Operating Instructions: Perkin Elmer UV-Vis Lambda 35

Turn on green instrument switch. Professor will login to PC and start UV WinLab
Explorer. Several Methods will be available. Choose one titled “Inorganic Lab”.
Scan Lambda35. Adjust data collection range as needed. Default is to collect data from
300-700 nm.
Fill two plastic cuvettes about ¾ full with DI H2O. Note front on cuvette facing beam.
Place in instrument. Perform Autozero with the two cuvettes containing only DI H2O.
Collect data in Range 300-700 nm or adjust as needed.
Set samples to zero, then 1 to start new run. Type in Sample ID and description. Include
chemical formula, your name, concentration, and other pertinent information.
Perform Autozero with only DI H2O in the cuvettes.
Empty the front test cuvette and fill with prepared solution. Click start.
Insert cuvette, containing sample of interest, into front cuvette holder in the same
orientation as before, while leaving DI H2O in rear cuvette.
Add peak labels via button, may need to shift threshold if too few or many peaks are
found with the search routine.
Right click on spectrum and print to lab printer.
The last person to use the UV-Vis spectrophotometer should logoff and turn off the
instrument to prolong the two bulb lives.

Additional information:
Do not use the plastic cuvettes with acetone, ethanol, or any organic solvent. Collection
in non-aqueous solutions requires glass or quartz cuvettes.
