Haemocytometer

• The hemocytometer is a device designed for the counting of blood cells. It is now also used to count other types of cells and other body fluids and semen analysis.

• hemocytometer consisting of 2 pipettes one for the red cells and other one for the white cells and a thick glass slide is on average 25 theater box.

• Each square is divided, including 16 square smaller.

The objective of the experiment:

• Set the number of red blood cells in a blood sample taken from a human using Haemocytometer.

 The principle of the experiment

• Rely on the experience of blood-thinning solution reduce red blood cells (a solution of isotonic sodium chloride, 0.9%).

1-Swab the hemocytometer and cover slipe with 70% EtOH and add cover on slide.

2-Swab towards the side of the tip of a little-used finger with 70% EtOH.and draw blood up to the 0.5 mark.

3-Continuing to hold the pipette as horizontal as possible, pull solution diluent up to the 101 mark.

4-Seal the tip with your finger and shake well to mix

5-Get rid of a few drops of diluted blood then put a drop of blood at the edge of the glass cover subject on the slide. Leave the slide for two minutes to precipitate the red blood cells and then placed under a microscope to examine.

• count and record the RBCs in each of five fields (each with 16 smallest squares). Include in the count all cells touching left and bottom sides, ignore cells touching top and right sides.

• Wash out the pipette thoroughly with soap and water, rinse well, finish with distilled H2O rinse, replace in case.
