Methods: Preparation of experimental animals, molecular characterization of LSDV circulating in Ethiopia, Sentinel herd monitoring, Experimental design for vaccine challenge experiment , Clinical and serological observations post challenge and Data Analysis Expected outputs from the project: Contribute important scientific information for vaccine production institutes Contribute to produce better immunogenic CPV vaccines in the future Economic losses due to LSD will be reduced. To identify molecular characterization of LSD virus circulating in Ethiopia, it gets opportunity to study further . Impact of the outputs on agriculture in Africa: If one of the transboundary diseases, Capripox disease could be controlled livestock and livestock products trade will be improved . Two different vaccines (Neethling strain and Kenyan SGPV ) have been widely used for the control of LSD in cattle populations in southern and eastern Africa)(OIE, 2010).They will be benefited from this study . The National Animal Health Diagnostic and Investigation Center (NAHDIC) is the referral and reference veterinary laboratory in Ethiopia. It is the center of excellence for animal disease surveillance, investigation, diagnosis and research which contributes substantial role in promoting export of animal and animal products, improvement of the livelihood of the smallholder farmers and pastoralists, provision of professional support for investors involved in animal farming and transfer technology for stakeholders. The implementation of quality system in NAHDIC has brought an important milestone for NAHDIC in the face of regional and international community’s being as the East African supportive referral Diagnostic Laboratory for avian flu and Newcastle and other Trans boundary animal diseases. This will have its own impact on international trade of animals and animal products from Ethiopia. Currently, NAHDIC has implemented ISO/IEC17025:2005 in three laboratories selecting 12 tests as its scope of accreditation since 2009. These tests have already accredited by accreditation body (SANAS). Has nine labs at BSL2 level, one at BSL3 level for the diagnosis of zoonotic diseases. The center is located in Sebeta 25kms from the capital, Addis Ababa. What types of molecular biology equipment do you have at your labs? 1. Agarose gel electrophoresis equipment 2. PCR thermocycler (Fast 7500 Applied biosystem , Biorad forr eal time PCR , Applied biosystem for conventional PCR and Lamp Temal cycler ( ESE – Melt) 3. PCR work station and safety cabinets class 2 4. Ice flack marker 5. Laminar flow hood and chemical hood 6. Wet ice maker 7. Liquid nitrogen 8. +4 0 C fridge or cold room 9. -20 0 C freezer 10. -80 0 C freezer 11. Pipettes for volumes in the range 1 ul to 1 ml 12. -Spectrophotometer (UV/VIS) 13. Rotary incubator (37 0 C) 14. Incubator cabinet (37 0 C) 15. Autoclave 16. pH meter 17. Hot plate stirrer 18. Magnetic stirrer 19.
