Separation of a complex sample: Extract from Hoodia gordonii
Interrogation of information-rich samples requires a UPLC system that provides the highest quality information. The separation was achieved using the ACQUITY UPLC I-Class with a Xevo® G2 QTof and a 1.8-µm ACQUITY UPLC HSS T3 2.1 x 100 mm Column.
Extract of Hoodia gordonii was analyzed in collaboration with Ikhlas A. Khan, Ph.D., and Bharathi Avula, Ph.D., from the National Center for Natural Products Research, Research Institute of Pharmaceutical Sciences at The University of Mississippi.

UPLC vs. UHPLC: A clear difference
UPLC separations are defined by the use of sub-2-µm particle column packing materials that reduce dispersion during the separation. When total dispersion from the column and the system together is minimized, resolution and sensitivity are maximized. To realize the full benefits of sub-2-µm columns, the LC instrument must be optimized to preserve the high-efficiency separation.
In this example, the impurity analysis of tetracaine was run under the same conditions on the ACQUITY UPLC I-Class System (system dispersion of 5.5 µL) and a UHPLC system (system dispersion of 20 µL). Although the UHPLC system was optimized for lowest possible dispersion and system volume, the impact of system dispersion is clearly evident. Significantly better resolution was achieved with the ACQUITY UPLC I-Class System.

Power for ballistic gradients
The low dispersion characteristics of the ACQUITY UPLC I-Class System allow you to push your separations to a new level. This ballistic gradient for a series of anesthetics achieved standard deviations for retention time as low as 12 ms for six replicates. The separation achieved a peak capacity of 46 in 10 s with a 1/2 height peak width as low as 88 ms.

Precision for reproducible injections
The ACQUITY UPLC I-Class System (SM-FTN) delivers precise, accurate injection volumes from 0.1 µL to 1.0 mL. For this series of anesthetics analyses, the injection linearity was assessed from 0.1 µL to 2.0 µL with six replicates at each level. Excellent linearity, repeatability, and accuracy were achieved across the injection volume range.

With carryover minimized for the assay of omeprazole, the calibration range on the ACQUITY UPLC I-Class System coupled with the Xevo TQ-S was determined to be 500 ag to 10 pg on-column with a correlation coef-ficient of 0.99997 across greater than four orders of magnitude.
Designed for mass spectrometry...
The ACQUITY UPLC I-Class was paired with a Xevo TQ-S for the analysis of omeprazole, achieving a quantification range of 500 ag to 10 pg on-column. No detectable carryover within 20% of the limit of quantitation was observed after the injection of the highest concentration standard.
