Abstract
For decades, the use of fullerenes for biomedical applications (e.g. photodynamic therapy and
magnetic resonance imaging) has been considered promising. One of the greatest obstacles
for their utilization is their lack of solubility in aqueous system due to the hydrophobicity of
the fullerene cage. To overcome the inherent lack of solubility, we derivatized the fullerenes
through various chemical modifications.
In this work, the derivatization of the fullerene C60 was carried out via the Prato
reaction to obtain a new and versatile derivative for further conjugation with water-soluble
moieties.
The Prato reaction was applied on M3N@C80 (M = Sc, Lu, Y, Gd) and studied in a
great detail. We found that the reactivity and the regioselectivity of the adducts were highly
dependent on the endohedral metal type. The cycloaddition occurred first on a [6,6]-bond
(junction between two hexagons) and isomerized to the [5,6]-bond (junction between a
hexagon and a pentagon) via a sigmatropic rearrangement. We also prepared other Prato
adducts of M3N@C80 to understand the important factors involved in the reaction.
Interestingly, the rate of isomerization was dependent on two essential factors: the endohedral
metal size and the type of exohedral functional group (substituent on the nitrogen atom of the
pyrrolidine exohedral group). Whereas the [6,6]-to-[5,6] isomerization was completed in the
presence of small metals (M = Sc, Lu), the same isomerization process was found to be
reversible upon heating with larger metal clusters (M = Y, Gd). The final regioisomeric ratio
was found to be solely dependent on the size of the metal.
Since water-soluble derivatives from fullerene monoadduct are highly suspected to
dissolve in water via the formation of small fullerene cage aggregates, preparation of
polyadducts with polar groups may prevent the fullerene aggregation. Because single
molecule is desired for Gd@fullerene MRI contrast agent materials, multiple addition of

M3N@C80 was carefully studied since the polyadducts may prevent the formation of
aggregates. Preparation of polyadducts was carefully investigated, and surprisingly with
larger endohedral clusters of metallofullerenes (M = Y, Gd), highly regioselective bis- and
poly-addition was observed.
Preparation of biorelevant C60 derivatives was carried out by conjugation with a
peptide, by conjugation with amine-terminated polyethylene glycol or poly(vinylpyrrolidone).
Amine-terminated polyethylene glycol polymers were commercially available whereas the
amine-terminated poly(vinylpyrrolidone) polymers had to be prepared in house. Synthesis of
well-defined poly(vinylpyrrolidone) polymer with functional end-group was accomplished by
RAFT polymerization.
Previous work by Ikeda and co-workers showed that water-soluble fullerenes could be
obtained via complexation with a water-soluble host molecule (e.g. cyclodextrins). In this
work, a new design of the host (cavitand) was evaluated. We prepared a quinoxaline-based
cavitand with amide groups on the upper rim. The amide groups are expected to interact with
each other to form a cyclic array of hydrogen bonds, thus creating a large cavity.
Optimization of the molecular design was done by varying the steric hindrance of the R2
group of the upper rim, and conformational study was investigated by NMR and crystal
structure analysis.

Résumé
Depuis plusieurs décennies, l'utilisation de fullerènes pour des applications biomédicales (p.
ex. la thérapie photodynamique ou l'imagerie par résonance magnétique) a été considérée
comme prometteuse. L'un des obstacles majeurs pour leur utilisation est leur manque de
solubilité dans les systèmes aqueux à cause de l'hydrophobicité de la cage fullerène. Afin de
dépasser ce manque de solubilité intrinsèque, nous avons dérivatisé les fullerènes à travers
différentes modifications chimiques.
Dans ce travail, la dérivatisation du fullerène C60 a été effectuée par la réaction de
Prato afin d'obtenir un nouveau dérivé versatile pour ensuite être conjugué avec des parties
hydrosolubles.
Cette réaction de Prato a été effectuée sur M3N@C80 (M = Sc, Lu, Y, Gd) et étudiée
en détail. Nous avons trouvé que la réactivité et la regioséléctivité des adduits était hautement
dépendante du type de métal endohédrique. La cycloaddition a d'abord lieu sur une liaison-
[6,6] (jonction entre deux hexagones) puis isomérise sur une liaison-[5,6] (jonction entre un
hexagone et un pentagone) via un réarrangement sigmatropique. Nous avons aussi préparé
d'autres adduits M3N@C80 de Prato afin de comprendre les facteurs importants impliqués lors
de cette réaction. Fait intéressant, la vitesse d'isomérisation fût dépendante de deux facteurs
essentiels : la taille du métal endohédrique et le type de groupe fonctionnel exohédrique
(substituant sur l'atome d'azote du groupe pyrrolidine exohédrique). Alors que l'isomérisation
de [6,6]-à-[5,6] fût totale dans le cas de petits métaux (M = Sc, Lu), celle avec avec de plus
larges métaux (M = Y, Gd) fût réversible en chauffant. Le ratio final de régioisomères s'est
révélé être dépendant seulement de la taille du métal.
Étant donné que les dérivés hydrosolubles du monoadduit de fullerène sont
soupçonnés de se dissoudre dans l'eau par la formation de petits agrégats de cages de
fullerènes, la préparation de polyadduits comportant des groupements polaires pourrait

empêcher l'agrégation des cages. Sachant qu'une molécule seule est désirée pour les agents de
contraste IRM de Gd@fullerene, plusieurs additions sur M3N@C80 ont été étudiées
soigneusement étant donné que les polyadduits empêcheraient la formation d'agrégats. La
préparation de polyadduits a été étudiée avec soin et étonnamment avec de larges clusters
endohédriques de métallofullerènes (M = Y, Gd), une bis- et poly-addition hautement
régiosélective ont été observées.
La préparation de dérivés de C60 applicable pour la biologie a été réalisée par
conjugaison avec un peptide, par conjugaison avec du polyéthylène glycol ou bien du
poly(vinylpyrrolidone) à terminaison amine. Les polymères de polyéthylène glycol à
terminaison amine sont commercialement disponibles alors que les polymères du
poly(vinylpyrrolidone) à terminaison amine ont dû être préparés en laboratoire. La synthèse
d'un polymère poly(vinylpyrrolidone) bien défini avec un groupement fonctionnel terminal a
été réalisée par polymérisation RAFT.
Des travaux antérieurs par Ikeda et ses collaborateurs ont montré que des fullerènes
hydrosolubles peuvent être obtenus par complexation par une molécule hôte soluble dans
l'eau (p. ex. les cyclodextrines). Dans ce travail, un nouveau modèle de molécule hôte
(cavitand) a été évalué. Nous avons préparé un cavitand à base de quinoxalines avec des
groupes amides sur la partie supérieure. Ces groupes sont censés interagir les uns avec les
autres pour former un arrangement cycliques de liaisons hydrogènes, qui créeraient ainsi une
grande cavité. L'optimisation du modèle moléculaire a été effectuée en faisant varier
l'encombrement stérique du groupe R2 de la partie supérieure et l'étude conformationnelle a
été étudiée par RMN et par analyse de la structure cristalline.
