Data Acquisition and Processing Software State of the Art Quantitation

Acetaminophen, a commonlyused analgesic (pain reliever)and antipyretic (fever reducer)was analyzed. It is a challenging compound to run by mass spec-trometry as a result of its lowmolecular weight (151) andfragility. It was run in the positive ion mode by electrospray usingMRM mode monitoring of152/110.

The HPLC separation was accomplished at the flow rate of 200ul/min by employing25/37.5/37.5 water/acetonitrile/methanol with 0.1% formic acid using a Beta Basic C18 2.1 X 50 mm 3micron column injecting 5ul. The signal to noise, based on 3 sigma, was 36/1 on the 50 fg/ul injection. The concentration range run shows excellent linearity.

The transition 609 to195 wasmonitored using MRM. Thesignal to noise is better then90:1. Here a 5 microliter injec-tion volume is used. The chro-matographic conditions were3/7 water/Acetonitrile 2mMammonium acetate with 0.1%formic acid at a flow of 0.2milliliter per minute.
