Temperature Control (Fig.1)The key to the design of any thermalcycler is uniform and accurate temperature control. Thermo's thermalcyclers can use up to five controlmodes for monitoring temperatureduring PCR reactions: Active TubeControl, Simulated Tube Control,Simulated Plate Control, SimulatedSlide Control and Block Control.
Temperature Uniformity (Fig.2)It is paramount for reproducibleresults to attain a uniform temperatureacross the block. In Thermo thermalcyclers this is monitored across theentire block during each stage of thethermal cycling protocol. Every instrument is calibrated usingequipment traceable to the UKNational Physical Laboratory stand-ards. A dynamic uniformity test isconducted as part of Thermo's QualityAssurance procedure to ensure theymeet this specification prior todispatch.
Temperature Control Options
Temperature Gradients (Fig.3)By using our gradient blocks, it is quickand easy to optimise primer to template annealing temperatures.Multiple annealing temperatures canbe assessed in one experiment bydefining a temperature gradient of upto 15°C across the block. The optimisedtemperature can then be transferreddirectly to standard cycling due to ourinnovative software and block design.
Active Tube Control (Fig.4)Thermo's unique method of choice formonitoring and controlling any thermalcycling reaction is to measure theactual temperature of the reaction mix during PCR. When this method isemployed, the machine actively com-pensates for differences in the programmed temperature and theactual temperature achieved in thereaction mix by adjusting the blocktemperature. This method of tempera-ture control ensures that the program-med temperatures and dwell times areaccurately achieved, resulting in optimal PCR performance.
Simulated Tube/Plate and Slide Control(Fig.5)When there is a requirement to useevery position within a block, werecommend that Simulated Control beused. We have developed advancedsoftware algorithms (Simulated Tube,Plate and Slide Controls) that achievetemperature accuracy similar to ActiveTube Control for tube, 96 / 384 wellplate and slide formats. The softwareconstantly calculates the sample tem-perature, factors in the sample volumeand then adjusts the block temperatureto ensure that the sample reaches theprogrammed temperature.
Block Control (Fig.6)Our thermal cyclers can also use BlockControl, which is the sole controlmethod used on some thermal cyclers.Block Control is best used for prolon-ged static incubations. If Block Controlis used during short incubation peri-ods, such as during a PCR reaction, thereaction mix is at the set temperaturefor a considerably shorter time thanthat programmed, which could lead toa reduced yield of PCR product.
Flexible Network of Thermal CyclingBlocksThe MultiBlock System (MBS) compris-es a central computer with sophisti-cated software, which can be net-worked to up to 30 individual satelliteblocks. The choice of the number andtype of each block is completelyflexible, giving you the freedom to customise your PCR system. Satellite blocks are available in 0.2ml(gradient and standard), 0.5ml (gradi-ent and standard) and 384 well for-mats, all of which can be added singularly providing further flexibility.

Space Saving DesignMBS satellite blocks can be positionedin any arrangement: next to each otheror at opposite ends of the laboratory.This gives you the freedom to design anetwork that makes the most of yourlaboratory space. The MBS does notrequire any bulky power supply unitsto operate. Slots in the lid and blocksurround maintain the airflow in theMBS blocks. This means that theinstruments can be placed up againstone another and still be adequatelycooled. In addition, the block orienta-tion (portrait) makes the MBS anextremely narrow unit, making themost of limited bench space.
Outstanding UniformityThe MBS exhibits an excellent blockuniformity, of ±0.3°C across the blockwithin 30 seconds of reaching the tar-get temperature (measured at 72°C).This block uniformity ensures that you

96 Well FormatThe PxE thermal cycler is available in a choice of two block formats, providing great flexibility in the choiceof consumable format and through-put. The 0.2ml block has a 96 tubecapacity and accommodates tubes,plates or strips. The 0.5ml holds48 samples in 0.5ml tubes and 96 samples in 0.3ml tubes, platesor strips.
