In some countries government regulations require a completenitrogen balance for effluents. Besides ammonium, nitrite andnitrate, organic nitrogen compounds have to be determined –the sum of all these nitrogen compounds is called total nitro-gen. The Standard Method DIN/EN/ISO 11 905-1 describesthe procedure for the determination of total N. After formationof nitrate by oxidative decomposition of all inorganic and or-ganic nitrogen-containing substances, a subsequent nitrate
determination gives the total nitrogen content in mg N per li-tre.In samples with low nitrate and nitrite concentrations thismethod directly gives similar results to the determination ofTotal Kjeldahl Nitrogen TKN (including only ammonium andorganic nitrogen compounds). In the presence of detectableamounts of nitrate/nitrite, the TKN can be determined by sub-tracting nitrate/nitrite nitrogen from total nitrogen.

Together with the chemical oxygen demand (COD) the bio-chemical oxygen demand (BOD) is the most important sum-mary parameter for an estimation of the pollution of a wastewater. The BOD is defined as the amount of oxygen per vol-ume, which is used by the microorganisms at 20 °C for oxida-tive degradation of the organic substances present in thewater. The AOX content represents the sum of organically boundhalogens (chlorine, bromine, iodine) which are adsorbable toa suitable adsorbent; their concentration is expressed aschloride. The rapid test NANOCOLOR® AOX 3 allows the determina-tion of AOX in natural waters, industrial or municipal wastewaters and sea water. The test is based on an easy three-step procedure:1. The chemical oxygen demand (COD) is one of the most im-portant parameters for the evaluation of industrial or munici-pal waste waters. It is defined as the amount of oxygen – inthe form of an oxidizing agent – consumed in the oxidation oforganic water components. For standard methods (titration)and in cuvette tests, potassium dichromate in sulfuric acidhas long been used as an oxidizing agent for COD analysis.In addition to organic substances, inorganic substances canbe simultaneously oxidized. Particularly higher chloride concentrations are known to inter-fere. They may cause falsely higher results; but more oftenchloride interference leads to totally irreproducible results.Therefore, during COD analysis, chloride ions are usuallymasked by mercury salts. However, mercury is toxic, maycause damage to the nervous system of human beings andanimals and in addition is persistant in nature. So meanwhilein many countries the use of mercury compounds for analyti-cal purposes is subject to stringent regulations. Application ofmercury-consuming analytical methods is forbidden as soonas they can be replaced by mercury-free methods.For the determination of the COD with tube tests one cannotsimply omit the mercury, since this may lead to false or nonreproducible results, due to chloride/salt interferences. Andusing a chemistry different to the potassium dichromatemethod (e.g. permanganate or manganese) may cause con-spicuous difficulties in comparison to results obtained withstandard methods or regular COD cuvette tests.Now, MACHEREY-NAGEL has developed a mercury-freeCOD test, which uses the established dichromate chem-istry, but substitutes the most toxic component – themercury salts – by a non-toxic chloride elimination tech-nique using a special cartridge.
Convenient chlorine removal cartridges eliminate chlo-ride interferences and the reliable dichromatechemistry leads to reproducible and compara-ble results. The basic procedure of theregular COD cuvette test has not beenchanged, so no additional methods mustbe learned.

The metabolic activities of microorganisms have a substantialinfluence on the purification efficiency of a sewage plant.Therefore it is important for the operator of a sewage plant toknow whether and in which way introduced waste waters af-fect the metabolic activity of the microorganisms (the activat-ed sludge activity). Microorganisms contain a number of enzymes (among othersdehydrogenases), which are responsible for the degradationof the waste water constituents. These enzymes are presentin all living microorganisms. The detection of dehydrogena-ses is therefore the proof for the occurrence of living biomass.The long-established tests for determination of the dehydro-genase activity – standardized as German Standard Methods(DEV) – are based on the so-called TTC method. In practice an analysis often recovers only the quantity of met-als which is present in the sample as dissolved ions. If the to-tal content of a metal is to be determined, particularly samplesof highly contaminated water or industrial waste water mustbe decomposed to avoid falsely negative or falsely low re-sults. Usually, by applying acid and heat, undissolved metaloxides are dissolved, metal ions are released from complex-

