4 Purpose: 4.1 The purpose of this Standard Operating Procedure (SOP) is to describe the digestion

procedure used for solid, plant and animal samples at Eurofins Frontier Global Sciences prior to total recoverable trace metals analysis.

5 Scope: 5.1 This method is an acidic digestion procedure for solid, plant and animal samples to

provide a homogenous matrix suitable for analysis of total recoverable trace metals by various methods.

5.2 Sample analysis may be performed by various methods, including Inductively Coupled Plasma-Mass Spectrometry (ICP-MS), Cold Vapor-Atomic Fluorescence Spectrometry (CV-AFS), and Hydride Generation-Atomic Fluorescence Spectrometry (HG-AFS). PUBLIC

6 Basic Principles: 6.1 Homogenized samples are accurately weighed into a tared 30 mL Teflon vial and

nitric acid is added.

6.2 The vials are capped; the threading is wrapped with PTFE tape and heated in an oven for 5 hours.

6.3 The samples are diluted to with reagent water.

6.4 Samples are diluted with reagent water prior to ICP-MS analysis.

6.5 Samples are diluted with HCl prior to HG-AFS analysis.

6.6 Mercury in animal tissue is most frequently digested according to FGS-011 “Digestion of Tissues for Total Mercury Analysis Using Nitric and Sulfuric Acids (70:30)”. For limited mass samples, this digestion is used for CV-AFS.

7 Reference Modifications: 7.1 No significant modifications were made to this method.

8 Definitions: 8.1 Batch – no more than 20 client samples grouped for preparation. 1 Preparation Blank

per 10 samples, 1 CRM or 1 LCS/LCSD (or BS/BSD) per 20 samples; 1 MS/MSD set is prepared for every 10 samples.

8.2 Celsius (C), conversion of Celsius to Fahrenheit: (C * 1.8) + 32.

8.3 Fahrenheit (F), conversion of Fahrenheit to Celsius: (F - 32) / 1.8.

8.4 Method Detection Limit (MDL) – the limit derived from an exercise as described in 40 CFR, Part 136, Appendix B. The exercise produces a defined value that is the minimum concentration that can be measured and reported with 99% confidence that the analyte concentration is greater than zero from a given matrix.

8.5 Certified Reference Material (CRM) – a standard of known composition that is certified by a recognized authority and representing a sample matrix. It is used to verify the accuracy of a method.

8.6 Laboratory Control Sample (LCS) and Laboratory Control Sample Duplicate (LCSD), is a sample containing known concentrations of the analytes of interest that is taken through the entire preparation and analysis process in the same manner as the samples to monitor complete method performance. A Certified Reference Material (CRM) is preferred as the LCS, but a blank spiked sample also meets the requirement.

8.7 Preparation Blank (BLK) – Method blanks consist of the same reagents used to digest the samples, in the same volume or proportion, and are carried through the complete sample preparation and analytical procedure. Teflon boiling chips are added to the preparation blanks. PUBLIC

8.8 Matrix Duplicate (MD) – a representative sample is selected and digested in the same manner. This QC sample will indicate sample homogeneity on the analytes of interest and is prepared at the client request.

8.9 Matrix Spike (MS) and Matrix Spike Duplicate (MSD) – a representative sample is selected and spiked at the same levels as a Blank Spike. These QC samples will indicate sample matrix effects on the analytes of interest.

8.10 May: This action, activity or procedure is optional.

8.11 May Not: This action, activity or procedure is prohibited.

8.12 Shall: This action, activity or procedure is required.

8.13 Should: This action, activity or procedure is suggested, but is not required.

9 Interferences: 9.1 Tissue samples with high levels of Calcium may cause interferences.

9.2 If a non-routine element is requested it is important that the Project Manager informs the Trace Metals Laboratory Manager before samples are received. In some cases these standards may not be available and will need to be purchased prior to the arrival of samples.

10 Safety Precautions, Pollution Prevention and Waste Handling: 10.1 Personnel will don appropriate laboratory attire according to the Chemical Hygiene

Plan. This includes, but is not limited to, laboratory coat, safety goggles, and nitrile gloves under clean gloves.

10.2 The toxicity or carcinogenicity of reagents used in this method has not been fully established. Each chemical should be regarded as a potential health hazard and exposure to these compounds should be as low as reasonably achievable. Chemists should refer to the MSDS (Material Safety Data Sheets) for each chemical they are working with.

10.3 All personnel handling environmental samples known to contain or to have been in contact with human waste should be immunized against known disease-causative agents. Eurofins Frontier will reimburse the expense of Hepatitis A and B immunizations for any laboratory staff member who desires this protection.

