1.0 PRODUCT INTRODUCTION AND DESCRIPTION

The BioXC™ 200GX is an affordable approach to continuous or triggered biothreat sampling. It is designed for seamless integration with the Cepheid GeneXpert® presumptive identifier. The innovative design of the BioXC 200GX eliminates the need to transfer samples from a collection vial or cartridge into the GeneXpert cartridge, significantly reducing risk of inadvertent human exposure and re-aerosolization of concentrated pathogens.

This approach helps accelerate the time from detection to confirmation testing while minimizing the risk of human contact with the sample. When a GeneXpert cartridge is placed into the BioXC 200GX dispensing chamber, sample is automatically dispensed into the cartridge and ready for subsequent analysis. A standard sample vial can be used similarly. By automatically dispensing the liquid into the cartridge, the BioXC 200GX eliminates the time-consuming and costly process of transferring sample manually with a pipette. Additional fluidics are not required.

The BioXC 200GX may also be integrated with the FLIR IBAC to create a trigger-based biothreat detection system. In that scenario, the IBAC would trigger the BioXC 200GX to take a sample only under threatening conditions, rather than continuously sampling, thus reducing the costs of consumables.

2.0 PRODUCT DESCRIPTION

The BioXC™ 200GX system is intended for use as part of a larger system. External power and computer control are required to run the system. Control and information request commands may be sent to the unit by use of HyperTerminal (available in the accessories/communications section of most computers) or other serial communication software.

The unit is equipped with a 9-pin Panel Mount D-Sub Connector (Figure 1) for interface to the computer serial port. A separate power supply and cord will connect to the circular port next to the serial port. Two Interface Cables and a Power Supply are included for this purpose.

WARNING: The BioXC may not be plugged directly into a PC serial port via a serial data cable. Use the provided custom Interface Cable only.

6.0 INITIAL SET-UP

The BioXC requires set-up before first operation. The Sample Fluid Bag is attached to the pump tubing and hooked inside the unit. The 24 VDC Power Supply with current (max/avg): 2.5/1.3 amps and custom Interface Cable are connected and the fluid lines are primed before the unit is placed in service. Perform the steps below to set-up the BioXC.

Step 1: Place BioXC on a secure, level surface.

Step 2: Press the Top Cover Release Tab down to unlock the Top Cover (Figure 6).

Step 3: Lift the cover up and away from the unit and set aside.

Step 4: Carefully lift the pump tubing out of the internal compartment to access the fitting at the end of the tubing.

Step 5: Twist and remove the cap from the fitting.

Step 6: Twist and remove the cap from the tubing on the Sample Fluid Bag.

Step 7: Attach the Sample Fluid Bag tubing to the pump tubing by twisting the fittings firmly together (Figure 7).

NOTE: It may be necessary to rotate the bag to ensure that the fittings do not untwist as the bag is placed inside the unit.

Step 8: Carefully place the bag inside the unit, fitting end first, and hang the bag from the triangle opening at its top over the Sample Fluid Bag Hook (Figure 7).

Step 9: Connect the Interface Cable to the BioXC by plugging in the 9-pin Circular Connection into the unit (Figure 1). Connect the other end to the host computer.

Step 10: Connect the Power Supply to the unit (Figure 4).

Step 11: Plug the Power Supply into a standard wall outlet to power. The Green LED on the BioXC will illuminate solid, indicating power is connected to the unit.

Step 12: Press and hold the Sample Fluid Bag Reset Button (Figure 5) for two (2) seconds to initiate the tubing priming sequence. The Green and Yellow LEDs will flash alternately. When priming is complete the Yellow LED will turn off and the Green LED will remain solid.

NOTE: Sample fluid will drain through the Sample Cartridge Interface down to the Waste Tray when the fluid lines are fully primed.

Step 13: Replace the cover by angling the front edge of the cover down under the lip on the case and then pressing down until it locks into place (Figure 7).

Step 14: Proceed to Section 7.0 LOCATION AND MOUNTING.

7.0 LOCATION AND MOUNTING

The BioXC 200GX is designed to sample an indoor space for biological aerosols. For best results, placement should be based on a threat analysis of the desired indoor space. The unit may sit on a level, secure surface or mounted to a horizontal or vertical surface. Do not locate the unit outdoors or near aerosol generating sources such as, but not limited to, indoor foliage, mail sorting machines, and unfiltered air exhausts.

CAUTION: Ensure that the unit’s Exhaust Vents are not blocked when locating the BioXC.

HORIZONTAL SURFACE MOUNTING

The BioXC 200GX may be mounted onto a horizontal surface. A Mounting Bracket and Hardware (Figure 9) has been included for this purpose.

NOTE: Ensure the BioXC 200GX is mounted on to a level surface.

