Do not waste your time leaving a message for the front desk, administrative assistant or human resource department. It is their job to screen the applicants.

You want to speak directly to the individual hiring for the job. They are the one feeling the stress of not having that position filled yet and they are the individual with the most detail about the opening.

Ideally, you would go to the company, wait in the reception area until the hiring manager is available and then approach him/her requesting an interview.
