so looking for a supplementaryprocedure was often not easy. In some cases, the arrangementseemed strange. Cell counting using a haemacytometer wouldseem more appropriate in the appendix on ‘Basic techniquesand useful data’, than in chapter 11. The ‘Further reading’sections at the end of each chapter would be more useful if thearticles listed were referred to in the body of the chapterwhich, in most cases, they were not. Perhaps there could havebeen concluding sections of the chapters that would lead thereader to want to look through this reference list to find otherprocedures that were not discussed. An important exclusionfrom the text was references to information on chemicalsafety. Websites for radioisotope health and safety were listedin several places in the text, but websites to alert the readerabout chemical safety for toxic reagents were not included.Such information could have been included with the list ofuseful websites at the end of Appendix B.
In conclusion, despite the trend to search for informationon the web, I think this book has a place in the laboratory.This book provides important basic information on immuno-logical techniques which is a must for students and researchassistants. Even for the more experienced, this book is an easyreference for a buffer or media formulation, or a reminder ofa technique that has not been used for a while. The book,however, would certainly have extra value if it was accompa-nied by a CD.
