SummaryA unique lipoteichoic acid (LTA) was isolated from a new Gram-positivebacterium, Streptococcus sp. PT DSM 8747. This organism was discovered in a
cancer patient suffering from recurrent erysipelas after surgery for Carcinoma of
the breast. The structure of this LTA is exceptional with respectto the glycolipidanchor and the very short hydrophilic chain length. It consists of a linear 1,3-linked poly(glycerophosphate) chain, attached to position 0-6 of a galacto-furanosyldiacylglycerol.The hydrophilicchain is substituted only with D-alanine.
This LTA was identified as a promising candidate for cancer therapy and is
currently being investigated due to its novel chemical structure and biologicalactivity. The concentrationof this LTA has been estimated to be around 0.1% of
cell dry weight. The Standard downstream process employing phenol was not
feasible for scale up.This thesis describes a new scalable downstream process establishedwith a
quantitative analytical system based on enzyme immunoassays(EIAs). A non-
ionic detergent, Triton X-114 (TX114) showed an interesting ability to selectivelyextract LTA at high yield and could replace phenol. For process development,in-house EIAs specific for LTA were designed and evaluated for their analyticalPerformance with samples from the lab scale process. A two-site assay with
an inhibition step and fluorescent detection showed best resultswith regard to
accuracy (96-105%), detection limit (2-10"11 M), precision and reproducibility(<15% CV). Cellular LTA was released with mechanical, enzymatic(mutanolysin M-1) and chemical (SDS) treatments as a basis for process yieldcalculations. The CMC of this LTA was the highest ever reported (3-10~5 M),suggesting a broad ränge of biological activity.The strain was cultivated with complex medium in up to 30 m3 bioreactor.The
cultivation yield of LTA could be increased at least two-fold with sucrose as a C-
source. The downstreamprocess comprised bead mill cell disruption, TX114extraction and liquid-liquid Separation with the detergent-based aqueous two-
phase system. LTA partitioned aimost entirely in the detergent rieh phase. This
phase was purified with two Chromatographiesteps. Scale up necessitated
cycling processes for chromatography with at least four runs. TX114 was

separated from LTA with anion-exchangechromatography. After salt addition to
remove excess 1-propanol, LTA was purified with hydrophobic interaction
chromatography. Residual lipid impurities were removed with a 1-butanol
extraction. Pure LTA without any detectable amount of proteins, lipids, nucleic
acids and carbohydrates could be produced in gram-scale for clinical studies.
Although the overall downstream process yield was 19% for the pilot scale (labscale: 43%), the feasibility of this process was proven to be successful in
industrial scale.

ZusammenfassungEine bisher nicht beschriebene Lipoteichonsäure(lipoteichoic acid; LTA) wurdeaus einem neuen Gram-positiven Bakterium, Streptococcus sp. PT DSM 8747
isoliert. Dieser Stamm wurde in einer Krebspatientin mit einem
Mammakarzinom entdeckt, die postoperativ an einem rezidivierenden Erysipel(Hautinfektion) litt. Die Struktur dieser LTA ist aussergewöhnlich wegen ihrem
Lipidanker und ihrer sehr kurzen hydrophilen Kettenlänge.Diese LTA besteht
aus einer linearen 1,3-verknüpften poly(glycerinphosphat)-Kettedie an die
Stellung 0-6 eines Galactofuranosyl-diacylglycerin gebunden ist. Die
hydrophile Kette ist nur mit D-Alanine substitutiert.
Diese LTA wurde als potentiellerKandidat für Krebstherapie identifiziert undwird jetzt wegen ihrer Struktur und biologischen Aktivität untersucht. Ihr Anteil
am Trockengewichtder Zelle erreicht ungefähr 0.1%. Der früher entwickelte
Standard-Reinigungsprozessmit Phenol war für die Massstabsvergrösserungnicht geeignet.Die vorliegende Dissertation beschreibt einen neuen skalierbaren Isolierungs¬und Aufreinigungsprozess entwickelt mit einem neuen quantitativenanalytischenSystem gestützt auf Enzym Immunassays (EIAs). Mit einem nicht¬
ionischen Detergens, Triton X-114 (TX114), als Ersatz für Phenol konnte die
LTA selektiv und mit hoher Ausbeute extrahiert werden. Selbst entwickelte
EIAs, spezifisch für LTA, wurden bezüglich ihrer analytischen Leistung mit
Proben aus dem Labormassstabsprozessfür die Prozessentwicklung überprüftund weiterentwickelt. Ein zwei-seitiger (Sandwich) Assay mit einem
Inhibitionsschritt und fluorimetrischer Detektion wies beste Ergebnissebezüglich Richtigkeit (96-105% accuracy), Nachweisgrenze (2-10~11 M),Präzision und Reproduzierbarkeit (<15% relative Standardabweichung) auf.
Zelluläre LTA wurde mit mechanischen,enzymatischen (Mutanolysin M-1) und
chemischen (SDS) Behandlungen als Basis für die Berechnungen der
Prozessausbeute in Lösung gebracht. Die kritische mizellare Konzentration
dieser LTA ist höher (3-10"5 M) als bei anderen LTAs. Es lässt sich einen
breiteren Bereich biologischer Aktivität vermuten.

