Standard DNA Polymerases
MyTaq™ is a new generation of very high
performance PCR polymerases, designed to deliver
outstanding results on all templates. MyTaq is
engineered to increase affinity for DNA, resulting
in significant improvements to yield, sensitivity and
speed. MyTaq is the perfect choice for all PCR
assays.
MangoTaq™ contains a colored reaction buffer to
make it an ideal choice for standard high-throughput
applications, as it allows the user to load directly on a
gel, without the need for separate gel-loading buffer.
BIOTAQ™ is a highly purified thermostable DNA
polymerase offering very high yield over a wide range
of PCR templates, and is suitable for routine assays.
DNA Polymerases for Specialized Applications
RANGER is a novel, antibody-mediated, hot-start
enzyme blend that offers 3.5x higher fidelity than
native Taq, designed to amplify templates from 10kb
or greater with extreme sensitivity.
BIO-X-ACT™ Short is specifically designed for
difficult/problematic PCR applications requiring both
high processivity and fidelity that would normally fail
with Taq DNA polymerase.
High-fidelity DNA Polymerases
ACCUZYME™ is a thermostable enzyme possessing
proofreading activities, offering high-fidelity, even
with demanding applications. ACCUZYME produces
blunt-ended amplicons up to 5kb.
VELOCITY is an ultra-fast thermostable enzyme
possessing proofreading exonuclease activity.
VELOCITY has high processivity, resulting in shorter
extension times, higher yield and the ability to amplify
long templates in a fraction of the time, even from low
template concentrations.
Hot-Start DNA Polymerases
MyTaq™ HS is a new generation of very high
performance, antibody-mediated hot-start PCR
products, designed to give outstanding results with
complex genomic DNA templates. MyTaq HS is
designed to increase affinity for DNA, so resulting
in significant improvements in yield, sensitivity and
speed.
IMMOLASE is a chemically-modified hot-start
containing a heat labile blocking group and requires
incubation at 95°C for 10 minutes, giving improved
specificity as compared to standard polymerases.
MyFi™ is a novel, antibody-mediated, hot-start
enzyme blend that offers higher fidelity and enhanced
specificity than native Taq. MyFi is ideally suited for
PCR amplification of difficult targets which require
both high processivity and higher fidelity.

MyTaq Extract-PCR Kit offers a quick and easy alternative for the extraction and amplification of DNA
from a variety of tissue types. Many other DNA extraction methods can be laborious and time consuming,
using phenol extraction, overnight incubations often combined with column purification. These methods
all contribute to the total time taken to prepare PCR-ready DNA and are rarely suited to medium or high-
throughput experiments. In addition, the longer protocols often require large amounts of starting material and
need to be further optimized for efficient extraction of DNA, particularly when samples are solid tissues such
as mouse tail or mouse ear.
When used with the same starting material, the MyTaq Extract-PCR Kit gave a better yield and was more
sensitive, compared to other suppliers of similar kits. Bioline’s MyTaq Extract-PCR Kit offers a convenient
alternative for the extraction of DNA for applications such as mouse genotyping and sequencing (Fig. 1).

Genomic DNA was extracted in a 100µl reaction and 1µl of the supernatant used for the PCR reactions. MyTaq One-Step RT-PCR has been formulated for highly reproducible first-strand cDNA synthesis and
subsequent PCR in a single tube. The combination of the latest advances in buffer chemistry together with
a reverse transcriptase and hot-start DNA polymerase system ensures that MyTaq One-Step RT-PCR Kit
produces fast, highly-specific and ultra-sensitive one-step RT-PCR, from either total RNA or mRNA using
gene-specific primers. The reverse transcriptase is thermally stable, so for RNA transcripts that form stable
secondary structures at low temperatures (including GC-rich sequences), higher temperatures can be used
(fig. 1), ensures high cDNA yields from all RNA.
MyTaq One-Step RT-PCR specially formulated enzyme blend ensures highly efficient and sensitive
transcription from as little as 3pg total RNA and is robust enough to work under different cycling conditions
(fig. 2). Even greater sensitivity and specificity can be achieved by use of gene-specific primers, maximizing
amplification of these targets while minimizing amplification of other sequences.
MyTaq One-Step RT-PCR Kit reduces reaction-to-reaction variability and helps prevent contamination
by minimizing hands-on steps. It is ideal for determining the presence or absence of RNA templates and
quantifying expression through qualitative, semi-quantitative or quantitative analysis of RNA transcription levels.
The one-step format is also perfect for the synthesis of double-stranded cDNA products for subsequent gene-
expression analysis.

