Investigating testosterone regulation of platelet thromboxane A2 receptors in healthy volunteers and in castrated and uncastrated patients with prostate. Radioligand binding technics, platelet aggregation, assay, molecular pharmacological and cellular methods were employed.

1982-5 Medical Training Fellow in the Clinical Pharmacology Training Program of the University of Glasgow Medical training Fellow, University Department of Materia Medica , Stobhill General hospital , G21 3UW , Glasgow , UK. ( Moved to Dept of Medicine and Therapeutics, Gardiner Institute, Western Infirmary, G11 6NT Glasgow, UK.). Conception and conduct of randomized controlled , single or double blind , or double dummy studies, using cross-over, parallel group or Latin squares designs. Pharmacokinetic analysis of drug concentrations involving both model dependent and independent were undertaken. Population pharmacokinetics using NONMEM ( Non-Linear Mixed Effect Model) was used to describe population kinetic parameter values for investigational new drugs such as enalaprilat ( MK422) or lisinopril ( MK521). Pharmacokinetic-Pharmacodynamic concentration effect models were employed in order to

predict drug effect from plasma drug concentrations by comparing, linear, log-linear or Sigmoid –E max models with or without an effect compartment. Statistical analyses of data were undertaken using parametric and non-parametric methods, including different types of analyses of variance, multiple and logistic regression analysis. Clinical Pharmacology Consultation to Physicians in the Greater Glasgow Health Board Area and within Stobhill Hospital.

The Phase 1 evaluation of Investigational New Drugs for the Pharmaceutical industry was also undertaken for Merck ( MK421, MK422, MK 521 – enalapril, enalaprilat and lisinopril ), Hoffman la -Roche ( Ro-3112 –cilazapril ), Sevier ( S-9490- perindopril) and Squibb (captopril). Low molecular weight heparin – Dalteparin and enoxaparin (Rhone-Poulenc –Rorer ) during the course of these training and practice.

II Clinical Practice and Clinical pharmacologist Roles :Consultant Physician Clinical Pharmacologist and Cardiologist 1989- 1999: Teaching medical students and residents in the Cardiovascular Medicine Unit. Ambulatory and hospital care of Cardiac patients.( 2 clinics a week) Routine echocardiography , cardiac autonomic function and Treadmill Exercise ECG reporting, Intra-operative monitoring of high risk patients and Intensive Care patients.Evaluation of new and old drugs in clinical trials and interaction with Pharmaceutical industry representatives. Monitoring of adverse drug reactions and interactions.Initiated the first hospital Adverse drug reaction form and arranged seminars for its utilization. Chairman of Hospital Drug Formulary Committee ( wrote new hospital drug formulary) and Essential Drug list. Member, Hospital Drug Revolving Fund Committee – Reported to Hospital Management Board and Government. Member Hospital and University Research and Ethics Committee of OAUTHC Hospitals.

Phase 1drug evaluation – Studies on healthy volunteers, involving combination of Alpha-1 blockers, ACE-Inhibitors, or Calcium Channel blockers in Nigerians. Beta-blockade effects on intraocular Pressure (IOP) in Nigerians.

Phase –II/IIIB drug evaluation – Enalapril in hypertension and heart failure, and hypertensive –diabetics ( Merck Sharp & Dohme UK ) ,

Lisinopril ( Astra –Zeneca UK) , Cilazapril in hypertension ( Hoffman –La Roche ) –UK and Germany. , -lacidipine in hypertensive blacks ( Glaxo –UK ) , Amlodipine in hypertensive blacks comparison to hydrochlorothiazide ( Pfizer Nigeria PLC ). Captopril-prazosin in hypertensive acute pulmonary edema ( Pfizer –Nigeria PLC ).

Phase IV -, safety , efficacy and adverse effects – ACE inhibitor induced cough and angioedema in blacks, ACE Inhibitor reduction in mortality in black Africans with congestive heart failure. [ See publications ]

Current Research ( i) The interaction of sex-hormones , hyperinsulinemia and paracrine autocoids in the progression of type-2 diabetic endotheliopathy in Zucker diabetic rats –.(ii) The influence of ethnicity and hemoglobin AS trait on platelet and microvascular reactivity and on the target organ complications of type-2 diabetic patients using doppler –laser flowmetry. (iii) Cross-talk among the heptahelical G-protein coupled receptors in health and disease. (iv) The role of angiotensin II receptor subtypes and nitric oxide synthase isoforms in the development of cardiac cachexia in experimental and clinical heart failure. (v) Determination of the chemistry of an African aphrodisiac penile erection enhancer, [ Burantashi ]with alpha-2 antagonist, endothelin –1 and nitric oxide actions. ( vi) Hyperglycemia – Protein Kinase C connections – the physiological and pathological role of androgen receptor in diabetic microvascular disease. (vii) The role of matrix metalloproteinases in valvular regurgitation and left ventricular remodeling in hypertensive heart disease.(viii) Combined Renin –Angiotensin-System Blockers Effects on the Cardiac remodeling in Hypertensive heart disease and Failure.