Der Stamm wurde mit Komplexmediumbis zum 30 m3-Massstab gezüchtet. Die
Ausbeute von LTA konnte mit Saccharose als C-Quelle mindestenszweifach
gesteigert werden. Der Aufarbeitungsprozess bestand aus Zellaufschlussmit
einer Rührwerkskugelmühle,TX114 Extraktion und flüssig-flüssig Trennung mit
dem Detergens induzierten zwei-phasigen System. LTA verteilt sich in der
detergensreichen Phase, welche in zwei Chromatographiestufenaufgereinigtwurde. Die Massstabsvergrösserungerforderte einen zyklischen Prozess fürdie Chromatographie mit mindestens vier Chargen. TX114 wurde mit
Anionenaustauschchromatographievon LTA abgetrennt. Nach einer
Salzzugabe für die Entfernung des überflüssigen 1-Propanol wurde LTA mit
hydrophober Interaktions-Chromatographie aufgereinigt. Restliche Lipid-verunreinigungen wurden mit einer 1-Butanol Extraktion entfernt. Reine LTA
konnte in Gram-Mengen für klinischeStudien ohne nachweisbareMengen von
Lipide, Proteine, Nukleinsäure und Kohlenhydrate produziert werden.Obwohl die Ausbeute des Downstream-Prozessesim Pilotmasstab einen Wertvon 19% (Vergleich Labormasstab:43%) erreichte, konnte die technische
Durchführbarkeitdieses Prozesses erfolgreichim industriellen Masstab gezeigtwerden.

Un unique acide lipoteicholque (lipoteichoic acid: LTA) a ete isole ä partir d'unenouvelle bacterie, Streptococcus sp. PT DSM 8747. Cet organisme fut
decouvert chez une patiente souffrante d'erysipeles recidivants apres ablation
chirurgicale d'une tumeur au sein. La structure de cet LTA est exceptionnelle en
considerant l'ancre glycolipidique et la tres courte chaTne hydrophilique le
composant. Elle consiste en une chaTne lineaire de polyphosphates de
glycerine ä liaison 1-3, attache ä la position 0-6 d'un
galactofuranosyldiacylglycerol. La chaTne hydrophilique est substituee
uniquement avec la D-alanine.
Cet LTA a ete identifie comme un candidat prometteur pour la therapie des
Cancers et est maintenant etudie ä cause de sa nouvelle structure et son
activite biologique. La concentration de cet LTA a ete estimee
approximativement ä 0.1% de la matiere cellulaire seche. Le proceded'extraction et de purification Standard utilisant le phenol n'est pas adapte pourl'echelle industrielle.
Cette these de doctorat decrit un nouveau procede d'isolation-purificationcapable d'etre transfere ä l'echelle de production industrielle dont les
Performances ont ete estimees avec l'aide d'un Systeme analytique base sur
les dosages immunoenzymatiques (enzymes immunoassays: EIAs). Un
detergent non-ionise, TritonX-114 (TX114) a la capacite interessante d'extraire
selectivement le LTA avec un rendement eleve et pourrait remplacer le phenol.Les EIAs speeifiques pour le LTA ont ete mis au point pour le developpementdu procede et evalues pour leur performance analytique avec des echantillons
provenant du processus ä l'echelle du laboratoire. Un dosage sandwich avecinhibition et detection fluorescente a presente les meilleurs resultatsconcernant
l'exactitude de quantification (96-105%), la limite de detection (2-10"11 M), la
precision et la repetabilite (<15% CV). Le LTA ancre dans la membrane
cytoplasmique est libere avec l'aide de traitements mechaniques,enzymatiques(mutanolysin M-1) et chimiques (SDS) comme base pour les calculs de
rendement des Operations unitaire du procede. La concentration critique

micellaire de cet LTA est la plus elevee jamais reportee (3-10~5 M), suggerantun large domaine d'activite biologique.La souche de bacteries a ete cultivee en milieu complexe dans des
bioreacteursau maximum de 30 m 3. Le rendement de la productiondu LTA a
pu etre ameliore au moins deux fois avec le Saccharose comme source de
carbone. Le procede d'extraction-purificationa consiste en l'ouverture des
cellules avec un extracteur billes, une extraction au TX114 et une Separationliquide-liquide avec le Systeme aqueux ä deux phases base sur le detergent. Le
LTA s'est reparti presqueentierement dans la phase riche en detergent. Cette
phase a ete purifiee avec deux etapes de Chromatographie liquide. Le
changementd'echelle de production a necessite des procedesde purification ä
plusieurs cycles de Chromatographie avec au moins quatre charges. TX114 a
ete separe du LTA par Chromatographie echangeuse d'anions. Apres additionde sei, afin d'eliminer l'exces de 1-propanol, le LTA a ete purifie parChromatographie ä interaction hydrophobique.Les impuretes lipides ont ete separees ä l'aide d'une extraction au 1-butanol.
Le LTA pur a pu etre produit en quantite ä l'echelle du gramme pour les etudes
cliniques sans quantite detectable en proteines, lipides, aeides nucleiques ouhydrates de carbone. Bien que le rendement total du procede d'extraction-
purification a ete de 19% pour l'echelle pilote (laboratoire: 43%), la realisation
de ce procede a ete un succes ä l'echelle industrielle.
