•How is the light reaction affected by the presence or absence of light?

•How is it affected by the boiling of chloroplasts?

LAB FOUR Photosynthesis

Isolated chloroplasts

DPIP replaces NADP+ in the light reactions of photosynthesis.

LAB FOUR Photosynthesis

Isolated chloroplasts

DPIP replaces NADP+ in the light reactions of photosynthesis.

As DPIP is reduced, it turns from blue to colorless.

Adjust empty spectrophotometer to 0%

Adjust with blank cuvette to 100%

Add 3 drops chloroplast to your cuvette and immediately measure transmittance.

Measure again at 5, 10, and 15 minutes.
