Why Prepare a Sample?It is often necessary to isolate specific compounds for qualitative and quantitativeanalysis. An effective sample preparation technique should remove interference, concentrate analytes for greater sensitivity, or modify the sample to achieve analyticalcompatibility. Without a pure, dry high yield sample analytical results are often meaningless. For this reasons, an effective sample preparation step is crucial for successful analysis.
Defining the SampleThere are three integral components to a sample:
Matrix: The body of the sample from which you are extractingAnalyte(s): The component of the sample you are trying to extractInterferences: Other components of the sample which need to be separated from the
analyte in order to effectively clean-up the sample
Traditional Sample Preparation TechniquesSample preparation is still often performed by archaic, time consuming, and inefficient methods. Of these techniques Liquid/Liquid Extraction is the most common.
Liquid/Liquid (L/L) extraction requires taking a raw sample, usually dissolved in anaqueous, buffered solution, and placing it in a separatory funnel with an organic solvent such as Ethyl Ether, Petroleum Ether, Methylene Chloride etc. The funnel isshaken vigorously, often for long periods of time, in order to distribute the samplecomponents between the immiscible solvent and aqueous layers. The organic compounds of interest are distributed in the organic solvent layer while the inorganicsalts, acids, bases, and other water soluble components remain in the aqueous layer.
The layers are then allowed to equilibriate and separate. The solvent layer is drawnoff and the aqueous layer is extracted two more times. The solvent extracts are combined, washed with aqueous solution, and dried through anhydrous sodium sulfate. The solvent is then evaporated from the extract to concentrate the analytes.
As described, L/L extraction is very time consuming, tedious, and costly. There aremany steps, leaving a large margin for error and analyte loss. An immense amount of expensive organic solvents are used, which need to be handled and disposed ofcarefully.
To summarize, L/L sample preparation can pose the following problems:! Time consuming! Tedious and difficult procedure! Expensive due to use and disposal of large amounts of solvents! Use of large quantities of toxic and flammable solvents! Wet, contaminated extracts! Results tend to be irreproducible

What is a Bonded Phase?The base silica (or polymer) used in solid phase extraction sorbents (or resins) consists of fine porous particles with a large surface area. This surface area permits adegree of functional interaction with the sample molecules passing through.
Silica gel is functionally modified by bonding organosilanes to the silanols to form abonded base.

What is endcapping?After bonding an organo-phase to the silica substrate, residual silanols remain due to steric exclusion. Since the free silanols are an adsorptive interaction they may compete with a different mechanism of the bonded phase. Endcapping the residualsilanols with a similar but lower molecular weight organo-phase ensures uniformmechanism interactions. In some specific applications residual silonal interactionsmay be desired. There are some non-endcapped sorbents available for use in this typeof application.
What are polymeric resins?Although silica based sorbents are most common, there are also polymer-based resinsavailable for use in solid phase extraction.
Polymeric resins work on the same principles as silica sorbents, however a polymertakes the place of the base silica. Similar to silica-based sorbents polymer-basedresins can be functionalized by either non polar, polar, or ionic functional groups.
There are some advantages to polymeric resins including:! Resins can be subjected to the full range of the pH scale (1-14), where as
sorbents break down in extreme pH conditions.! There are no residual silanol interactions because polymer based resins are
not adsorptive as are silica gels.

PolarityTwo of the four retentionmechanisms, normal phase,and reverse phase, arerelated to the physicalproperty of polarity.Polarity is often defined asthe separation of electronic charge of a molecule or ion. The electronic charge can belocailized in the functionalgroup of a molecule offering a polar site forinteration.
The Four Retention Mechanisms
PolarityTwo of the four retention mechanisms, normal phase and reverse phase, are related to the physical property of polarity. Polarity is often defined as the separation of electronic charge of a molecule or ion. The electronic charge can be localized in thefunctional group of a molecule offering a polar site for interaction.
Normal PhaseNormal phase solid phase extraction is used to extract polar compounds from lesspolar or weaker solutions. Polar compounds are adsorbed onto the sorbent surface viapolar functional group interactions. More polar or greater solubilizing solvent thanthe sorbent is used to elute the analyte of interest form the sorbent.
The advantages of normal phases are their compatibility with aqueous solutions andtheir ability to reversibly retain polar compounds.
Reverse PhaseReverse phase SPE is used to extract non-polar or hydrophobic compounds frommore polar or weaker solutions. Non polar compounds are adsorbed onto the surfacevia non polar functional group interactions. A more non-polar or greater solubilizingsolvent than the sorbent is used for elution.
The advantages of reverse phases are their excellent compatibility with aqueous samples.
Ion exchangeIon exchange mechanisms are used to extract ionic compounds from aqueous andnon-aqueous solutions. Ionic compounds are exchanged on the sorbent surface viaopposite charges attracting eachother. A solution that will neutralize the charge interaction and has a stronger affinity to the ionic analyte is used for elution.

