Suitability teStSSyStem

Total Organic Carbon (TOC) is a general test for the presence of organic compounds in water samples. For purified pharmaceutical waters, the United States Pharmacopeia (USP), European Pharmacopoeia (EP), Japanese Pharmacopoeia (JP), Chinese Pharmacopoeia (ChP) and Indian Pharmacopoeia (IP) all require TOC to be monitored and reported for pharmaceutical-grade waters.

One ubiquitous test that is accepted globally as a means of confirming acceptable TOC analyzer performance, is the System Suitability Test, sometimes referred to as SST. W hile pharmacopeial bodies may have their own version of the test, the concept is the same: if an analyzer can successfully oxidize and compare two different sources of organic compounds according to individual acceptance criteria, it can be used to monitor TOC levels in pharmaceutical waters.

Pharmaceutical and medical grade waters used in product manufacturing must meet conductivity requirements as specified in USP <645>/ EP 2.2.38, where the off-line conductivity must be less than 1.3 μS/cm at 25°C. Deionized water has a theoretical “purity” of 0.055 μS/cm at 25°C, so the question is often asked why “ultrapure” water for injection (WFI) and purified water (PW) is considered acceptable in these applications when it displays conductivity levels 20 times the theoretical “purity” of water. T he answer is simple; atmospheric carbon dioxide (CO2) readily dissolves into ultrapure waters, “contaminating” the water with conductive ions and adding approximately 1.0 μS/cm of conductivity to ultrapure water at 25°C. As such, appropriately accurate/stable CRMs in the range of interest (0.055-1 μS/cm) are not available, and alternative approaches to validating the detection limits, linearity, precision and accuracy, of conductivity meters are necessary.

Validating the detection limits, accuracy and precision of a conductivity sensor in ultrapure water has been very elusive using traditional calibration solution schemes. T herefore, ERA is offering a three solution Validation Kit that will allow an accurate, verifiable, and approved approach to validating the sensor for your USP <645> methods. T he kits come in single-use 125 mL volumes as well as multi-use 500 mL bottles. Each Validation Kit consists of three (3) conductivity calibration solutions. Measurements are made at each level and the results used to determine the Method Detection Limit (MDL), Limit of Detection (LOD) and Limit of Quantitation (LOQ). In addition, the data are used to verify probe accuracy and linearity. I would like to share with you two significant milestones that we have recently experienced.

The first involves designing and moving into a new facility in Golden, CO. Our business requires sizable amounts of lab and analytical space; something we had grown out of in our previous facility. We are excited that this new space has improved and increased our manufacturing and analytical capabilities, allowing us to positively impact the quality of our products and breadth of our offerings.

T he second milestone includes some significant anniversaries. In December 2006, a small company named ERA was purchased by Waters Technologies Corporation. At that time, ERA was celebrating 30 years in business and Waters 48. ERA was one of the largest proficiency testing and reference material suppliers and Waters a global leader in developing analytical instrumentation. With the publication of this catalog, we mark our fifth anniversary as part of the Waters organization and 35th year in business.

What do we celebrate with these anniversaries? How has ERA changed? Well, although we are still ERA, we have proudly added our Waters affiliation and now you see us as ERA, A Waters Company. We have improved our website and online data tools by housing all of our servers in the Waters’ server facility. We have added staff to improve our ability to meet your needs with new products and personalized customer service. We have matured, expanded our laboratories and made significant operational improvements.
