Significant attention has been given to the beneficial effects of

prebiotics

• Since concept was first defined, it has been subject of intensive research

• Prebiotic research has continued at a rapid pace with >2000 research articles published

over the past 5 years>2000 articles

Prebiotics and probiotics are both beneficial for health

But what is the difference?

* https://isappscience.org/prebiotic-definition-updated-isapp/ ; https://isappscience.org/probiotics/

• What are prebiotics?

• Food/feed for beneficial microbes

that live in our colon.

• eaten by the ‘good’ gut microbes as

their “favorited dish”

• positive impact on the gut environment,

with an overall health benefit."

• What are probiotics?

• live microorganisms from other

sources than the human gut

• Swallowed and need to survive the

stomach and small intestine passage

• Need to provide strain-specific evidence

in humans about their health benefit

Imbalances in the gut microbiota have been linked to various

diseases….

*Literature search and interpretation by Beneo Insitute, based on 24 pertinent publications13

Imbalances in the gut microbiota are linked to:

• Asthma and allergy

• Infections

• Colic and functional gastrointestinal disorders in infants

• Inflammatory bowel disorders

• Autism spectrum disorders

• …. Low numbers of bifidobacteria are linked to

several disease risk associations*:

• Type 2 diabetes

• Inflammatory bowel disease (IBD)

• Ulcerative colitis

• Crohn’s disease

• Coeliac disease

• Obesity / metabolic disorders

A substrate that is selectively utilized by host

microorganisms conferring a health benefit

Changed:

• Prebiotics not limited to gastrointestinal tract

• Possibly includes non-carbohydrate substances and

diverse categories other than food

• Definition applies also for the prebiotic use in animals

Confirmed:

• Selectivity is key! ( criterion that

distinguish prebiotics from many other

substances)

• Selective effect must evoke a net health

benefit!

Strong body of scientific evidence for prebiotic chicory root fibres

The prebiotic effect of chicory root fibers

(inulin and oligofructose) is a

scientifically proven fact

>30 studies in adults

With as low as 5g chicory root fiber

per day the increase in Bifidobacteria

is demonstrated

>15 studies in infants and children

Belgium by Professor Jeroen Raes from the

Department of Microbiology and Immunology and

Professor Kristin Verbeke from the Translational

Research in Gastrointestinal Disorders Unit

• Randomised, placebo controlled, double-blind, cross-over design

• 44 participants with mild constipation consuming 12 g/d (3x4 g) Orafti®Inulin (compared to

maltodextrin) over 4 weeks

• Feacal samples were obtained before and after intervention and wash-out period for:

Prebiotic effect proven with state-of-the-art methodology

Vandeputte D, et al. Gut 2017;0:1–7. doi:10.1136/gutjnl-2016-31327120

The umbrella view on the colon’s

ecosystem demonstrates:

• Selective effect of chicory root fiber on the

human gut microbiota with newest

methodology

• Amongst almost 300 genera detected the

abundance of only 3 genera changed

significantly with inulin intake

• Bifidobacterium increased

• Anaerostipes increased

• Bilophila decreased

Improved well-being! Wellbeing parameters assessed with PAC-QoL(Patient assessment of Constipation Quality of Life)

Marquis et al. (2005) Scand J Gastroenterol 40(5):540–551. doi: 10.1080/00365520510012208 21

• PAC-QoL• Subscores used for „discomfort“ and „satisfaction“ comprising in total 9 questions

• Ratings from 0 (not at all, none of the time) to 4 (extremely, all of the time)

• Measuring the impact constipation has had on daily life during the past 2

weeks

• Discomfort: • „To what extent or intensity have you felt bloated to the point of bursting?“

• „How much of the time have you felt the need to have a bowel movement but not

been able to?“

• …

• Satisfaction: • „How much of the time have you had fewer bowel movements than you would

like?“

• „To what extent or intensity have you been satisfied with the regularity of your

bowel movements?“

• …

RESULTS of PAC-QoL:

Physical discomfort

score: No difference

Satisfaction: significant

improvement with chicory

root fibre

Improved digestive well-being by chicory root fiber

Vandeputte D, et al. Gut 2017;0:1–7. doi:10.1136/gutjnl-2016-313271 22

• Correlation between Bilophila abundance variation and changes in stool consistency was

found

&

• Correlation between the reduced numbers of Bilophila in the gut and improved

constipation-related wellbeing parameters ( Quality of Life questionnair) was observed

Author’s conclusion:

“the observed association between Bilophila decreases on inulin consumption

and quality-of-life increase urges the need for further research on

the role of this taxon in host well-being.”

Orafti®Synergy1 prevents a leaky gut!

Schroeder et al. Cell Host & Microbe 23, 27–40, January 10, 2018

46

Background:

The inner colonic mucus layer separates

gut bacteria from the host and thus is an

important protective physical barrier

between the outside world including trillions

of gut bacteria and the inside of a human

being.

Diet affects microbiota composition and the

microbiota influences the mucosa.

Key results (mice study):

• A Western-Style diet (WSD) leads to

• increased mucus penetrability and

• decreased mucus thickness, and

• a reduced growth rate of the inner mucus

layer, and

• is accompanied by distinct alternations in the

microbiota composition

• Orafti®Synergy1 supplementation

prevents the Western-Style-diet-induced

increase in mucus penetrability!

Health benefits of lowering blood glucose levels

International Scientific Consensus:

• Reducing the blood glucose response of

a meal is a beneficial physiological effect

• Carbohydrate-based diets with low effect

on blood glucose levels

• Improves blood glucose control in people with

type 2 and type 1 diabetes

• Reduces the risk of type 2 diabetes

• Reduces the risk of coronary heart disease

• Likely reduces total body fat mass and supports

body weight management

2017 review continues to confirm

The low blood sugar response of chicory root fibres

Lightowler et al. (2017) Eur J Nutr. https://www.ncbi.nlm.nih.gov/m/pubmed/28255654 51

The effect of sugar reduction with chicory root

fibres on blood sugar and insulin response

has been investigated in:

• 9 human trials (including normal & overweight

individuals) showing that the more sugar you

replace with chicory fibres the lower the

blood sugar response

• Already 20% sugar replacement shows a

significant effect

• Demonstrated in different product applications

All studies show a reduced blood glucose response with chicory fibres
