Operating environment of instruments

In order to extend the machine’s service life, please pay attention to the following factors which may damage the machine:

• EnvironmentalConditions;• Effectsofenvironment,includingnaturalultravioletradiation(ie:DirectSunlight)• Erosionandabrasionoftheprotectivecoverandothersafetyparts.

• Indooruse;• Altitude:<2000m;• Ambienttemperature:+5°C~+35°C;• Relativehumidity:<80%;• Operatingvoltage:230V,50/60Hz;• Pleaseensureadequateventilationinareaofuseandappropriatesealingmeasurewhenperformingthe separationofpoisonoussubstancesandpathogenicmicro-organismcells;• Ensurethereisnovibrationand/orastrongdrafttoinfluencetheperformance.• Ensurethereisnodustorflammableandcorrosivegaspresentintheambientair.

Please read this manual carefully when using the instrument for the first time!The maintenance of the instrument should be carried out by the agents authorized by Biovac.

The following materials should not be used in conjunction with this instrument:

• Flammableandexplosivesubstances;• Strongchemicalmaterials;• Toxicorradioactivesubstances,• Pathogenicmicroorganisms

TheBiovac13RHighSpeedRefrigerationBench-topCentrifugecanbemaintainedonlybyqualifiedserviceengineersusingappropriatetools.

Shouldtherebeaproblemnotmentionedinthismanual,pleasecontactaBiovacauthorizedagentforthecorrectprocessingmethods.

OnlyaccessoriesprovidedbyVacutecshouldbeused.Ifotheraccessoriesareused,Vacutecwillnotbeheldresponsibleforanydamageorinjurytoinstrumentoroperatorasaresult.

TheBiovac13RHighSpeedRefrigerationBench-topCentrifugeshouldbeinspectedandmaintainedatstatedintervals.

1. Safety clauses

1.1 Operating requirements Biovac13RHighSpeedRefrigeratedBench-topCentrifugeisdesignedaccordingtopresenttechnical

standards.Thereforeitmustbeusedaccordingtothedesignrequirements.WhentheBiovac13RHighSpeedRefrigeratedBench-topCentrifugeisusedaccordingtothefollowingimproperorinappropriatemethods,equipmentmybedamagedortheoperatorinjured. • Notusedinaccordancewiththedesignrequirements/intendeduse. • Theoperatorandmaintainerhavenotbeentrainedinitscorrectuseandoperation. • Impropermodificationstothedesignwithoutauthorisation. • Failuretoreadtheregulationsforsafeuse.

PersonswhouseormaintaintheBiovac13RHighSpeedRefrigeratedBench-topcentrifugeshouldreadandunderstandrelatedusageandsafetyregulationsinthisOperatingManual.

Thefollowingregulationsshouldbeobeyedtoavoidaccidents: OperatingManualshouldbekeptneartheBiovac13RHighSpeedRefrigeratedBench-topCentrifugeforreferencebyoperators.

Biovac13RHighSpeedRefrigeratedBench-topCentrifugeisdesignedtoseparatebiologiccells,organismsandbacteria.Thedensityofthesampleshouldnotexceed1.2g/cm3atthemaximumspeed.

Duringoperation(whenthesampleisbeingseparatedorrotorsarerunning),operatorsanddangeroussubstancesshouldmaintainadistanceofatleast30cmfromthecentrifuge.Theventilationholeofthecentrifugeshouldnotbecloggedbysubstances.

IfthefollowingsafetyprecautionsarenotobeyedwhenusingtheBiovac13RHighSpeedRefrigeratedBench-topCentrifuge,operatorsorotherpersonsmaybehurtorthecentrifugeandsamplesmaybedamaged:

• Thecentrifugeisnotcorrosionprooforimplosionproof,thereforeitshouldbeusedinacorrosion andimplosionfreeenvironment.