Ionic strength! counter ion concentration less than the analyte favors extraction! Counterion concentration greater than the analyte favors elution
pH
For acidic compounds:! extract 1-2 pH units above the analytes pKa! elute at an acidic pH
For basic compounds:! extract 1-2 pKa units below the analytes pKa! elute at a neutral to basic pH
Organic solvent! for ionic compounds that lose solubility in aqueous buffered eluates,
add 40-80% water miscible organic solvent
Use a weak ion exchanger if the analyte has a strong ionization. For exampleuse a weak cation exchanger if your analyte is a strong cation. Using a strongcation exchanger in this scenario may cause the analyte to be irreversiblybound to the sorbent if a counter-ion is not available to displace the analyteform the sorbent bed.
Size Exclusion MechanismsTo separate large molecules from a sample, size exclusion mechanisms areoften used. Specific resins designed for sized exclusion implement a limitingpores size to allow small molecules larger than the pores are excluded and passthrough the resin bed. This method is often used in applications such as protein and DNA purification.

Cartridge SelectionThree Basic Extraction MechanismsThere are three basic extraction mechanisms used in solid phase extraction:non-polar, polar, and ion-exchange. Sorbent mechanism selection is primarilybased upon the funcional groups present on the analyte and the composition ofthe sample matrix.
Each sorbent within a given group exhibits unique properties of retention andselectivity which may be very specific for a given anaylte. As a result, even ifan extraction calls for a non-polar extraction mechanism, it may still be neces-sary to test several sorbents to find the optimal balance between high recoveryand good clean-up.

Spe-ed Flangeless CartridgesToday’s fully automated SPE processing systems require an SPE cartridge configuration that has beenspecially adapted to fit these instru-ments. Applied Separations’ Flangelesscartridges do not have the flange, ortab that is found at the top of AppliedSeparations’ Standard SPE cartridge.
They are identical in every other way.
Applied Separations provides the same widevariety and high quality packings in these specially modified cartridges as available in its standard SPE cartridges.
The flangeless design allows for closer packing in specialized automated SPE systems, such as the Gilson® SPE215. This format will accommodate 96 x 1mL cartridges, or 48 x 3mL cartridges in the standard 96-well footprint.
If non-standard sorbents, packingamounts, or cartridge sizes arerequired, Applied Separations canprovide them.
Empty cartridges with a wide varietyof different frit material are alsoavailable.

MiniSpe-ed CartridgesMiniSpe-ed cartridges contain the widevariety of sorbents found in standard Spe-ed cartridges. The MiniSpe-edconfiguration can be easily used in manualsyringe operations or on a vacuum mani-fold. MiniSpe-ed cartridges are also fullycompatible with instruments such asMilliab™. MiniSpe-ed cartridges have a1.0mL volume. For silica gel, this is 450mg.
MiniSpe-ed Plus CartridgesMiniSpe-ed Plus cartridges have the sameconfiguration as MiniSpe-eds, but holdalmost three times as much sorbent. TheMiniSpe-ed Plus cartridge has a volume of 2.8mL. The weight of different sorbents will varydepending on their bulk density; for silica gel this is 1265mg.
ConvenienceMiniSpe-ed / Plus cartridges stack together, interchangeably, allowing the use of more thanone sorbent, or further increasing capacity. Both cartridge types are available empty or pre-fritted for your own packing. The MiniSpe-ed and MiniSpe-ed Plus cartridges use standard20µ hydrophobic polyethylene SPE frits. Other frit material is available.

Applied Separations, Inc., has developed Spe-ed Advanta, a new chemically modifiedpolymeric resin for solid phase extraction (SPE).Spe-ed Advanta is packed in standard 1, 3 and6mL SPE cartridges, and in 96-well plates (othercartridge sizes available upon request) and iseffective in removing polar organic compoundsfrom aqueous samples.
Unlike standard C18 and polystyrene divinyl-benzene SPE cartridges, Spe-ed Advanta is:• easily wetted with water• requires no tedious conditioning steps• eliminates poor recoveries associated with the drying of a
conditioned sorbent bed.
Spe-ed Advanta markedly increases the recoveries of polar compounds fromaqueous matrices when compared to C18 and polystyrene divinylbenzene SPEcartridges. Spe-ed Advanta enhances the preparation of pharmaceutical, envi-ronmental, biological, clinical, biotechnical, food and cosmetic samples.

Amberlite® XAD Spe-ed Amberlite® XAD ResinsXAD-2, XAD-4, XAD-7, and XAD-16 resins are neutral, macrop-orous polymeric beads and derive their adsorbative characteristicsfrom their macroreticular porosity, pore size distribution, high surfacearea, and chemically homogeneous non-ionic structure. These resinsare primarily used to extract non-polar, hydrophobic compounds fromaqueous or polar solutions.
Pre-cleaned and Pre-swollen to Eliminate VariabliltySpe-ed cartridges are prefilled with measured amounts of speciallyprepared and cleaned XAD resin for critical analytical use.Commercial XAD resins are normally not suitable for analytical orother applications requiring high purity. Applied Separations’ XADproducts are given mulitiple solvent washes to assure the highest levelof quality. Variablity problems are eliminated with pre-swollen, pre-conditioned beads. To ensure continuing quality, the cartridge with theXAD resin is then capped and sealed to maintain the resin in itshydrated form.
Resin SelectionXAD-4, XAD-7, XAD-16,XAD-1180, XAD-2000, andXAD-2010 differ in poresize, surface area, and chemical composition. Select a pore size largeenough to allow the analyteto enter the pore and adsorbonto the pore surface.

