1. Research and Development (Core research activities undertaken)

Animal Health Program: Under PPR project-ALP, a local isolate of PPR virus (PAK-Fjg-07/NARC4)

from AHP repository is being attenuated for the development of PPR vaccine by serial passages on Vero cells. During month under report 49 serial passages have been completed.

Seven lyophilized isolates of PPR virus stored in animal health laboratory were reconstituted and 6/7 isolates were found positive by RT-PCR using newly synthesized primers of M-gene.

Quantitative real time polymerase chain reaction (qRT-PCR) was optimized and a standard curve was made using PPR vaccine as a positive control.

Eleven PPR virus infected tissue samples were inoculated onto Vero cells for virus recovery and the flasks were subjected to 2nd blind passage by freeze/thaw method. Six samples showed PPRV characteristic cytopathic effects.

Complete N gene consisting of 1578 nucleotides was amplified employing RT-PCR from tissue samples collected during outbreaks at Barnala, AJK, Bhakhar and Faisalabad using newly synthesized N-gene primers of PPR virus.

Four tissue samples (Spleen, Liver, Lymph node and Lungs) collected during a suspected outbreak of PPR at Layyah were found positive by RT-PCR.

Repository of different local isolates of PPR and FMD viruses are being maintained by the program. Besides, regular maintenance and sub-culturing of Vero and LFBK cell line were performed.

RNA was quantified from various reference FMD strains (A, O, Asia 1) using nano-drop and subjected to quantification using real time PCR.

A trial to determine appropriate age for PPR vaccine was initiated at BLPRI, Kherimurat in collaboration with FAO.

Animal Nutrition Program:

ALP project “Generation of reference values on amino acid availability in local feedstuffs for poultry and capacity building at NARC/PIASA” is in progress. Analyses of feed and animal samples are under progress. Second trial using 100 broiler chicks has started in which feedstuffs and droppings will be analyzed.

An experiment entitled “Effect of varying protein sources of concentrate on growth and nutrient utilization in Sahiwal cattle calves fed maize silage as basal diet” has been started using 12 calves received from LRS. These calves have been divided into 3 groups of 4 calves each. Three concentrate diets (iso-nitroginous) have been prepared having different protein source as under:

Diet A having Maize Gluten meal 30% while diet B having Cotton Seed Cake and diet C having Rape Seed Cake as protein source. These concentrate are being offered to calves @ 1.5% of body weight on dry matter basis and maize silage being fed ad-libitum basis. Data regarding daily feed intake and refusal, and fortnightly body weights are being recorded.