• Thefollowingmaterialsareprohibited: Flammableandexplosivesubstances: Strongchemicalmaterials; • Toxicorradioactivesubstances,orpathogenicmicroorganisms. • Corrosivesubstancesmaydamagematerialsinsidethecentrifugeandtherotors,thereforethey shouldbeputinsidesealedcontainerswhileseparating.

1.2 Precautions • Beforerunning(whenseparatingsamples),appropriaterotorsmustbefixedfirmly; • Duringoperation(whentherotorisrunning)orafterthecentrifugehasbeenstopped(therotoris

stillrunning),openingthelidbyhandisstrictlyforbidden. • AccessoriesmustbeprovidedandapprovedbyVacutec.Somecommonparts,suchasglassand plasticcontainersforseparating,shouldbeVacutecapprovedandbeinaccordancewiththe requirementsofthehighestspeedandmaximumcentrifugalforceofrelatedrotors; • Donotusethecentrifugeorseparatesampleswhilethelidisopen; • Changingmechanicalpartsandelectronicsshouldbecarriedoutbyqualified Vacutecpersonnelonly; • Whenusingthecentrifuge,usetheappropriaterotorsaccordingtoload.DoNotoverload; • Maintenanceshouldbecarriedoutperiodically.See“Cleanandpurify”chapterformore information.

2.2 General Biovac13RHighSpeedRefrigeratedBench-topCentrifugewithmultifunctionprotectionintroduces

advancedtechnologysuchasspeedadjustmentviafrequencycontrol,microcomputercontrol,touchbutton,dialsetup,digitalLCD.Threerotorvariationsareavailable(fordetails,see“Table1:Typeofrotorandtechnicalparameters”).Thiscentrifugeisusedfortheseparationofbiologiccells,organismsandbacteria.

2.3 Device structure 2.3.1 The Device Lid

Onlywhenthelidlocksarelockedonboththeleftandrightofthelidmaythecentrifugerun; otherwisethewarningsystemwillsoundanalert(alightwillcomeonandabuzzerwillsound) andthedevicewon’trun.

Toopenthelidpress“”(openbutton)onthecontrolpanel.Oncethelidrisestoacertain height,thelidhingeandspringswillsupportthelid. Ifapowercutoccursortheopenbuttonisnotresponding,thesamplemustberemoved.Referto

2.3.2 The Chamber Thechambermaintainsastableworkingtemperature.

2.3.3 The Rotor Fordetailssee“Table1:Typeofrotorandtechnicalparameter”.Therotorrotatesata predeterminedspeedtocreatetherelativecentrifugalforcerequiredtoseparatevarioussamples. Thecentrifugalforceisseveralthousandtimesmorethang(accelerationofearthgravity),itis therefore important to use and maintain the rotor carefully and safely!

2.3.4 The Refrigeration System Therefrigerationsystemisusedtocreatealow-temperatureoperatingenvironmentforthe centrifugechamber.Thetemperatureisautomaticallycontrolled.Thecentrifugeshouldbe operatedinanenvironmentwithaconstantambienttemperaturebetween5ºCto35ºCandan ambienthumidity≤80%.Optimumoperatingconditionscanbeachievedwithadequate ventilationaroundthecentrifugeinaroomwithambienttemperature controlledbyair-conditioning. Aheatsinkisinstalledinthefrontofthemachinewhereairflowandcirculationisessential.Any dustshouldberemovedfromtheradiatorwithabrushorcompressedaironceeverythree monthstoensureeffectiveheatdissipation.Thedigitaltemperaturesensorinstalledatthebottom ofthechamberisusedtomonitorthetemperatureinthechambercontinuously.

Thetemperatureinthechambermayvaryfromthatofthesamplesatfirst,andtake aperiodtoequalise.

2.3.5 The Control System The control system allows the setting of the rotor speed, running time, temperature and control of displaysystemandwarningsystemoftheentiremachine.Toensureproperrunningofthe machineandthesafetyoftheoperator,DONOTopenthefrontcasingofthedevice,asthemain controlcircuitboardandotherelectronicsareinstalledontothefrontcoverofthedevice.

