Hydro Air Research Italia

Since 1979 HAR has gained extensive expertise inprocess and waste water applications, becoming aleader in membrane separation technology.Over 10% of HAR annual revenues are devoted to thecompany’s research and development efforts. The resultis an on-going technological upgrading of its productsand applications, including separation technologies suchas evaporation/crystallization, electrodialysis,chromatography and pervaporation.

Industrial process applications include biotechnologyand pharmaceutical applications, where HAR developedits own know-how, as well as applications in food anddairy, fine chemicals, petrochemical and chemical field.

According to the increasingly stringent environmentallegislation, HAR designs and supplies complete plants“Zero Discharge Water Recycling”, an advanced processto treat effluents obtaining water suitable to reuse or todischarge according to the law restrictions. The goal isreached through the combination of differenttechnologies, from bioHAR® Membrane Bioreactorthrough membrane processes, pervaporation andevaporation.

HAR has developed and realized patented systems forLandfill Leachate Treatment, plants with an economicalprocess, effective and reliable.

HAR pursues ISO 9001 QualityPolicy, to enable the maximumsatisfaction of its customersby responding appropriately totheir needs and requirements.

HAR approach to separationproblems

The beginning of all HAR systems design is theevaluation of the customer’s objectives andrequirement. Test facilities and pilot plant are availablefor feasibility study and bench or semi-industrial trials.

Preliminary tests are carried out in HAR laboratories onsmall liquid samples providing separation performanceand data useful for industrial plant scale-up as well ascost projections or economic models to determinate full-scale operation and required capital costs.Pilot plant verification at customer’s site isrecommended to generate data under actual fieldconditions.

Membrane

Membrane technology can be used in a wide range ofseparation processes.HAR expertise covers all the range of membraneprocesses, from Microfiltration and Ultrafiltrationthrough Nanofiltration and Reverse Osmosis toElectrodialysis.Depending on the purpose of the separation, differenttype of membranes can be used, characterized bydifferent material of construction, configuration andselectivity.Organic and inorganic membranes, spiral wound andhollow fiber, tubular and plate type can be selecteddepending on the application and on processrequirements.

Chromatography

Considering the increasing demand on products puritylevels, specially in biotechnology, food andpharmaceutical field, chromatographic separation is themost powerful of separation technologies for purificationprocesses, mainly for impurities that are difficult toseparate by conventional separation methods.HAR activity includes process feasibility through pilottesting, design and manufacturing of chromatographiccolumns.

Technologies

Ion Exchange

Many kinds of substances can be purified by ionexchange treatment with specific resins. The separationgoal is pursued through different techniques, based oncomplex ion formation, ion exclusion, affinity difference,selectivity difference or ion retardation.

Evaporation / Crystallization

HAR continuous evaporation systems use a wide rangeof technologies: single or multieffects, forcedcirculation, falling film, thin film with thermalcompression or mechanical vapour compression. Whendesigned as crystallizers for chemical, pharmaceuticalsor food industry the engineering of the equipmentguarantees product quality and long operating cycles.

Pervaporation

A low pressure, low temperature membrane process fororganic solvent drying and concentration. An optimalprocess to break azeotropes and solve bottleneckingproblems.

Microfiltration is the most open cross-flow filtrationdevice. Typical application of this low pressure processis the clarification of chemical, biological,pharmaceutical or food solutions, for the removal ofhigh molecular weight compounds such as bacteria,fats, yeasts and fungi.

Membrane Technology

Ultrafiltration is a low pressure process for selectivefiltration of molecules of specified size and weight,typically in the range from 1000 MW to 1.000.000 MW.Typical applications of UF membrane are concentrationand recovery of high molecular weight compoundsseparation/removal of contaminants, clarification,removal of colloids and suspended solids.

Nanofiltration is a pressure driven process similar toRO applied in the area between RO and UF. Monovalentions and low molecular weight organics are passingthrough the membrane, while bigger compounds arerejected. MW cut off of a NF membrane is in the range150-500 and typical application is in pharmaceuticalpreparations, removal of colour, of hardness from water.

Reverse Osmosis is a high pressure separationprocess to remove low molecular solutes (salts, smallorganics molecules) from a solvent (water).The separation occurs with an imposed pressure higherthan the osmotic pressure, forcing the solvent to passthrough the membrane while the solute is rejected,monovalent ions included.

The large variety of membranes and modules in allcommercial configurations (tubular, spiral wound,hollow fiber, plate and frame) as well as manufacturingmaterials (polymeric and inorganic) provides numerousalternatives allowing to solve most separation problemswith custom made systems.

HAR technical specialists work with the end-user inorder to satisfy the widest spectrum of requirements interms of application, volume and rules, tailoring thesystem on the customer’s needs.