Real-time PCR is one of the most powerful and sensitive gene analysis techniques available and has become
the standard in most laboratories. Its effectiveness, particularly at amplification and quantification of low levels of
nucleic acids, has driven the emergence of numerous applications, including cellular mRNA and quantification,
biomarker discovery and validation, microbial quantification, and detection of extremely low copy targets
for forensic investigations. The increase in the use of real-time PCR has increased the demand for a higher
throughput and faster assays, to reduce the overall protocol time. This has been achieved by improvements in
instruments, using faster enzymes (without sacrificing accuracy), and shortening or even combining PCR steps.
The new SensiFAST Kits have been designed to take up this challenge, with an antibody mediated hot-start and
very rapid enzyme-extension rates this enables fast cycling conditions, the SensiFAST formulations provide the
fastest cycling times whilst maintaining the performance, reliability and high reproducibility of conventional real-
time PCR reagents.
Selection of detection chemistryThe two types of chemistries that have been developed for real-time PCR are:
• Intercalating dye chemistry (including SYBR® Green I dye)
• Probe detection chemistry (including TaqMan®, Scorpions® and molecular beacon probes)
SYBR Green I dye chemistrySYBR Green I dye fluoresces when bound to double-stranded DNA, so the amount of fluorescence is directly
proportional the amount of double-stranded DNA. So if fluorescence is measured after each successive round
of amplification in a PCR, the increase in PCR product can be determined.
Probe detection chemistryAn oligonucleotide probe is constructed with a fluorescent reporter dye bound to the 5’ end and a quencher
on the 3’ end. The proximity of the quencher to the reporter dye greatly reduces the fluorescence emitted. If
the target sequence is present, the probe anneals and is cleaved by the 5’ nuclease activity of the Taq DNA
polymerase during extension, increasing the fluorescence emitted. Additional reporter dye molecules are cleaved
with each cycle, resulting in an increase in fluorescence intensity proportional to the amount of PCR product.

• Does not require expensive probes, so costs are reduced
• Multiple dye molecules can bind to a single amplified target sequence, increasing sensitivity
• The ability to perform melt curve analysis provides a quality control step to verify amplification of the desired amplicon
• Less specific than probes – only primers determine specificity
• Non-specific binding of SYBR dye to any double-stranded DNA may generate false positives
• Not possible to multiplex multiple gene targets

• Specific hybridization between probe and target, significantly reduces background and false positives
• Multiple targets can be detected in the same reaction, reducing cost and improving precision
• It is possible to distinguish between similar sequences with small differences e.g. SNPs
• A unique probe has to be synthesized for each target sequence

The new Bioline SensiFAST Genotyping Kit has been developed for highly reproducible genotyping of
sequence variants, including difficult single nucleotide polymorphisms (SNPs) loci and has been validated on
commonly used real-time instruments. The SensiFAST Genotyping Kit contains a combination of the latest
advances in buffer chemistry and enhancers, together with an antibody-mediated hot-start DNA polymerase
system. This ensures that the SensiFAST Genotyping Kit produces highly-specific and ultra-sensitive real-time
PCR with clear allelic discrimination, providing tight allele clustering and outstanding separation to ensure high
call rates, enabling reproducible and accurate results.
SensiFAST™ Genotyping KitExceed the limit
DNA samples from three Caucasian patients were genotyped using a custom TaqMan™ Drug Metabolism Genotyping Assay with the SensiFAST Genotyping Kit, using a set of thirty replicates for each sample. Cycling conditions: 95oC 3min, 35 cycles 95oC 10s, 60oC 30s. The results illustrate the SensiFAST Genotyping Kit gives very tight clusters between the replicates.

Bioline ISOLATE II Kits for nucleic acid isolation are perfect to start the analysis of your samples. The kits are designed
for fast and efficient isolation of DNA and RNA from a wide range of biological materials, including animal tissue,

Extended Storage LifedNTPs from Bioline have the distinct advantage of being presented as lithium salts, these are more resistant to repeated freeze/thaw cycles helping to keep them free from bacterial contamination during storage (lithium ions being bacteriostatic). Lithium salts are also more soluble than sodium salts in ethanol, so their removal by ethanol precipitation is more efficient, reducing the chances of salt artifacts.