2.3.5 The Display System Thedisplaysystemcanshoweverysettingparametersimultaneously,andcantrackanychange inrealtime.Inaddition,itdisplaysrelativesystemerrorsandwarnings.

2.3.5 The Warning System Thewarningsystemwillalerttheuserof“overspeed”,deviceoverheating,lidwarning,etc. Analertisindicatedbyaflashing,red,warninglightonthecontrolpanelandabuzzerwillsound simultaneously.Ifrunning,thedevicewillbestoppedautomaticallyandtheuserwillbeunable tostartasequenceagainuntiltheerrorhasbeenresolved.NOTE:usermaypressthe“Esc”button onthecontrolpaneltosilencethewarningbuzzer.

2.4 Safety Protection Device

TheBiovac13RHighSpeedRefrigeratedBench-topCentrifugehasaseriesofsafetyprotectiondevices:

• Themountingrackismadeofsteelplatingandtheprotectionsteelringismadeofsteelcasting.Theinternalbladderoftheinnerchamberisstainlesssteel.ThefrontframeismadeofABS.Thelidisexplosionproof.Thereisaviewportinthecenterofthelid.

Onlywhenthecentrifugeisonandtherotorhascompletelystopped,willthe“”buttonopenthelid.

• OverspeedStop Thewarningbuzzerissoundedwhentherotorspeedexceedsthesettingspeedby200r/min.Therotorstopsautomaticallyifthespeedexceedsthemaximumratedspeedby250r/min.Waituntiltherotorstopscompletely,thenopenthelid.Restartoncetheerrorhasbeenresolved.

• Openingisstrictlyforbiddenduringoperation. If“”isaccidentlypressedduringoperation,thedevicewon’trespond,however,thewarningalarmis

indicated.

• Openinginthecaseofanemergency Iftheelectricityisofforthereisanurgentproblemwiththerotor,youmayopenthelidmanually

whenitisnotpossibletodosobypressingthe“”button(ENSURETHEROTORHASSTOPPEDCOMPLETELYFIRST,TOAVOIDINJURY).

DONOTUSETHEEMERGENCYOPENINGFACILITYUNNECESSARILY!

4.Preparationsbeforeuse4.1Transportationandinstallation Biovac13RHighSpeedRefrigeratedBench-topCentrifugeistransportedinspecial-purposecartonwith

protectionmaterialsforbufferinginside.Afteropening,theprotectionmaterialsforbufferingshouldberemoved.

• Thenetweightofthedeviceisabout47kg,thereforeitisnecessarytoliftthedevicefrombothsides(theleftandright)tokeepitsbalance.Thefrontcovershouldnotbepressedonorusedforlifting.

• Keeplevelandavoidvibrationwhentransporting• Pleaseusespecialpackingcasesduringlong-distancetransportation.Keeplevelandhandlewith

care!

4.2ProperplacementoftheCentrifuge Biovac13RHighSpeedRefrigeratedBench-topCentrifugeisforindooruse,andtheworkareashould

meetthefollowingrequirements:

• Asafetyclearanceof30cmmustbemaintainedaroundthecentrifuge.Whilerunning,thereshouldbenoindividualorhazardoussubstanceincloseproximitytothecentrifuge.

• Theadapterortabletopshouldbesolidtoavoidvibration;ifmobileracksortrolliesareused,theyshouldbeequippedwithlockingdevicestoensurethesafeoperationofthecentrifuge.

• Ifthecentrifugeisplacedbesidethewallorinacorner,thereshouldbeaspaceof10cmbetweenthebackofthecentrifugeandthewalland15cmbetweeneachsideandthewalltoallowsufficientventilationandheatemission.

