Yeast Lab Background Information

Yeast are Fungi.Yeast are single celled and eukaryotic.Yeast have mating types instead of sexes.Mating types are “a” and “alpha”.A deliberate mating is called a CROSS.There are many strains of yeast.

Strain 2 = pinkish red colored (both the a2 and alpha2)Strain 3 = cream colored (both a3 and alpha3

Materials

• Sharpie• Petri dish• Waste cup• Toothpicks• Class yeast- 4 plates (alpha 2, a2, alpha3, a3)

Yeast Lab Procedure

1. Neatly label petri dish along bottom edge with lab group names and date.

2. Neatly draw and label a mating grid on bottom of petri dish (see your data section!).

3. Wash hands.4. Streak parents by taking a TINY bit of each and

putting it on the plate along the top or side….PAY ATTENTION TO YOUR LABELING!!! (use a different toothpick for each mating type and strain..…a total of 4 toothpicks; be careful not to touch toothpick end or damage the agar).

5. Throw away these toothpicks.

Yeast Lab Procedure

6. Carry out the crosses (mate the yeast!!!) (use a new toothpick for each mating type and strain…a total of 8 toothpicks; be careful not to touch toothpick end or damage the agar)7. Throw away all toothpicks; wash your hands.8.Incubate at 30°C until next class… upside-down!!
