InSite Salmonella is a quick and easy test for the detection of Salmonella species in
food processing environments. The test contains a liquid medium formulated with
growth enhancers and chromogenic compounds selective for Salmonella species. A
color change from purple to bright yellow in 24 – 48 hours is presumptive positive for
Salmonella species. InSite Salmonella is a self-contained screening test which is easy
to perform while reducing wait time, labor costs, and materials.

There is no simple single testprocedure that can give a definitiveresult for the detection of pathogens
such as listeria. All methods involve samplecollection devices, enrichment in culturemedia, process treatments and entailseveral handling or transfer steps.

These procedures generate a presumptivepositive result which only becomes a truepositive result when the identity of theisolate is confirmed. A presumptive positiveresult that subsequently does not confirmthe identity, is a negative result and not afalse positive. More specific and definitivedetection methods are more expensivethan simple screening tests.The benefit of screening tests is that
negative samples can be eliminated quicklyto verify process control procedures andreduce the need for expensive confirmatorytests. This enables testing to be more cost
effective by directly reducing costs or usingthe same budget to increasing the numberof samples tested, thus minimising the riskexposure and improving quality assurance.
3-in-1 cleaning tool
A new listeria screening test provides asimple convenient cost effective solution.InSite L. mono Glo is a single device
intended for monitoring environmentalsurfaces after cleaning. It provides a 3-in-1screening tool for listeria species with abuilt-in verification for L. monocytogenesgiving visual results in 24-48 hours.Requiring nothing more than a small
portable incubator and a UV flash light, thetest is simple to use and self-contained tominimise the risk of cross contamination.The procedure is a simple swab, snap and
squeeze, incubate and read using the nakedeye. The visual result can be read at anypoint and any number of times during theincubation period to get the earliestindication of the result.The device consists of:
l A 1-inch foam swab prewetted with aneutralising buffer that allows bacteria torecover, grow and to counter the inhibitoryeffects from any residual sanitiser that mayremain on the test surface after cleaning.The large foam swab has been validated tocollect and detect listeria after swabbingeither 4 x 4-inch or 12 x 12-inch surface areaand detect listeria at low numbers afterdrying overnight at room temperature to
apply stress that typically results in a largeloss of viability.
l The bulb of the device contains a newimproved media that is released when thedevice is activated by a simple snap andsqueeze action. The media is selective forlisteria and reduces the incidence ofpresumptive positive results from bacteriawith similar diagnostic characteristics, suchas enterococci. The chromogenic mediarelies on the ability of listeria to hydrolyseesculin enzymatically and produce ablackening of the media.
l The media in the bulb also contains afluorogenic substrate which is hydrolysedby pathogenic strains of listeria viz L.monocytogenes to release a product thatfluoresces green under low level UV light.The fluorescence is visible in the media butalso binds to the plastic of the swab tubewhich makes it more vivid and a definitivepositive result.InSite L. mono Glo showed very good

The procedure is a simple swab, snap and squeeze, incubate and read using the naked eye.

validated with >95 strains of listeria speciesof which >55 were L. monocytogenes from12 serotypes, and all were detected. Morethan 35 non-listeria species were alsotested and none of the staphylococci,bacilli, coliform or enterobacteriaceae weredetected even at very high inocula. Someenterococci were detected but only at highinocula of ~500,000cfu.The time to detection with a high
probability of detection was dependent onthe inoculum. At the lowest inocula at 1-10cfu per swab area on both 4 x 4 and 12 x 12inches dried on to stainless steel coupons,the test required 48 hours to detect andverify a positive. The limit of detection after 30 hours’
incubation was 25 cfu/swab whereas after24 hours’ incubation the limit of detectionwas >1000 cfu (see Table 1).
Cost effective format
The test format of InSite L. mono Glofacilitates cost saving since a neutralisingswab and selective media are contained in asingle, pre-assembled, ready to use device.The foam swab enables both large andsmall surface areas to be covered and itsnarrow diameter also enables samples to becollected from small crevices and nozzles. Minimal expertise is required to activate
and read the test and no opening ortransfers are required, thus minimising therisk of cross contamination. Presumptive positive results for both
listeria species and L. monocytogenes areobtained in 24, 30 and 48 hours giving arapid semi-quantitative result as well asscreening out the negative samples in 48hours.The media is compatible with other tests
such as BAX System PCR tests to confirm
the presence of the pathogen in threehours or less. Similarly, the media givingpresumptive positive results contains viablecultures from which bacteria can beisolated and identified by conventional ormodern methods, and source tracking usingfingerprinting methods such as theRiboPrinter.InSite L. mono Glo is a convenient, cost
effective, rapid screening tool for listeria inenvironmental samples. n
