It is espe-ciallyadvantageous forslicing small or ir-regular shapedspecimens as onemight obtain frombiopsies. The Mcll-wainTissue Chopper causes much less disturbance of the cellstructure than homogenizers or blenders and has been usedon various tissues of the central nervous system as well asfor liver and kidney.Slices up to 1mm thickness and cubes or prisms up to 1mmcross section can be prepared with the chopper in 30 sec-onds. Stepless variation of slice thickness from zero to themaximum is done by means of an indexing clutch in conjunc-tion with a micrometer head calibrated in microns. The speci-men is placed on the circular, white disc on the stainless steeltable and is transversed automatically from left to right at anadjustable speed. At the same time, the blade is raised anddropped at speeds varying from zero to over 20 strokes perminute. The process may be repeated with the table turned45° if prisms are desired.Turn the table 90° on the second cutfor cubes.Electrical: 110 and 220VAC

Pattern Design

The MRS-4 is fabricated using the high accuracy direct write elec-tron beam manufacturing equipment. The pattern is anti-reflec-tive chromium on a glass standard. Imaging contrast in bothsecondary and back scattered mode is very high. The pattern iscoated with a proprietary conductive material which allows forSEM imaging at any accelerating voltage.

Geometric Design

The MRS-4 has groups of nested squares with pitches of 1/2µm,1µm, 2µm, 50µm and 500µm. The largest pattern has an overalldimension of 8mm square with lines and spaces of 250µm. Thiscan be used to check magnifications between 10 - 100x. The50µm pitch patterns are useful from 100 - 1000x magnifications. The 2, 1 and 1⁄2µm pitch patterns allow calibrations up to200,000x. Incorporated into the standard are X & Y rulers with6mm length with 1µm increments.

DimensionsThe overall size is ap-prox. 9mm x 9mm x2.3mm thick. TheMRS-4 can be pro-vided in an aluminumretainer with a size of25mm diameter and3mm thickness to pro-tect the standard.

 Citifluor Antifadent Solutions

In the technique of fluorescence microscopy, the fluorescence isstimulated by high intensity UV and /or visible light. Absorption ofthe light populates an excited state of the dye (usually a singlestate) and this leads to fluorescence. However the excited state ofthe dye may undergo chemical reactions which lead to its de-struction as evidenced by the fading or bleaching of the fluores-cence and consequent loss of the image or in the case of assays,a change in signal intensity during measurement. To overcomethese problems Citifluor developed antifadent (antibleaching) so-lutions for use as mounting media when specimens are examinedby fluorescent microscopy or other detection system and as addi-tives for assays. continued on next page

Unconjugated Gold and Silver Sols - Gold/Silver Colloids (Sols)Nanometer-sized gold and silver particles of uniform shape and size are invaluable tools in nanotechnology (assembled arrays), lightscattering (elastic - plasmon resonance; inelastic - Raman effect) and for single molecule detection. Gold nanoparticles can be attachedto proteins, alkanethiols and DNA by various methods. Silver nanoparticles are evolving similar functional characteristics to their goldcounterparts.Ted Pella, Inc. is the leader in offering a wide range of both silver and gold nanoparticles for these applications. Talk to our applicationsspecialist to find out about the new developments/applications. We are constantly developing new gold and silver colloids for newnanotechnology applications.

The nanometer sized gold colloids can be characterized as follows:

• Monodispersed gold nanoparticles supplied in water, having trace amounts of citrate, tannic acid and potassium carbonate

• Citrate stabilized with a net negative surface charge (Langmuir 2005, 21:9303-9307)

• 5-20nm gold colloids are supplied at a pH 6.0, larger particlesizes at pH 9.0

• Available in a size range from 2 to 250nm

• The particles are invaluable tools for light scattering, eitherRaman or plasmon resonance (Analytical Chemistry 1999,71:4903-4908; Analytical BioChemistry 1998, 262:137-156)

Unconjugated Gold and Silver Colloids or SolsThe 2nmn colloid is too small to scatter light and the solution isclear. The remaining sizes scatter light to different degrees and thesolution color changes with increasing particle size.