The extensive design, engineering, manufacturing andquality assurance capabilities as well as the experiencein thousands of industrial systems combine to producethe most competitive cost efficient membrane systems.

Depending on the type of operation (i.e. batch orcontinuous) and the extent of interface with existingequipment required, the operating system controls areranging from a simple manual or programmable logiccontrol (PLC) to a distributed control system.

System design and componentsselection criteria are evaluatedfunction to each specific process, ensuring that safetystandards are incorporated such as explosion proofing,ATEX compliance, analytical process detection, CIPand/or SIP, full automatic sequence, full or partialanimation of the process, data logging to PC, sanitaryrequirements, GMP criteria and FDA approval.

Special membranes can be used to concentrate anddesalt small organic solutes in a wide spectrum oforganic solvents, for pharmaceutical/chemicalprocesses or waste treatment applications.

Developing a chromatographic or adsorptive purificationtechnology for a target product involves three steps:

� selection of media and scouting for suitable protocol� optimizing media and column performance for

defined separation objectives� scale-up and operation of the production scale

system with SOPs and validation procedures

In a collaborative effort with international institutesand adsorbent manufacturer, HAR built an expertise indesigning processes for different industrialchromatographic separations for molecules fromantibiotics, natural active ingredients of plant origin,synthetic peptides, synthetic APIs, to macromolecules likeenzymes, antibodies and plasmid DNA, DNA and RNA.

Resins offered by HAR are rich in range of interactionsas well as range of particle and pore sizes; they offeradvantages like rigidity and stability to autoclaving and

harsh CIP (cleaning-in-place) and SIP (sanitization-in-place) procedures.

Considerable work has lead HAR to design columnhardware for both packed bed and expanded bed formatof adsorption chromatography. Packed bed andexpanded bed purification technologies have beendeveloped for molecules from penicillin G, rifamycin,vitamin B12, and ß-carotene to proteins like lactoferrin,lactoperoxidase, lysozyme, insulin etc.

HAR offers laboratory scale to plant scale columnchromatography setups complete with column, pumps,piping system and measurement & controls.

HAR is also proposing of a new variant of expanded bedadsorption that attempts to overcome all disadvantagesof packed bed and expanded bed operations.Fluidized Moving Bed (FMB) is a combination ofexpanded beds of adsorbent mounted above each otherin the form a single composite column. The distinctsections individually handle different operations likeadsorption, washing, elution and regeneration in asequential manner with the adsorbent always movingcountercurrent to the different mobile phases indifferent sections. The concept of FMB is useful inlarge scale separations like in whey or soya proteinrecoveries, recovery of carotenoids from palm oil and inantibiotic purifications, lowering adsorbent inventorieswhile providing better solid-liquid mass transfer ratesand high productivities on adsorbents used..

Pervaporation is a low pressure, low temperaturemembrane process for organic solvent drying andconcentration. Pervaporation or vapour permeation (depending if theseparation is performed in liquid phase or gas phase) isan optimal process to break azeotropes and solvebottlenecking problems at competitive costs.It offers the possibility to separate miscible liquids ofsimilar molar mass and it is an alternative method todistillation, overcoming the limitations of standarddistillation in presence of azeotropes. Pervaporationtechnology can offer considerable savings both in termsof costs and performances.

� The solvent can be concentrated down from anywater level to very low water content

continuous high flux operation�Water produced is highly pure and can be

reprocessed� It is a low energy process

Pervaporation

Batch pervaporation is a simple system with greatflexibility, however a buffer tank is required for batchoperation. Continuous pervaporation operates best withlow impurities in the feed and is best for largecapacities. Vapor phase permeation is preferred fordirect feeds from distillation columns or for streamswith dissolved solids

The membranes used inpervaporation processes areclassified according to thenature of the separationbeing performed.Hydrophilic membranes areused to remove water fromorganic solutions.Organophilic membranes areused to recover organicsfrom solutions.

organics�Recovery of valuable organic compounds from

process streams�Dehydration of organic solvent-water solutions�Harvesting of organic substances from fermented

broth

Other products separated or purified by pervaporationinclude alcohols (methanol, ethanol, propanol, buthanol,benzyl alcohol), ketones (acetone, MIBK), aromatics(benzene, toluene, phenol), esters (methyl acetate, ethylacetate, buthyl acetate), amines (triethylamine, pyridine,aniline), aliphatics (chlorinated hydrocarbons) and ethers(THF, dioxane).

Pharma / Biotech

HYDRO AIR RESEARCH membrane systems areoperating successfully in downstream processing forantibiotics and biological active principles extractionfrom fermentation broths including such products asCephalosporin, Erythromycin, Penicillin, Clavulanic Acid,Tetracycline and others.The systems, when compared with the conventionalrotary precoat filters often used in this service, aresuperior in terms of product yield, filtrate quality,operating costs and waste problems.
