The suite is equipped with a high pressure homogenizer (Aves- tin) which will allow the disruption of dissimilar samples with a minimum maintenance and a low cost. The equipment has a built-in compressed air supply (100 psi) which will allow to pro- cess different samples: liposomes (70 psi), E. coli (70 psi) and Pichia (100 psi). The installed cooler of the equipment, con- nected to tap water, keeps the temperature low during sample disruptions. The suite is also equipped with Dyno-Mill cell dis- ruptor to process yeast samples. The equipment is fitted with a circulating bath which allows the disintegration of samples at low temperatures. For small volume of samples, BeatBeater is also available. The suite is equipped with one large-scale fermentation system (70-L stainless steel bioreactor). A steam generator is installed for one-step in-suite sterilization. It has an Applikon bio-controller (Model ADI 1010) and Pilot system (ADI 1075) with three pneumatic pumps (acid, base, and antifoam), stirrer controller, and rotameters, which provide user-friendly setup and automated control/recording of temperature, pH, dissolved oxygen (dO2) during the fermentation process. One computer is installed with BioXpert software for large-scale fermentation. The suite is equipped with six small-scale fermentation systems (three with 3-L bioreactors and three with 7-L bioreactors) available for Escherichia, Streptomyces, Rhodobacter, Salmonella, Bacillus and others. Each fermentation system has an Applikon bio-controller (ADI 1030), stirrer controller (ADI 1032), three rotameters (air, O2, N2), and three pneumatic pumps (acid, base, antifoam/label), which allow automated control/ recording of temperature, pH, and dissolved oxygen (dO2) during the fermentation process. One computer is installed with BioXpert software for bacterial fermentation. Bacterial Fermentation System (3-L or 7-L) The suite is equipped with a CEPA high speed centrifuge to harvest large volume of cells and media. It allows the collection of 50 liters of media within 45 minutes. The sample port of the 70-L fermenter can be directly connected to the high speed centrifuge to allow the harvest of samples after the fermentation process. The suite also has one separate bioreactor (3-L) with marine type impeller and sparger suitable for animal cell culture. The suite is equipped with one DASGIP OD4 module and three DASGIP Optical Density Probes, 12 mm diameter, 225 mm inser- tion length w/ 5 mm light path. From Jan 2011, the suite started this fermentation optimization package to help finding optimal condition to maximize produc-tion in fermentation. Design of Experiment (DOE) is a widely used method in process optimization in chemical engineering and industry. Optimization package provided from our suite is for 3 factors (such as temperature, pH, or dissolved oxygen) with 20 runs of small scale (700 ml) fermentations. It requires 7 days of experiments (3 fermentations per day) to achieve data. This service provides data analysis using DOE software to find a fit model for each responses and optimal condition for the re-quirement (goals). Fermentation Optimization Package (NEW)

Fermentation Annual Workshops Pichia workshop provides hand-on experiments for handling bioreactors and pichia culture. It provides detailed theory as well as experimental know-how by suite manager. Pichia pasto-ris strains are used to produce human Cathepsin K (with coop-eration from Dr. Dieter Bromme lab) for easy assay for produc-tion estimation. DOE workshop provides basic statistical introduction about Design of Experiment software from Stat-East Inc. Company and basic hands-on experience for handling bioreactor as well. Design experiment for optimization, conducting fermentation, and data analysis will be discussed during the workshop. These workshops are not credited courses nor certification but for training purposes. Each workshop will be announced via LSI Email list.