The Sequenza™ Slide Rack, holding 10 glass slides and Cover-plates™, is a system designed for the immunolabeling of sectionson glass slides. While originally designed for bench techniques, this system is uniquely suited for microwave-assisted applications. The Slide Rack holds glass slides covered with a Coverplate™. Thiscombination forms a capillary gap between the slide and theplate. Reagent volumes, especially antibody, are greatly reducedusing this versatile system. A reagent hopper forms the upper-most part of the plastic Coverplate™ allowing for easy dispensingof reagent into the capillary gap. Using a PELCO® MicrowaveProcessor with a PELCO ColdSpot®, rapid immunostaining of sec-tions on glass slides is reliable and easy.

Applications:

• Live Cell Microscopy• Multi-Photon Laser Scanning Microscopy (MPLSM)• High Level Laser Fluorescence• Fluorescence Lifetime Imaging Microscopy (FLIM)• Fluorescence Resonance Energy Transfer Microscopy (FRET)• Fluorescence Redistribution After Photobleaching Microscopy

(FRAP)• Epifluorescence Microscopy• Fluorescence Imaging• Immunofluorescence Microscopy• Confocal Microscopy• Confocal Laser Scanning Microscopy (CLSM)• Phase Contrast Microscopy• Polarized Light Microscopy• Differential Interference Contrast• High Resolution Image Analysis• Time Lapsed Photography• Video-Enhanced Microscopy• Infrared imaging• Calcium imaging• Colocalization studies• Green Fluorescent Protein (GFP)• Micromanipulation• Microinjection

Advantages are:

1. Much larger effective range, using an objective lens with ashort working distance

2. No birefringence effects because of thin (0.17mm) glass bot-tom

3. Low rim/side wall of the dish and a larger effective workingdistance make micromanipulation and microinjection possible.

4. Higher effective N.A. make brighter images (in e.g.: fluores-cence)

5. Higher effective N.A. make higher resolutions in image analy-sis

6. Microscope light passes through the bottom glass in a rectan-gular way, glass rests on stage

7. Direct heating from the microscope-stage, because the glass is(flush) in direct contact with stage

8. In IVF, no more oocyte-/embryo transfer is needed from oneplastic dish to another, unless different media are being used, saving a lot of time

9. Easy "perfusion" in the dish for continuous in and outflow ofsolutions, using "perfusion chamber insert"

 Immunostain Moisture Chamber

This is a low-cost, high precision moisture chamber for ten slides.Every moisture chamber is divided into ten individual compart-ments with approximately 12.7mm (1⁄2") empty space betweenthe compartments. When the chamber lid is closed, eight barrierdividers are placed into the empty spaces between the micro-scope slide compartments completely isolating all compartments. This is a very desirable feature when doing immunostaining.

The microscope slides are placed on four pedestal posts and fourcorner posts each 11.5mm (0.460") high, thus raising the surfaceof the slides approximately 12.7mm (1⁄2") off the floor to keep theslides away from the water below and making the slides easily re-trievable (either by hand or by forcep).

The chamber is fabricated out of heavy-duty polystyrene with anair-tight design to keep moisture in. The chamber is designed tobe stackable in order to save counter and/or refrigerator space

Universal slide rack and container for autoradiography gel stain-ing and staining techniques. Molded of black polyoxymethylene-poly-acetal plastic which has great advantages over glass in allcytological and histological laboratories.

Staining Dish

The Staining dish has a light tight lid, essential to easily evapo-rated baths and a compartment for a drying agent. The dish ac-commodates one slide rack, Product No. 21078, not included. The perforated slide partition which supports the drying agenthas index numbers and a writing surface. A removable clip isplaced on the top edge of the dish, permitting the stain solutionto drain back into the reservoir from the tilted rack. The actual ca-pacity of the small dish is 250ml but only 150ml is needed, whichprovides a saving on staining liquids, developing fluids and inter-mediates. Made from black polyoxymethylene-poly-acetal plasticthe dish is easy to clean and especially suited to store enzymes. The cover will not jam in freezer storage. Maximum use tempera-ture is 85° C, (185° F). Dimensions: 94 x 81 x 49mm high (3.7" x3.2" x 1.9").