NANOCOLOR® reagent set Sludge 50Liquid reagents for oxidative decomposition of sludge containing heavy metals In Germany, the use of sludge for soil improvement in farmingand forestry and in gardening is regulated by government reg-ulations which define the maximum contents of 7 heavy met-als (Cd, Cr, Cu, Hg, Ni, Pb, Zn) allowable in sludge for theabove applications. The NANOCOLOR® reagent set sludge50 allows even non-chemists to determine these contentswith high accuracy. It is, however, recommended that you ar-range for the supplier of the kit to provide instructions to thosemembers of your team likely to be involved in its use. With the exception of mercury and lead all of the above met-als can be determined with the NANOCOLOR® analyticalsystem. The photometers NANOCOLOR® 250 D and 350 Dcannot be used for this sludge analysis. When using the pho-tometers NANOCOLOR® Linus, 400 D or 300 D, you can di-rectly read results as mg/kg dried residue of sludge (S). NANOCOLOR® membrane filters Removal of turbidities (fine filtration) is recommended for tworeasons:
Turbidities interfere with photometric determinations, in-tense turbidities must be removed prior to the analysis.
When both soluble and total amounts of a substance(e.g. iron, manganese, COD etc.) have to be deter-mined, part of the sample has to be filtered to removeundissolved particles. The German Standard Methodsuse a pore size of 0.45 µm as standard for differenti-ation between dissolved and undissolved particles.
Additionally we offer a pore size of 1.2 µm.

NANOCONTROL standard solutionsReagents, measuring equipment and personal handling are al-ready subject to validation, or frequent internal control in manylabs. Therefore, numerous customers rely on our NANOCON-TROL standards to obtain an objective picture of the quality ofanalyses carried out with the NANOCLOR® system.NANOCONTROL standards are ready-to-use standard solu-tions, which contain one or several substances in known con-centrations. Separate spike solutions (NANOCONTROL100+ solution) allow tracing of interferences, which may bepresent in the sample. Composition and concentrations ofmultistandards meet the requirements of specific user groupse. g. in waste water treatment, drinking water production andothers. The standards are supplied with a documentationform, in which expected concentration and given tolerancerange (from/to) are stated for each parameter; in additionthere is plenty of space for documentation of quality control.Application of NANOCONTROL standards permits easy ana-lytical quality control and simplifies the corresponding docu-mentation. The consistent use of NANOCONTROL standardsleads to an objective proof of the accuracy of the analysis car-ried out with the NANOCOLOR® system and confirms the op-erator’s confidence in the reliability of the measured results.

The principle of standard addition is, that you increase theconcentration of a sample in defined steps. The recovery rateis then an indication of possible interferences. When con-stant, proportional deviations occur, they can be counterbal-anced; thus, the result can be corrected.

Besides application of multistandards for quality control an-other means for certification of analytical results is the partic-ipation in a NANOCONTROL interlaboratory trial.The principle of the interlaboratory trial is, that all participantsobtain an unknown sample, which they analyse independent-ly with the instrumental equipment and tests available. Theresults are then returned to the company who organises theinterlaboratory trial and evaluated in accordance with DIN38 402 part 41 and 42. While for accredited laboratories thismethod has been obligatory since a long time, it is not yet es-tablished in the field of routine process control. We recom-mend a participation once or twice per year. The MN interlaboratory trial is suited for checking your individ-ual working procedures as well as the waste water parame-ters determined with NANOCOLOR® tube tests. 100 mlsolution for the interlaboratory trial are sufficient for at leastone repeated test of each of the following parameters: COD,NO3-N, NH4-N, total N and total P.
Procedure of a NANOCONTROL interlaboratory trial1. The participant gets 1 bottle with 100 ml solution for the
interlaboratory trial. The concentrations of this solutionhave been certified by an independent DIN laboratory.
2. The concentrations of the solution for the interlaboratorytrial have been adjusted to the ranges of the NANO-COLOR® tests and are used without dilution.
3. Together with the solution, the participant obtains a pre-printed evaluation chart, instructions for use and an en-velop for returning the chart with the analytical results.
4. At MACHEREY-NAGEL all returned protocols are sub-jected to an objective evaluation.
5. Finally every participant obtains an individual evaluationfor every returned result, a certificate for participationand technical support, if necessary.
