xMAP Technology Overview
Bead-based Multiplexing of Up to 500 Bioassays per WellOpen-architecture xMAP Technology enables multiplexing of biological tests (assays), reducing time, labor, and costs over traditional methods. Systems using xMAP Technology perform discrete assays on the surface of color-coded beads known as microspheres, which are then read in a compact analyzer. Using multiple lasers or LED and high-speed digital-signal processors, the analyzer reads multiplex assay results by reporting the reactions occurring on each individual microsphere particle.
xMAP Technology Advantages and Benefits• Speed/High-Throughput - Because each microsphere serves as an individual
test, a large number of different assays can be performed and analyzed simultaneously
• Versatility - A single xMAP Technology-based system can perform assays using several different methods and chemistries, including nucleic acid hybridization, antigen-antibody binding, enzyme reactivity, receptor-ligand binding and other protein interactions

How xMAP Technology WorksFirst, Luminex uses proprietary techniques to internally color code microspheres with various fluorescent dyes. Through precise concentrations of these dyes, up to 500 distinctly colored bead sets can be created, each of which can be coupled to a target-specific molecule. These target-specific molecules may be antigens, antibodies, oligonucleotides, enzyme substrates, or receptors. After an analyte from a test sample is captured by this target-specific molecule, a reporter molecule, labeled with a different fluorescent dye, is introduced to complete the reaction on the surface of each microsphere. Next, the microspheres internal dyes are excited by the laser or LED, identifying the microsphere set. A second laser or LED excites the fluorescent dye on the reporter molecule. Using these signals, highspeed digital-signal processors identify each individual microsphere and quantify the amount of target molecule bound to its surface.

Luminex internally color-codes microspheres with precise concentrations of multiple fluorescent dyes, yielding up to 500 distinctly colored bead sets.
The microspheres can be coupled with reagents specific to a particular bioassay, such as antigens, antibodies, oligonucleotides, enzyme substrates, or receptors.
The microspheres pass through a red laser or LED, which excites the internal dyes to distinguish the microsphere set. Then a green laser or LED excites the fluorescent reporter dye to determine the result of the assay.

Data Acquisition and Analysis for Luminex Instrumentation xPONENT is a modular and flexible softwarepackage for controlling the MAGPIX,Luminex 100/200, and FLEXMAP 3D instruments. Designed for ease-of-use, the software allows the novice and advanced user alike, to set up and run an assay in a minimal amount of time.
The software’s graphical user interface follows the typical assay workflow but can be customized to meet the needs of a specific laboratory. Navigation wizards and automated routine operations such as startup, shutdown, calibration and performance verification, further enhance system usability and reduced hands-on-time.
xPONENT Software Offers:• Automated daily operations – Use of calibration and performance verification
kits to automate routine system maintenance as well as Startup, Shutdown, Calibration and Performance verification operations.
• Normalization bead support for internal standard – For all analysis types, users can select one bead for the intra-well normalization bead.
• Allelic ratios and allele call graphing function – Allele call analysis with new bar and line charts are now available for the analysis of allelic ratio data.
• Notification, alert options and remote monitoring – Users can log on remotely to monitor the system status. The software can also be configured to send out email notifications when an assay has been completed or if the system has encountered a problem.
• Low bead count detection – The software can be configured to alert the user if a user-defined number of successive wells do not reach the minimum bead count.
• On-the-fly regression analysis capability – Enables the user to quickly and easily reanalyze batch data with alternate curve fit algorithms.
• Luminex or third-party data analysis programs – The user can select from a user-defined set of analysis programs when setting up a protocol and the software will automatically launch the program once the data acquisition has been completed.

xPONENT Software is available with three levels of accessibility:
Basic The basic xPONENT software package allows the user to acquire samples, perform analysis, run system maintenance routines and remote web monitoring. This package includes the ability to calculate allelic ratios to analyze and/or normalize genotyping data.Three seat licenses are included with every copy of xPONENT software, but additional seat licenses are available.*
Secure*The secure xPONENT package includes all the basic functionality, and allows an administrator to set up user access levels.
21 CFR Part 11*For laboratories working under the FDA’s 21 CFR part 11 regulations, xPONENT offers multilevel user management, full audit trail, electronic records and electronic signatures.
xPONENT Software offers two additional licensed modules:
Automation Connectivity*The automation module, along with a device driver, enables communication between the reader and a front end automation platform. Luminex device drivers are offered by many automation providers. For more information on available drivers please contact Luminex Technical Support.*
LIS Connectivity*The LIS connectivity module allows communication with an external Laboratory Information System (LIS) by allowing the export of patient result data and the import of patient sample information in ASTM format. Microsphere Shelf-Life: The shelf-life of a xMAP microspheres is the length of time after manufacturing during which a microsphere should be used for coupling to a capture molecule. After coupling, microsphere stability will vary depending on a variety of customer-defined factors (molecule, buffer and storage temperature, etc.). Luminex recommends that customers verify the stability of coupled microspheres in their own lab under their own conditions.

