Organs of Immune System

Primary lymphoid organs-maturation of lymphocytes.

Secondary lymphoid organs- trap antigens & provide sites for mature lymphocytes to interact with that antigen.

Tertiary lymphoid tissues- cutaneous associated lymphoid tissues.

Primary Lymphoid Organs

Immature lymphocytes generated in bone

marrow mature, becomes specific for a particular antigen in central lymphoid organs.

Such educated cells are immunocompetent cells.

T cells arise in thymus . B cells in bone marrow.

– First organ to become lymphoid in the body

– Site of T cell development and maturation.

– Precursors of lymphocytes reach thymus–differentiate into thymocytes.

– Maximum size- just before birth.

– Grows till 12th year.– Undergoes involution after

puberty.

Effects of thymectomy or congenital thymic aplasia:

● deficient CMI (Di George syndrome)● increased chance of getting infectious

diseases● diminish antibody response towards

Bone Marrow

All lymphocytes originate in bone marrow. T lymphocytes develop in thymus. B lymphocytes develop in bone marrow itself.

Phagocytose foreign materials including microorganisms especially from local tissues.

Help in the proliferation and circulation of T & B cells.

Central cells of the immune system-

LYMPHOCYTES– They are responsible for:

1. adaptive immunity.

2. immunological aspects of diversity, specificity, memory & self / nonself recognition.

Functions of other WBCs

1. engulf and destroy micro organisms

2. present antigens.

3. secrete cytokines.

CD4 cells = MHC II Restriction - TWO types – TH1 & TH2. TH1 – secrete γ interferon & IL – 2 , which activate macrophages & T cells promoting CMI. TH2 – secrete IL – 4 , IL – 5 , IL – 6 = stimulate B cells to form Abs.

CD8 cells = MHC I Restriction – can kill & lyse target cells carrying new or foreign Ags = down regulate immune response.

Major HistocompatibilityComplex (MHC)

History Early work of Gorer (1930) on

antigens responsible for allograft rejection in mice led to the discovery of the ‘MAJOR HISTOCOMPATIBILITY COMPLEX (MHC)’.

Development of congenic & recombinant strains of mice by Snell enabled the detailed analysis of various loci of this complex.

Dausset pioneered studies on HUMAN LEUKOCYTE ANTIGENS MAJOR HISTOCOMPATIBILITY ANTIGENS in human being.

Benacerraf & colleagues established genetic basis of immune response.

For their work on MHC & genetic control of immune response, Snell, Dausset & Benacerraf were awarded Nobel Prize for medicine in 1980.

Major HistocompatibilityComplex (MHC)

History Early studies on MHC were

carried out on mice.

All species of animals including HUMAN BEING were found to possess a similar complex of genes on a segment of one chromosome pair.

These genes coding for…………. 1.Class I proteins ---- determining histocompatibility & acceptance or rejection of allograft.2. Class II proteins ---- regulating the immune

response.3. Class III proteins ---- including some

components of the complement system.

Major HistocompatibilityComplex (MHC)

History Name histocompatibility complex because of its

discovery based on transplantation experiments.

Human MHC antigens are found on surface of leucocytes hence synonymous with Human Leukocyte Antigens (HLA) & MHC complex of genes with the HLA complex.

HLA Complex– HLA complex of genes located on short arm of chromosome 6.

– It is comprised of three separate clusters of genes:

1. HLA class I A,B & C loci.

2. Class II or D region DR DQ & DP loci.

3. Class III or the complement region genes for complement components C2 & C4 of the classical pathway, properdin factor B of alternative pathway, heat shock proteins, tumor necrosis factors C.

Heavy chain (alpha) and “microglobulin” (beta two)

Heavy chain is 45 kilodaltons, has three domains + a transmembrane component (40 aa) + a cytoplasmic tail (30 aa)

The three alpha domains are called: 1, 2, & 3

1 and 2 interact to present processed Ag

Processed Ag is optimally a monomer

MHC - II An alpha and beta chain, 33 kDA

and 28 kDA, respectively.

Chains are non-covalently associated.

Each chain has two domains.

1-1 interact to present processed Ag

Processed Ag is optimally 13-18 aa

2 & 2 are part of immunoglobulin super family

HLA Typing

– SEROLOGY used to be the ‘gold’ standard. Now being superseded by molecular techniques as they become more robust and time efficient.

– CELLULAR rarely used now. Originally used for Class II typing.

– MOLECULAR fast becoming the method of choice. Many laboratories test of choice.

HLA Typing

– Earlier antisera were obtained from multiparous women ,as tending to have antibodies to HLA antigens of their husbands owing to sensitization during pregnancy.

– Now monoclonal antibodies to HLA antigens have been developed.

HLA Typing

– HLA Typing is done serologically by MICROCYTOTOXICITY ( microlymphocytotoxicity) which tests for complement mediated lysis of peripheral blood lymphocytes with a standard set of typing sera.

– Serological typing not possible for HLA – DR antigens, which are detected by mixed leukocytic reaction (MLR), & primed lymphocyte typing (PLT).

– Genetic methods – RFLP, gene sequence – specific oligonucleotide probe typing.

MHC Restriction

T cells respond to processed antigens on the macrophages & other secretory cells only when they are presented along with the self MHC antigens - k / a MHC restriction.

MHC – I molecules ---- CD 8 + T cells ( Cytotoxic T or Suppressor cells).

MHC – II molecules ---- CD 4 + T cells ( T Helper cells or Inducer cells).