• Thecentrifugeshouldbeplacedawayfromwindowstoavoidtheheatfromdirectsunlight.• Thefoursupportinglegsofthecentrifugeshouldstandevenlyona“swing-out”table-board.• Thecentrifugeshouldbeputinacleanroomwithaconstantambienttemperaturebetween5ºCand

35ºCandanambienthumidityoflessthan80%

4.3Fixedplacementofequipment Afterthecentrifugehasbeenplaced,don’tmoveitfrequently.Alwaysensurethecentrifugestandslevel

onthetable-board.Theshouldbenovibration.

Toavoiddamagetotherotoraxis,pleaseremovetherotorwhenmovingthecentrifuge.

Theeachoftheelectricplugconnectedtothecentrifugemustbeingoodcondition.Thevoltageandfrequencyofthepowersupplyshouldbeinaccordancewiththerequirementsstatedinthetechnicaldescriptionorspecificationslabelledonthenameplateofthecentrifuge.Thepowerjack/switchisattherearofthemachine,“I”foronand“O”foroff.Onlyusethepowercableincludedwiththemachine.

Neverpress“▷/☐”or“QTemp”buttonimmediatelyafterstartup;thedoormustbeopenedfirsttoremovetherotorfixingexpandedpolyethylene(EPE)androtor,andthenthechamberandrotormustbeinspectedandcleanedbeforerunning.

5.2 Start Connectthepowercablesuppliedwiththemachineintothepowersocketontherearpanelofthe

machineandconnecttheotherendintomainssupply.ThepowersupplyusedforthemachineisAC230V,50/60Hz.Oncepluggedin,turnonthepowerswitchnexttothepowersocket.Thebuzzerwillsoundandthesoftwareversionwillbedisplayed.Oncetheself-checkiscompleted,itwillenterthemaininterface(asshowninFig.7),thenextoperationcannowbeconducted.

5.3 Open Whilethepowerswitchisturnedonandtherotorhascompletelystopped,press“”button(door-

openbutton)onthecontrolpaneland“”willappearinthedisplaywindow;thelidwillautomaticallyopenviaspringaction,andthelidneedstoberaisedtherestofthewayandleanedbackwardagainsttheframe;theinnerchamberwillnowbevisibleandtheiconrepresentingdoorstatusonthemaininterfacewillbedisplayedasunlocked.

ATTENTION:whenthedoorisopened,thelidwillautomaticallybounceuptoacertainheight,keepclearofthelidduringthisoperationtoavoidinjury.Inthecaseofanerror,wherethedoorcannotbeopenedautomaticallyandthesamplesinthechamberneedtoberemoved,thedoorcanbeopenedmanuallybythemethodstatedinchapter7.1

5.4 Close Presstheliddownuntilthedoorhookontheliddrivesthelockingdeviceandyouhearthelockclick;

whentheicondisplaysas“”,itindicatesthatthedoorhasbeencorrectlyclosedandlocked. LidAdjustment:whenclosingthelid,ifyoufeelthelidistootightortooloose,unscrewthelocknutonthedoorhookwitha17mmsocketwrenchandthenadjustthedoorhookappropriately;turnclockwisetotightenandanti-clockwisetoloosen.Aftercompletingtheadjustment,tightenthelocknutagain.

Whenclosingthedoor,anappropriateforceshouldbeappliedtopresstheliddowntoavoiddamage to the door lock.

5.5 Installation/Dismounting of the rotor Ifinappropriaterotorandadapterareused,itwillcauseanunsatisfactoryseparationeffectand

could also damage the centrifuge.

TherotorusedmustbeaproductconfirmedandappointedbyVacutec,otherwiseitmaypotentiallycauseafault.ThespecificationandmodelofalltypesofrotorsconfirmedbyVacutecareprovidedinthismanual(fordetailsonspecificationandmodel,seeTable1“RotorTypeandTechnicalParameters”).