Similar to MicroPlex Microspheres, MagPlex Microspheres are carboxylated polystyrene micro-particles, or “beads,” that have been dyed into spectrally distinct sets, or “regions”, allowing them to be individually identified by an xMAP Instrument. These uniquely coded beads provide a user an addressable substrate on which to perform multiple bio-analytical reactions in a single well.
All microspheres are general use components for further manufacture of xMAP bioassays. MagPlex Microspheres are available at concentrations of 2.5 million and 12.5 million beads/mL and can be ordered in 1mL and 4mL vial sizes. MagPlex Microspheres are also offered in a variety of custom volumes. Please see the MagPlex Custom Volume Ordering Guidelines online or contact Luminex to inquire about custom volume options.

MagPlex-TAG Microspheres are oligonucleotide-coupled polystyrene micro-particles, or “beads,” that have been dyed into spectrally distinct sets, or “regions”; allowing them to be individually identified by an xMAP Instrument.
Each one of these uniquely color-coded beads has a unique 24 base DNA sequence, called an “anti-TAG,” covalently coupled to its surface. These beads enable the user to quickly and easily design custom bead arrays, simply by adding a complementary “TAG” sequence to their primers or probes of interest and hybridizing those primers or probes to the anti-TAG sequence on the addressable microsphere.

xMAP Instrument Reagents
MAGPIX Reagents
The MAGPIX instrument requires drive fluid to serve as the delivery medium to transport the beads to the instrument’s optics. Due to the compact nature of the MAGPIX system, drive fluid is available in small pre-filled disposable containers which are stored within the instrument. Each drive fluid container provides enough fluid to read approximately ten plates of samples, and they are sold in packages of four. MAGPIX uses a calibration kit and a verification kit, each providing 25 uses.
Luminex 100/200 Reagents
The Luminex 100 and Luminex 200 instruments require sheath fluid to serve as the delivery medium to transport the beads to the instrument’s optics. Both instruments have the option of a 20L disposable external sheath fluid container or a compact 20X sheath fluid concentrate pack; which can be reconstituted, with water, to a working volume of 20L when needed. The Luminex 100/200 instruments, operating on xPONENT software, are calibrated using a calibration and a verification kit, each providing 25 uses. Luminex systems running on the older IS 2.3 software require the purchase of individual vials of calibration and control microspheres.
FLEXMAP 3D Reagents

Shelf-life: Sheath Fluid (20L) and Drive Fluid have a 2 year shelf-life, from the data of manufacture, and Luminex guarantees a shelf-life of no less than 6 months from the shipment date. Sheath Concentrate Pack (1L) has a 9 month shelf-life, from the date of manufacture, and a guaranteed 3 month minimum shelf-life. Calibration and Verification reagents all have a 2 year shelf-life and a guaranteed 6 month minimum shelf-life from the shipment date.

xMAP Antibody Coupling (AbC) Kit
The xMAP Antibody Coupling Kit, or “AbC” kit, is a reagent kit that contains everything a researcher needs to couple an antibody to MagPlex Microspheres. Similar to an ELISA plate, MagPlex Microspheres can be used as the substrate on which immunoassays can be performed. Unlike a traditional ELISA, however, this assay method requires much less antibody, sample, and time; with equal or better sensitivity and dynamic range. Since each microsphere is uniquely addressable, multiple immunoassays can be performed simultaneously with the same sample, in the same well. Once your desired antibody is coupled to the surface of the bead you can begin optimizing monoplex or multiplex capture-sandwich immunoassays to suit your needs.
NOTE: This kit does NOT include microspheres or antibodies.

Magnetic Separators
Magnetic Plate SeparatorThe Luminex Magnetic Plate Separator is a modified plate magnet intended to separate a 96-well plate of MagPlex Microspheres during assay preparation steps. The side clasps on this magnet have been specially designed to hold a round-bottom plate securely to the
magnet, enabling the user to quickly invert and decant it over a waste container. Similar to the method used in an ELISA, this manual plate washing method has been shown to effectively remove nearly all of the supernatant from every well on the plate, with virtually no bead carry-over.
Magnetic Tube SeparatorThe Luminex 1.5mL Tube Magnetic Separator is intended for a single reaction tube. Typically used in bead coupling reactions, this magnet can quickly separate 1mL MagPlex Microspheres from solution and is a handy addition to any xMAP bench top.
