Strategies for lipid secretion Jinjing Wang 2009-09-25 Slide 2 How does Lipid secret from yeast cells ? Halo forming Slide 3 The process of lipid transportation across the membrane of oleaginous yeast Yarrowia lipolytica W29 Slide 4 Several algae strains that secrete lipid Ochromonas danica Chlorella vulgaris Spirulina platensis Trebouxia sp. Slide 5 plan Cell wall structure changes Algae cells can secrete lipid when cultured under environment pressure (e.g. Temperature). There should be some changes of cell wall structure to insure the lipid can be secreted into the medium. We can examine the changes of algae cell wall structure during lipid transportation across the cell wall using electron microscope. Some genes should be in charge of this process. When the structure of cell wall changes, there should be some differences in the expression of protein. Comparing the different expression of protein using Proteomics analysis, so we can find the genes controlling this process. Autolysis It has been reported that lipid is excreted because of diffusion. The lipid excreted from yeast cells or algae cells is 6-10% of the total lipid, this process often become a homeostasis. Introduce heat-inducible autolytic vectors to yeast cells or algae cells.