Bioline offers a wide range of DNA Ladders premixed with Loading Buffer, enabling accurate sizing of DNA ranging between 25bp and 10,000bp and optional mass determination. 5x Sample Loading Buffer is supplied for a ready-to-use format, minimizing the time spent diluting and adding tracking dye to the DNA ladder. Simply transfer the Ladder from the vial to the gel.
The HyperLadder range has been created for accurate sizing of DNA fragments on agarose gels. EasyLadder is designed for shorter runs and can be run on standard or high-throughput agarose gels, the reduced number of bands being easier to distinguish in a short run.

HyperPAGE is a broad range protein marker prestained with two fluorescent dyes that resolves into 9 bands from 10kDa to 190kDa. HyperPAGE is lyophilized for convenient storage and only needs to be reconstituted prior to use. HyperPAGE II is a broad range protein marker prestained with five fluorescent dyes that resolves into 9 bands 10 to 250kDa. HyperPAGE II is supplied in a convenient ready-to-load format; eliminating the need to heat, reduce, dilute or add loading dyes.
Both are suitable for monitoring the efficiency of electrophoretic migration in SDS-PAGE and the transfer of proteins from gel to membrane in Western blotting, without the need for additional staining dye. Prestained protein markers are suitable for determining the approximate molecular weights of proteins. For accurate sizing, the use of unstained markers is recommended.