PuriPON Columns eliminate the need forcentrifugation and phenolextractions because thedenatured protein isretained on the top fritand the phenol isadsorbed to the gelmatrix.
PuriPON ColumnsReliable methods for preparing pure DNA and RNA are essential formolecular biology research. Newly synthesized oligonucleotides oftenneed to be purified, desalted or exchanged into new buffers.
PuriPON Columns are prepacked, gravity flow columns used for therapid purification of nucleic acids, oligonucleotides and proteins. Eachcolumn contains a medium grade of Applied Separations’ Spe-ed DEXgel filtration media packed between two polyethylene frits. The fritsalso protect the gel from running dry under gravitational buffer flow.
Spe-ed DEX is biologically inert, bead-formed, cross linked dextrangel. Molecules that cannot enter the porous beads because of theirlarge size are eluted first. The smaller molecules enter the beads andare eluted in order of decreasing size.

Vacuum ManifoldPlace the Spe-ed DRI cartridges on your vacuum manifold.Place your collection glassware in the vacuum manifold.Introduce the liquid into the Spe-ed DRI cartridge and draw vacuum.Draw vacuum until as much liquid as possible has been extracted from

Today’s fully automated SPE processing systems requirean SPE cartridge configuration that has been speciallyadapted to fit these instruments. Applied Separations’Flangeless cartridges do not have the flange, or tab that isfound at the top of Applied Separations’ Standard SPEcartridge.
They are identical in every other way.
Flangeless Standard Applied Separations providesthe same wide variety and
high quality packings in thesespecially modified cartridgesas available in its standard
SPE cartridges.
The flangeless design allows for closer packing in specialized automated SPE systems, such as the Gilson® SPE215. This format will accommodate 96 x 1mL cartridges, or 48 x 3mL cartridges in the standard 96-well footprint.
If non-standard sorbents, packing amounts, or cartridge sizes are required, Applied Separationscan provide them.
Empty cartridges with a wide variety of different fritmaterial are also available.

If gravity flow of an aqueous solution is desired,select a specially treatedpolyethylene hydrophilic frit.
FritsPolyethylene Hydrophobic FritsPolyethylene hydrophobic frits are the standard and are used in mostcommercial SPE products. They are also used with non-aqueous solutions such as solvents, alcohols, acids, bases, halogenated and aromatic hydrocarbons, and ketones. These hydrophobic frits can bewetted with organic solvents.
Polyethylene Hydrophilic FritsPolyethylene hydrophillic frits are used for general filtration and as forclarification of aqueous solutions and certain other solutions. Thesehydrophilic frits are specially treated and can be wetted with aqueoussolutions. This frit is ideal for gravity flow of an aqueous solution.
PTFE and Stainless SteelPTFE and stainless steel frits can be used to filter almost any organicsolvent. They are used for trace analysis and high sensitivity workwith solvent detection systems which are not compatible with polyethylene or polypropylene frits. PTFE is a hydrophobic membraneresistant to most organic solvents, acids, and bases.
PolypropylenePolypropylene frits are hydrophobic and are best used for lowextractables. Polypropylene frits are similar to PTFE frits in their filtration behavior and serve as an economical alternative to the moreexpensive PTFE frits.

Spe-ed Mate-10The Spe-ed Mate-10 offers a compact option for preparation of up to 10 samples.
Special Features of the Spe-ed Mate Vacuum Manifold! Universal Collection Rack - Provides eluate collection in test tubes,
volumetrics, LSC vials, WISP cups, micro and standard centrifuge tubes, and other analytical containers. Custom racks are also available.
! Removable Luer-fitted Needles - A variety of sizes, lengths, and materials, including Teflon®, stainless steel, and polypropylene - compatible with most solvents, acids, and samples.
! Chemically Resistant Cover - Inhibits surface deterioration and contamination.
! Individual Control - With the addition of stopcocks, the simultaneous vacuum, extraction, elution, or filtration of samples can be controlled.
! Special Coating with Inert Polymer available - for long life and uninterrupted operation in very hostile and corrosive environments.

Spe-ed Mate Glass Vacuum ManifoldSpe-ed Mate Glass Vacuum Manifolds offer the advantages of our standardSpe-ed Mates PLUS are an easily cleaned, chemically-inert, clear glass chamber for visually monitoring vacuum operations. Simultaneous vacuum,extraction, elution, or filtration of samples can be controlled for each valve.The individual vacuum gauge and bleed valve assembly allows control andmonitoring of vacuum applied to the chamber. Luer connections on the valveassemblies accept a broad range of SPE cartridge sizes from 1mL to 60mL.Adjustable racks inside the glass walled vacuum chamber accommodate a variety of collection vessels. Eluants are deposited directly into collection vessels via polypropylene needles.