10.4 Nitric acid (HNO3): Corrosive. Strong oxidizer. Contact with other material may cause a fire. Causes eye and skin burns. May cause severe respiratory tract irritation with possible burns. May cause severe digestive tract irritation with possible burns. For more information see MSDS.

10.5 NEVER HANDLE heated digestion sample vials or bottles without safety goggles, clean gloves, and a lab coat!

10.7 Pollution prevention information can be found in the current Eurofins Frontier Global Sciences Chemical Hygiene Plan (CHP), which details and tracks various waste streams and disposal procedures.

10.8 All laboratory waste is accumulated, managed, and disposed of in accordance with all federal, state, and local laws and regulations. Any waste generated by this procedure should be disposed of according to SOP FGS-099 “Waste Disposal Procedure for Client Sample Waste,” which provides instruction on dealing with laboratory and client waste.

11 Personnel Training and Qualifications: 11.1 An analyst must perform an initial demonstration of capability (IDOC) that includes

four replicates of a secondary source before being qualified to analyze samples without supervision. Continuing DOC will be maintained and monitored via performance on CRMs and other QC samples, as well as obtaining acceptable results on proficiency testing exercises.

11.2 The analyst/laboratory technician must have read this SOP and other relevant SOPs and have the training documented on the applicable form(s). The analyst may be questioned on SOP by supervisor(s) and/or trainers.

11.3 Training is documented by the employee and supervisor, and is kept on file in the QA Office. The employee must read, understand, and by signing the training document, agree to perform the procedures as stated in all Standard Operating Procedures (SOPs) related to this method.

11.4 Reading of the SOP must be documented on the correct form such as “Standard Operating Procedure Training Record,” Appendix F in FGS-094, the last page of this SOP, Appendix A “Standard Operating Procedure Training Record” or a similar document.”

11.5 All employees must also, on a yearly basis, read the Quality Manual (QM), and complete the yearly Ethics training.

11.6 All training documents including IDOCs, CDOCs, SOP reading, Initial QA orientation, and Ethics training are stored by the Quality Assurance Manager in the employees training file for ten years after the employee is no longer working for Eurofins Frontier Global Sciences.

11.7 Chemical Safety Training, Compressed Gas Training, Chemical Hygiene Plan documentation, and Shipping of Hazardous goods, are stored by the Health and Safety Officer for ten years after the employee is no longer working for Eurofins Frontier Global Sciences.

12 Sample Collection, Preservation, and Handling: 12.1 Samples must be collected in accordance with established ultraclean sampling

techniques (see FGS-008 “Ultra Clean Aqueous Sample Collection”). Samples may be in commercially available clean glass containers with Teflon-lined caps or 125 mL or 250 mL HDPE jars. PUBLIC

12.2 Tissue sample preservation - The tissue sample must be frozen in the sampling container at less than -15°C or freeze-dried and stored at room temperature. The holding time for tissue samples is 1 year.

12.3 Just prior to digestion, samples are thawed and if necessary homogenized. The sample is well mixed to ensure the most representative sample possible.

13 Apparatus and Equipment: 13.1 LIMS

13.2 Teflon vials (30 mL)

13.3 Centrifuge tubes for post digestion storage: 50 mL Digestion vessels (CLASS-A) from SPC Science, or equivalent, with white digestion caps, or Centrifuge Tubes from CPI International or equivalent that have volume tested at 50 mL size and have been cleaned according to FGS-065 are used for common contaminant metals. If a volume tested tube is used, volume testing must be verified for the lot number used and must be submitted for review.

13.4 Analytical Balance: A laboratory analytical balance capable of weighing to ± 1 mg, with documented calibration.

13.5 Pipettors: All-plastic, pneumatic, fixed volume and variable pipettes in the range of 5 µL to 10 mL. Pipettes are to be calibrated weekly according to SOP FGS-003 and FGS-155.

13.6 Clean-Air Station

13.7 Oven: A lab-type oven, capable of reaching 160°C.

14 Reagents and Standards:

14.1 Reagent Water: 18 MΩ ultra-pure deionized water starting from a pre-purified (distilled, R.O., etc.) source.

14.2 Nitric Acid (HNO3): Low trace-metal grade nitric acid. This reagent should be from a lot number that has been previously tested to be low for the analytes of interest. This reagent shall be entered into LIMS and the expiration date is set to 3 years after receipt date.