1. Terminologya. The “Company” shall mean either Bioline Ltd or Bioline GmbH in line with the entity
with whom any order is placed.b. The “Goods” are products supplied by the “Company”. c. The “Buyer” means the person or organisation to whom the Company has
contracted to supply Goods. d. Headings are given for ease of reference only and shall not affect the products
described. e. References to individuals shall be treated as applying to corporations and vice
versa. 2. Contract Conditions These conditions shall apply to every sale made or agreed to be made by
the Company. They shall govern each contract defined as an agreement to sell, either by acknowledgment of the Buyer’s order, or by the dispatch of the Goods requested. These Conditions cannot be varied or waived except with an expressly written approval of the Company. Any conditions submitted by the Buyer, in whatever form, are expressly waived and excluded except where written agreement is given by the Company in exceptional cases.
3. Contract Cancellation The Buyer may not cancel the contract without written agreement of the Company.
If this is given, the Buyer shall indemnify the Company against all losses, damage, claims or action arising as a result of such cancellation.
4. Changes to Contract After accepting an order, the Company may make modifications to the Goods
without informing the Buyer, it being understood that such modifications are improvements, not adversely affecting the performance of the Goods.
5. Prices The price of the Goods does not include the following items: Packaging, delivery, insurance and taxes, duties and other impositions, all of which
shall be borne by the Buyer. The Company shall not modify prices before delivery to the Buyer, except to
reflect additional costs resulting from any alteration or addition to the Buyer’s requirements.
6. Payment Termsa. Payment shall be made in full in the currency invoiced, within 30 days from the date
of the invoice.b. In the event of late payment the Company reserves the right to: Suspend all further deliveries until payment has been made, or cancel the contract
with respect to Goods still to be delivered. In either case this shall not prevent recourse to other rights or remedies available to
the Company.c. The Company may charge interest at 2 per cent per 28 days on any sum overdue,
calculated on a day-to-day basis until final payment. The Buyer shall not be entitled to withhold payment on the grounds of having other claims against the Company.
7. Returns Owing to the temperature requirements of our products, the quality of returned
goods cannot be guaranteed. consequently, we regret that we cannot accept returned goods in the event of a purchasing error.
8. Deliverya. Unless otherwise agreed in writing, all Goods shall be dispatched by methods
judged the most appropriate to ensure the safe delivery of the Goods. Whilst expedient delivery is considered most important, any times quoted for delivery shall be treated as estimates only and the Company shall not be liable for any loss or damage arising from delays in delivery.
b. Delivery is on an ex-factory basis. After delivery, the Goods shall be solely the Buyer’s risk in respect of loss or damage arising from any cause, unless the Buyer notifies the Company in writing: of any damage, discrepancy or shortage in the Goods delivered, within 8 days of delivery or of total loss or non-receipt of the Goods, within 10 days of the Company’s invoice.
In such cases, the Company’s sole obligation shall be limited to the replacement of the Goods in question and the Company shall not be under any other liability whatsoever.
c. Where delivery of any product requires an Export Licence or other authorisation before shipment, the Company shall not be responsible for delivery delays resulting from a delay in the granting, or refusal of, such authorization.
9. Property Title to the Goods shall remain vested in the Company and shall not pass to the
Buyer until the purchase price has been paid in full and received by the Company. However from delivery the Buyer shall keep the Goods fully insured until the time of payment. If the Goods are lost or destroyed before payment, the Buyer shall hold the proceeds of insurance to the order of the Company pending payment. The Buyer agrees not to dispose of or re-sell the Goods before full payment is made for them.
10. Limited Product Warranty The Company warrants that its products will conform to the standards stated in its
product specification sheets in effect at the time of shipment. The Company will
replace free of charge any product that does not conform to the specifications. This warranty limits the Company’s liability only to the replacement of the product.
This warranty is exclusive and the Company makes no other warranty, expressed or implied, including without limitation any implied warranty of marketability or fitness for a particular purpose.
The stated express warranties and the remedy provided for breach thereof, are in lieu of all other liability or obligations of the Company for any damages whatsoever, arising out of or in connection with the delivery, or the inability to use any of its products.
In no event shall the Company be legally liable in the basis of contract, negligence, strict liability in tort, or warranty of any kind for any indirect, special, incidental, consequential or exemplary damages (including but not limited to lost profits).
Without limiting the effect of the preceding sentence, the Company’s maximum liability, if any, shall not exceed the purchase price paid by the Buyer for the product.
11. Patent Disclaimer Unless explicitly stated, no license or immunity under any patent is either granted or implied by the sale of any of our products.
The Company does not warrant that the resale or use of its products delivered will not infringe the claims of any patents, trademarks or copyright covering use of the product itself, or its use in combination with any other products, or its use in the operation of any process. Furthermore, the purchaser assumes all risks of patent, trademark or copyright infringement associated with any such use, combination or operation.
12. Claims and Liabilitya. To the extent permitted by law, all conditions, warranties or obligations
expressed or implied by statute, common law or otherwise are excluded and replaced by the provisions of these conditions.
b. No claim under the Warranty (see Condition 9) will be met if, in the opinion of the Company:
The defect is not due solely to defective materials or manufacture. The Goods have been misused, treated with carelessness, contaminated,
involved in an accident or dealt with in a manner at variance with the Company’s directions.
c. The warranty of condition 9 is specifically limited to the Buyer and does not apply to any other person.
d. The Buyer acknowledges that the Goods will be used for laboratory and research purposes and undertakes not to make them available for human consumption, drugs or household use, directly or indirectly. The Buyer shall indemnify the Company permanently against claims by any third party, arising out of the use or sale of the Goods by the Buyer.
e. All descriptions, representations, illustrations and samples furnished or statements made orally by the Company are given for general information purposes only. Any possible claim can be based only on written specifications agreed at the time of contract.
13. Assignment The Buyer shall not transfer to any other person, any contract to which these
conditions apply or the benefit thereof.
14. Force Majeure Any contract to which these conditions apply is subject to cancellation by the
Company or to any such variation as it might find necessary by reason of strike, lock-out, trade dispute, weather conditions, hostilities, legislation, Acts of God or any cause whatsoever, beyond the control of the Company.
15. Governing Law a. For Goods purchased from Bioline Ltd, the contract shall be governed by and
construed in accordance with the laws of England and Wales and the parties hereby submit to the exclusive jurisdiction of the English courts.
b. For Goods purchased from Bioline GmbH, the contract shall be governed by and construed in accordance with the German civil code (BGB) and the parties hereby submit to the non-exclusive jurisdiction of the German courts.
16. Trademarksa. The Bioline logo is a trademark of Bioline Ltd. The following trademarks have
been registered by Bioline Limited and/or Bioline USA Inc: ACCUZYME, BIOTAQ, BIO-X-ACT, HyperLadder, IMMOLASE, MangoMix,
MangoTaq, MyTaq, TRIsure.b. The following are trademarks registered by other companies: SensiMix™ and SensiFAST™ (Bioline Reagents Ltd), SYBR® Green (Molecular
