Food Additives Sectional Committee, FAD 8

FOREWORD

This Indian Standard ( Second Revision ) was adopted by the Bureau of Indian Standards, after the draft finalized by the Food Additives Sectional Committee had been approved by the Food and Agriculture Division Council.

This standard is one of the series of Indian Standards for edible synthetic colours permitted under the plevention of Food Adulrerution Rules, 1955, Ministry of Health, Government of India. These rules, inter-alia prescribe:

‘All food colours including natural colouring matter and permitted synthetic food colours and their preparations or mixtures excluding saffron and curcumin shall be sold only under the BIS Certification Mark.’

The Indian Standard specification’for erythrosint was first issued in 1960. It was revised in 1974 to bring it in line with the FAO/WHO specifications and also taking into account the indigenous data generated. This standard is being revised again taking into consideration the latest publication for the food colour issued byFA0 ] see Food and Nutrition Paper 31/l ( 1984 ) ‘Specification of identity and purity of certain food additives 1, the latest specifications laid down under Food Chemical Codex, EEC Directives, Canadian Food Laws and Food and Drugs Act of USA In addition, due consideration has been given to indigenous data.

In this revision the minimum requirement for total dye content has been raised and the maximum requirement of loss on drying at 135’C and chloride and sulphate expressed as sodium salt has been decreased. The limits for subsidiary dyes, fluorescein, otganic compound other than colouring matter, zinc and heavy metals have been included in line with the International requirements.

Requirements for erythrosine, food grade have been prescribed under the PFA, Rules. As food colours are under compuIsory BIS Certification, the modified values given in Table 1 have been referred to CCFS for adoption under the PFA Rules. While formulating this standard, necessary consideration has been given to the Standards of Weights ond Mensures ( Pnckqed Commodities ) Rules, 1977. The standard is however subject to restrictions imposed under these Rules, wherever applicable.

Chemical Name - Disodium or dipotassium salt of 2’, 4’, 5’, 7’. tetra-iodofluorescein.

Molecular Weight - 879.87 ( d&odium salt ).

For the purpose of deciding whether a particular requirement of this standard is complied with, the final value,obsetved or calculated, expressing the result of a test or analysis, shall be rounded off in accordance with IS 2 : 1960 ‘Rules for rounding off numerical values ( revised )‘. The number of significant places retained in the rounded off value should be the same as that of the specified value in this standard.

3.2 Freedom from Contaminants

Precautions shall be taken to ensure that the material is free from mercury, copper and chromium in any form; aromatic dmines, aromatic nitro compounds, aromatic hydrocarbons, and cyanides. 3.3 The product shall be processed, packed, stored and distributed under hygienic conditions in licenced premises (see IS 2491 : 1972).

4 PACKING AND MARKING

4.1 Packing

The material shall be packed in glass containers, metal containers, polyethylene containers, or cardboard containers suitably lined with polye- thylene. Any other suitable container may also be used subject to agreement between the purchaser and the vendor.

4.2 Marking

4.2.1 Each container shall be legibly and indelibly marked with the following information:

( Packaged Commodities ) Rules, 19771 Prevention of Food Adulteration Rules, 1955.

4.2.2 BIS Certification Marking

The product may also be marked with the Standard Mark.

4.2.2.1 The use of the Standard Mark is governed by the Provisions of the Bureau of Indian Standards Act, 1986 and the Rules and Regulations made thereunder. The details of conditions under which the licence for the use of Standard Mark may be granted to manufacturers or producers may be obtained from the Bureau of Indian Standards.

5 SAMPLING

Representative samples of the material for tests shall be drawn and criteria for ascertaining con- formity to the requirements of this specification shall be determined according to the method prescribed in 4 of IS 1699 : 1994.

6 TESTS

6.1 Tests shall be carried out as prescribed in co1 4 and 5 of Table 1.

6.2 Quality of Reagents

Unless specified otherwise, pure chemicals and dis- tilled water (see IS 1070 : 1992) shall be employed in tests.

NOTE --‘Pure chemicals’ shall mean chemicals that do not contain impurititi which affect the results of analysis.

DETERMINATION OF TOTAL DYE CONTENT

A-O GENERAL

“Iwo methods, spectrophotometric and nitric acid method, are specified. In case of dispute, spectro- photometric method shall be regarded as the ref- erence method.

A-l SPECTROPIIOTOMETRIC METIIOD

A-l.1 Apparatus

Suitable spectrophotometer with properly calib- rated scales for both wavelength and optical den- sity. However, suitable spectrocolorimeter may also be used after proper calibration against a spectrophotometer.

A-l.2 Procedure

Weigh accurately about 125 mg of the dye sample and dissolve with 0.1 N sodium hydroxide in a 250- ml volumetric flask; dilute this with the same solvent to make a final concentration of 0.5 mg per 100 ml ( approximately ). Find out the optical density of this dilutesolutionagainstO.lNsodiumhydroxideasblank and 527 nm in a glass cell with 10.0 mm light path.

A-1.2.1 Simultaneously weigh accurately about 2 g of dye-sample and dry this in an air-oven at 105 + l°C for 2 hours. Calculate the loss of mass on drying; from this data, calculate the dry mass of the sample ( M ) in the final solution taken for measurement of optical density.

A-2 NITRIC ACID METIIOD

A-2.1 Reagents

A-2.1.1 Nitric Acid - 1.5 N and 0.5 percent.

A-2.1.2 Silver Nitrate - 4.25 percent ( nzlv ) solu- tion of silver nitrate ( AgNO3) in water ( 0.25 N ).

Dissolve 1.000 g of erythrosine in.250 ml of water, transfer to a clean 500-ml beaker, add 8.0 ml of 1.5 N nitric acid and stir well. Filter through a sintered glass crucible ( porosity 3, diameter 5 cm ) which has been weighed containing a small glass stirring rod. Wash thoroughly with 0.5 percent nitric acid until the filtrate gives no tuibiditywith silver nitrate and then wash with 30 ml water. Dry to constant weight at 135 + 5°C. carefully breaking up the precipitate by means of the glass rod. Cool in a desiccator and weigh.

ANNEX D

[ Table 1, Item (vii) ] DEI’ERMINATION OF FLUORESCEIN

D-l PRINCIPLE

The fluorescein is separated from the erythrosine by TLC and compared with a standard chromatogram prepared from fluorescein at the concentration corresponding to the limit figure.

D-l.1 Solvent

Methanol 500 ml Water 400 ml Ammonia (sp gr 0.890) 100 ml

D-l.2 Sample

Weigh 1 g sample, dissolve in about 50 ml solvent and dilute to 100 ml in a volumetric flask.

D-l.3 Standard

Weigh an amount of the fluorescein standard corresponding to 1 g at the colouring matter con- tent of the erythrosine sample. Dissolve in water (or in water with 10 ml ammonia sp gr 0.890 if fluorescein free acid is being used) and dilute to

Spot 25 ,ul of the sample and standard solutions side by side on a cellulose plate. Develop for 16 hours in the chromatography solvent. Allow the plate to dry. View under a UV light source and compare the fluorescence of the standard with the fluorescence oft he corresponding area on the chromatogram of the sample. The intensity of the latter shall not be greater than that of the former.

NOTE -Take special care not to allow the chromatogram to be exposed to direct sunlight.