15 Procedure: 15.1 If needed, the sample is dissected and homogenized.

15.1.1 The process used for homogenization, number of samples, work order number, client name, and initials of the lab assistant are entered into the Tissue Homogenization Log.

15.2 Turn on the oven. Place a calibrated oven thermometer inside the oven. This thermometer is used to measure the actual temperature in the oven.

15.3 Place a piece of lab tape on each Teflon vial so that it goes all the way around the vial. Write the sample IDs on the tape of each Teflon vial.

15.4 Tare vial on the balance. Using a clean spatula, accurately weigh out homogenized wet or dry sample into the vial. Weigh out Teflon boiling chips to the prep blanks and liquid blank spike samples. Add appropriate spikes to QC samples.

15.5 Record the sample ID and the sample mass in the Oven and Digestion Logbook as well as on the tape on the vial.

15.6 After all samples have been weighed out, wrap Teflon tape around the threads at the top of each vial.

15.7 Pipette HNO3 into each vial.

15.8 Tightly screw the caps onto the vials and place in the oven and heat samples at for a total digestion time of approximately 5 hours.

15.9 After digestion, allow the samples to cool to room temperature, and then dilute each sample with reagent water and invert.

15.10 Once diluted, allow samples to sit for to allow solid material to settle. Decant the sample into a centrifuge tube for long term storage.

15.11 Instrument Maintenance and Troubleshooting

15.11.1 The oven is verified to be functioning correctly each day through the use calibrated thermometers. In addition, annual calibration and maintenance is performed through an outside vendor.

15.11.2 If the oven is found to not be able to maintain the appropriate temperature range on the date of analysis the analyst will notify their supervisor and QA immediately and mark the oven as non-functioning and to not use. An external repair group will be called out to correct any problems with the oven and it will be recalibrated prior to resuming digestions using the oven.

16 Calculations: 16.1 This preparation procedure does not involve calculations.

17 Statistical Information/Method Performance: 17.1 Method Detections Limits (MDL) are determined according to 40 CFR Part 136,

Section B. Ten replicates (9 degrees of freedom) are run with a sample that is spiked

3-10 times the expected MDL. The standard deviation (σ) is taken from the resulting data and the MDL is calculated as follows: MDL=2.821* σ .This value should not be interpreted as the method reporting limit.

17.2 The Practical Quantitation Limit (PQL) is the reporting limit for this method and is included as the lowest calibration point (2003 NELAC regulation 5.5.5.2.2.1.h.3). The PQL is determined by running ten replicate samples with a concentration that will produce a recovery of 70-130% for most analytes, but the recovery requirements are analyte dependent. The PQL is referred to as the Method Reporting Limit (MRL) in LIMS.

17.3 Current LODs and PQLs are stored at: \General and Admin\Quality Assurance\MDLs & PQLs.

18 Quality Assurance/Quality Control:

18.1 Method Blank, 1 method blank per 10 samples must be digested, no more than 20 per grouped preparation. The Method Blank (MBLK) is prepared with a similar mass of Teflon boiling chips as the samples, with the same reagents, and put through the same preparation process as the samples.

18.2 Laboratory Control Spike and Laboratory Control Spike Duplicate, a blank spike and blank spike duplicate (BS1/BSD1) is prepared with a similar mass of Teflon chips and is spiked with a known concentration of the metals of interest.

18.3 For every set of 20 samples or less digested, at least one relevant matrix Certified Reference Material (CRM) should be digested. Consult with Trace Metals Laboratory Manager and/or the PM when choosing an appropriate CRM.

18.4 For every set of 20 or less samples, matrix spike/matrix spike duplicate (MS/MSD) should be digested per 10 client samples. Please refer to analytical method for control limits. Spike the MS/MSD exactly the same as the BS1/BSD1.

18.5 Follow the flow charts in EFGS-SOP-038 to determine if any QC falling outside the established control limits can be qualified. Make sure to always use the latest version.

18.6 All the Quality Control limits for the analysis method are included on the “Data Review Checklist.” Make sure to always use the latest version.

19 Corrective Action:

19.1 If the sample was not stored at a temperature less than -18°C and stored at room temperature the Quality Manager must be notified.

19.2 If the oven temperature exceeds 120±5°C, inspect the samples for any obvious volume loss. 19.3 If proper procedures are followed and the analysis QC is still out of range, re-digestion may be required.

19.3.1 In the event that the method blanks are bias high in an analyte of interest, the blank is reanalyzed for confirmation. If the blank confirms the bias high, if the samples within the batch can not be qualified, samples are to be re-digested.
