Supplemental Figure 3. The sly1 mutants show increased sensitivity to GA biosynthesis inhibitor PAC in seed germination. Seeds of WT Ler (30 mo-old), ga1-3 biosynthesis mutant (27 mo-old), sly1-10 (29 mo-old, #3) and after-ripened (A) sly1-2 (28 mo-old, #21) were incubated in the presence or absence of 100 µM PAC for 3 d during seed imbibition at 4oC. Then seeds were transferred to plates containing 0 µM GA3 and increasing concentrations of GA and incubated under light at 22oC. Germination rates are given as percent radicle emergence after 7 d incubation. In the absence of PAC, the germination rate was 100% for WT and sly1-10, 75.4% for after-ripened sly1-2 and 0% for ga1-3 in the absence of GA. Age of the seed is given below the genotype (month). Error bars show S.E. (n=3). % Germination was calculated based on cotyledon emergence.