Wheninstallingrotors,usethe5mmhexagonspannersuppliedalongwiththemachine. Installation steps for rotors are as follows:

• Turnonpowerswitchandwaitforself-checkcompletion. • Press“”buttonandopenthelidofthecentrifuge;makesurethechamberisclean andimpurityfree;

• Cleanexternalsurfaceofmotorshaftandinspectthesealringonthemotorshaftensuring itisintact; • Preparetherotoryouwanttouse;holdtherotorwithbothhandsandalignthecenterholeofthe rotorwiththemotorshaft,andthenverticallyloweritontotheshaft; • Screwonthelocknutbyhandandthenusethe5mminnerhexagonspannertotighten thenutclockwise; • Rotorinstallationiscompleted.

Beforeuse,especiallybetweenprolongedperiodsoftime,theseatingoftherotorshouldbeinspectedforanychanges;whennecessary,thelocknutshouldbere-tightenedtoensuretherotorissecure.

5.6Maintenanceofmotorsupportshaft Maintenancestepsofmotorsupportshaft: • Turnonpowerswitchandwaitforself-checktocomplete • Press“”buttonandopenthelidofthecentrifuge;thendismounttherotorusing5mminner

hexagonspanner.NOTE:tightenclockwiseandloosenanti-clockwise. • Removedust,impuritiesandseparatedsampleresidueinthecentrifugechamberensuring

itisclean. • Cleantheconicalsectionandthescrewthread(showninFig.8)ontherotatingshaft,ensuringno

dirtordustremains;thenevenlyapplythelubricatinggreasesuppliedalongwiththemachine.

5.7 Calculation of rotor load Calculationofmaximumload:

Biovac13Rdeskcentrifugewillgenerateahighcentrifugalforceduringhigh-speedrunning;eachrotorisdesignedtohaveenoughmechanicalstrengthtowithstandthisforcewhenworkingattheratedmaximumspeed,buttoavoidaccidentorinjury,therotorloadmustnotexceeditsmaximumratedload.

Whenseparatingsamples,thesamplesarefirstplacedintoacontainerandthenplacedtogetherintoa rotor, the sum total of the container and sample must not exceed maximum rated load of the rotor, otherwiseyoumustreducethesampleweightorcalculatepermissiblerunningspeedofrotor(NPERM)toensuretherotorloaddoesnotexceeditsmaximumratedload.

Where: Nmax=maximumratedspeed; Maximumpermissibleload=maximumpermissibleload; Actualload=actualload

Don’tusetherotorunderoverloadedconditions,asthismayresultinseveredamageorinjurytotheequipment or operator.

5.8 Sample filing in centrifugal container Whenthecentrifugeruns,ifbalancedcorrectly,separationzoneofsampleswillnotinterferewitheach

otherduetotheabsenceofvibration,resultinginanimprovedcentrifugaleffect.Therefore,samplesshouldbeplacedintothecentrifugalcontainerassymmetricallyaspossibletoachievethebestbalanceduringthesequence.

Carefullyinspectthecentrifugalcontainer(centrifugetube,etc)beingused,ensuringitisinaccordancewiththeallowablemaximumratedacceleration(centrifugalforce);alwaysusethelowestrunning speed possible.

Pay strict adherence to the service life of the centrifugal container in use; especially when frequently runningundermaximumpermissibleload(speedandtemperature). Ifdamaged,itshouldbereplacedimmediately!

5.9 Safe use of rotor • Beforetherotorruns,sampleshouldbeloadedcorrectlyandsymmetrically.

5.13 Temperature Control

5.13.1 Pre-Refrigeration Function

Whenthelidofthecentrifugeisclosedandpriortherunningofthepresetprogramme,oraftercentrifugalseparationiscomplete,thecentrifugewillenter“pre-refrigerationmode”accordingtothesettemperature.Inthisway,itcanreachtherequiredtemperaturepriorcentrifugalseparation;andmaintaintheappropriatetemperatureafterseparationtoprotectsamples.Ifambienttemperatureorchambertemperatureishigherthanthatofthesample,usersareadvisedtopre-refrigerateandtheninsertsamplesforcentrifugalseparation,otherwisethe“TEMPOVER40ºC”warningmayappear.