Step 1: Attach the Mounting Bracket to the desired surface. The holes for mounting to a

horizontal surface are 4” apart on center. Step 2: Slide the BioXC 200GX down onto the mounting bracket (Figure 9).

WALL MOUNTING

The BioXC 200GX may also be mounted vertically to a wall using the mounting bracket.

CAUTION: Ensure the unit is mounted level.

Step 1: Secure the Mounting Bracket to the desired surface. The holes for mounting to a

horizontal surface are 3.5” apart on center. Step 2: Slide the unit on to the bracket (Figure 9).

Alternately, the BioXC 200GX may be mounted to a vertical surface without the mounting bracket using the keyholes on the back of the unit for this purpose (Figure 10). The keyholes are spaced 5 3/8” apart on center.

8.0 ELECTRICAL INTERFACE

A custom interface The single end of the custom Interface Cable plugs directly into the unit’s 9-pin Panel Connector. The other end of the cable connects to a host computer (Figure 2).

Data interface for communications is a RS-232 protocol set up with the following parameters:

• Bits per second (Baud rate): 57600

• Data bits: 8

• Parity: none

• Stop bits: 1

• Flow control: none (no handshake)

The unit can transmit data related to status of various components. All data strings transmitted from the unit are followed by ‘\r’ (0x0A) and ‘\n’ (0x0D) characters.

Table 5 lists messages from the BioXC 200GX.

9.0 SAMPLE RETRIEVAL

At the end of sample collection the Yellow LED will illuminate, indicating the sample is in the internal reservoir ready to be dispensed. The Countdown timer displays on the host computer, indicating how long the user has to retrieve the sample. The Sample Cartridge Interface has been specially designed such that when the user inserts a Cepheid GeneXpert Sample Analysis cartridge, the BioXC 200GX automatically dispenses the sample fluid for use with the GeneXpert System. Sample not dispensed within the user-specified set time will be automatically dumped to the Waste Tray. The default sample holding time is four (4) hours. Perform the steps below to retrieve a collected sample.

CAUTION: Ensure both the Green and Yellow LEDs are solid after Sample Collection before performing these steps.

Step 1: Prepare a new GeneXpert Sample Analysis Cartridge by puncturing and opening the sample, marked with an “S”, hole using standard methods. Open the “S” hole as large as possible.

Step 2: Insert the cartridge into the Sample Cartridge Interface (Figure 11). A blue LED in the Sample Cartridge Interface will illuminate the cartridge. This will command the BioXC 200GX to dispense sample fluid into the cartridge. The pump will turn on and the Green and Yellow LEDs will flash.

Step 3: Remove the cartridge after the pump has turned off and the Yellow and Green LEDs change to solid.

Step 4: If another sample is desired, repeat steps 1 through 3. Each 10 mL collection has enough fluid to dispense sample to four (4) cartridges plus archive.

Step 5: If an Archive Sample is desired, or to return the unit to stand-by mode, proceed to Section 10.0 MANUAL ARCHIVE DISPENSE/RINSE BUTTON.

10.0 MANUAL ARCHIVE DISPENSE/RINSE BUTTON

The user may also command the unit manually to dispense an Archive Sample by pressing the Manual Archive Dispense/Rinse Button. The Sample Cartridge Interface has been designed with a recess to hold the 15 ml Archive Vial included with the unit for this purpose (Figure 12).

The Manual Archive/Rinse Button is also used to initiate a manual Rinse Cycle. Perform the steps below to retrieve an Archive Sample and/or initiate a manual Rinse Cycle.

ARCHIVE SAMPLE

Step 1: Insert the Archive Vial into the recess in the Sample Cartridge Interface (Figure 12).

Step 2: Press the Manual Archive Dispense/Rinse Button for 2 seconds. The Green and Yellow LEDs will flash to indicate dispense.

Step 3: Remove vial when LEDs return to solid.

NOTE: The volume of sample dispensed to the Archive Vial will range depending on the amount of sample previously dispensed and the environmental conditions (temperature, relative humidity, etc) during sampling.

MANUAL RINSE

Step 1: Press and hold the Manual Archive Dispense/Rinse Button for 4-5 seconds to initiate a manual rinse. Once the rinse cycle is complete the unit will return to Standby and wait for next trigger.

Note: All of the sample remaining in the collection reservoir will be pumped to Waste Tray.

11.0 SAMPLE FLUID BAG REPLACEMENT

The Sample Fluid Bag holds 200 ml of fluid. Each bag is marked with an expiration date. Bags installed inside the BioXC should be changed every two months, even if it is not empty. Store unopened bags at room temperature. Perform the steps below to change the Sample Fluid Bag.

Step 1: Remove the unit from service.

Step 2: Disconnect Power Supply.

Step 3: Press the Top Cover Release Tab down to unlock the Top Cover (Figure 13).

Step 4: Lift the Top Cover up and away from the unit and set aside.

Step 5: Unhook used bag and carefully lift out of unit.

Step 6: Twist fittings to remove bag from pump tubings. Set bag aside.

Step 7: Twist and remove the cap from the fluid line fitting of the Sample Fluid Bag.

Step 8: Attach the Sample Fluid Bag tubing to the pump tubing by twisting the fittings firmly together (Figure 14).

NOTE: It may be necessary to rotate the bag to ensure that the fittings do not untwist as the bag is placed inside the unit.

Step 9: Carefully place the bag inside the unit, fitting end first, and hang the bag from the triangle opening at its top over the Sample Fluid Bag Hook (Figure 14).

CAUTION: Ensure the tubing does not catch on the unit. As much as possible avoid touching the electrical connections when placing the Sample Fluid Bag inside the unit.

Step 10: Reconnect the power supply.

Step 11: Press and hold the Sample Fluid Bag Reset Button for two (2) seconds to initiate the tubing priming sequence. The Green and Yellow LEDs will flash alternately. When priming is complete the Yellow LED will turn off and the Green LED will remain solid.

NOTE: Sample fluid will drain through the Sample Cartridge Interface down to the Waste Tray when the fluid lines are fully primed.

Step 12: Replace the Top Cover by angling the front edge of the cover down under the lip on the case and then pressing down until it locks into place (Figure 15).

12.0 WASTE SPONGE REPLACEMENT

The BioXC has been designed with a Waste Tray in the bottom of the unit for drainage of waste fluid from unused collection and rinse cycles. The tray contains a sponge to soak up the excess fluid. Since the BioXC is designed to be triggered infrequently, the fluid evaporates off over time between tests. The sponge should be replaced every six months. Perform the steps below to change the sponge.

Step 1: Remove the unit from service.

Step 2: Disconnect power.

Step 3: Place the unit on its back on a flat, secure surface.

Step 4: Remove the four Philips Head screws that are recessed in the rubber feet (Figure 16).

Step 5: Pull the bottom away from the unit (Figure 16).

Step 6: Replace the sponge.

Step 7: Replace the bottom of the unit and secure the four screws.

Step 8: Reconnect power.

Step 9: Return the unit to service.

Note: Maintenance beyond what is stated here may ONLY be performed by technicians trained and certified by FLIR Systems. Maintenance performed without training by FLIR will void the warranty.

13.0 TROUBLESHOOTING

Possible troubleshooting solutions are provided in Table 8. Contact FLIR Systems for more information.

Problem Possible Solutions

Green LED is not illuminated Check connection to 24 VDC Power Supply.

Unit does not trigger Check the cable connection to the unit. Check connection to 24 VDC Power Supply.

Sampling cycle runs but no fluid is dispensed

Ensure the Sample Collection Cartridge is installed and the blue LED in the cartridge housing is illuminated. Use the Manual Archive/Rinse Button (hold for 2 seconds) if sample does not automatically dispense.

Stopped pumping fluid during the sampling cycle

Wait a few seconds. The pump is designed to stop for a few seconds before the end of the cycle to allow most of the liquid to drain.

Check if Sample Fluid Bag is empty; replace if necessary Inspect tubing for kinks

Sample volume is less than 8 mls

The timing of the sample recovery may need to be adjusted. Contact ICx for more information.

Fluid is being dumped into the bottom of the unit

The unit is designed to dump liquid into a Waste Tray Sponge located in the bottom of the unit.

15.0 WARRANTY

Limited Warranty and Remedy. FLIR Systems warrants that, upon delivery, each Product will be free from defects in materials and workmanship and will operate in all material respects in accordance with applicable instructions and manuals provided by FLIR. Coverage under this Limited Warranty shall commence immediately upon delivery and is valid for a period of one (1) year from the date of the invoice. This warranty will not apply to any Product that (a) has not been operated and maintained in accordance with applicable instructions and manuals, (b) has been repaired or altered by unauthorized personnel, or (c) has been misused, abused, damaged or subjected to operation for which it was not intended. FLIR will use commercially reasonable efforts to correct any failure of any Product that does not substantially conform to the foregoing warranty (e.g., by modification or repair of the noncomplying Product) provided that Customer gives FLIR written notice of the noncompliance within thirty (30) days after delivery. If Customer gives FLIR notice of any such noncompliance in accordance with the foregoing and FLIR fails to correct the noncompliance within a reasonable period of time after receipt of such notice, then Customer may return the Product and elect to either (i) receive a full refund of any amount paid by Customer for the Product or (ii) require FLIR to promptly deliver a replacement Product, in either case without cost to Customer.