Attention:Pre-Refrigerationfunctionwillstopafter1hour;Itisresetbyopeningandclosingthedooragainorduetopowerfailure.

5.13.2 Quick temperature function

Thecentrifugehasaquicktemperaturefunction.Firstsetthetargettemperatureparameterandexpectedtimedurationandmakesuretherotoriscorrectlyinstalledandlocked;closethelidandthenpressthe<Qtemp>buttontostartthequicktemperaturefunction,andtherotorwillspin.

Ifsettemperatureishigherthanchambertemperature,thecompressordoesnotstartandheatisgeneratedbyfrictionbetweenthehigh-speedrunningrotorandair,toquicklyreachthetargettemperature.

Ifsettemperatureislowerthanthechambertemperature,therotorrunsatthespecifiedspeed;thecompressorwillstartandtherefrigerationsystemstartstoquicklyreducethechambertemperaturetoreachthetargettemperature.

Afterthetemperaturereachesthesetvalue,therotorcontinuesrunning,butmainparameter<Qtemp>ondisplayscreenwillflickerandsimultaneouslybuzzersoundsintermittentlytoremindtheuser

6. Maintenance

6.1 Cleaning and sterilization

Usershavetheresponsibilitytocleanandpurifythecentrifugewhendangeroussubstancesspilloverintothemachine.Usersshouldcleanandpurifyitaccordingtothemethodrecommededbythemanufacturerstoensurethatthemachineisnotdamaged.

Inappropriate detergents and incorrects methods of cleaning may do harm to the centrifuge or internal parts. Cleaning and sterilization should be carried out according to operating procedures described in the operating and technical instructions.

6.1.1 Implementation of cleaning and sterilization

Turnoffthepowerandremovethepowercablebeforecleaning!

Thecentrifugeshell,rotorchamber,rotorandcentrifugalcontainersusedandsoonshouldbecleanedandmaintainedregularly;thuscontaminationcanbeclearedavoidingdamagetothepartsusedandapollutedenvironment.Ifyouarenotclearonwhichcleaningagentsshouldbeusedforcleaningthecentrifuge,pleasecontactVacutec.

• Organicsolventsareforbidden,astheycandegreasethebearingsofthemotor; • Liquid,especiallyorganicsolventsshouldnotcomeincontactwiththesupportingshaftandball

bearingswhenbeingcleaned; • Ifthereisfrostorathinicelayer,theyshouldbewipedawayoncetheyreverttowater.

6.1.2 Implementation of high-pressure sterilization

• Rotorsusedbythiscentrifugecanendurehigh-pressuresterilization.Temperature:121ºCwith apressureof250kPa,maintainedforatleast10minutes.Pleaserefertotheinstructionsofferedby manufacturersvariousseparatingcontainers.

• Alwaysconfirmwhetheritispermissibletosterilizerotorsandcontainerswithhigh-pressure sterilizationmethod.Thetemperatureanddurationofhigh-pressuresterilizationshouldbestrictly controlledaccordingtothedifferentrotorsandcontainersusedforseparating;

• Dontusearotororcontainerforseparatingiftheyarecorrodedordamaged.

6.1.3 Maintenance

6.1.3.1 Donotbringtherotorintocontactwithsharpobjects.Whenmovingordisassemblingthe centrifuge,youshouldtakecarenottodamagetherotor,asscratchesandindentationsmaycause cracksduringoperation.

6.1.3.2 Regularlycheckforspots,groovesorminutecracksintherotor(especiallyatthebottomofthetest tube).Ifanyarefound,stopusingthemimmediatelyandcontactVacutec.
