Virus Morphology
Single-stranded RNA genome surrounded by
an icosahedral nucleocapsid and covered by a
lipid envelope
The genome is composed of three structural
protein genes, encoding the nucleocapsid or
core protein C, a membrane-associated
protein (M), an envelope protein (E) and seven
non-structural (NS) protein genes

Antibody Responce
Primary Infection
IgM antibodies appear approximately 5 days after onset of symptoms and rise forthe next 1-3 weeks.
IgM antibodies detectable for up to 6 months.
IgG are detectable at approximately 14 days after onset of symptoms and aremaintained for life.
Secondary Infection
Approximately 5% patients do not produce detectable levels of specific IgM.
IgM titer can be slower to rise in secondary infection.
IgG appears approximately 2 days after symptoms appear.
IgG titre significantly higher in secondary infection.

Dengue fever is an acute viral illness characterized by:
Fever, often with sudden onset;
Severe headache, often described as retro-ocular;
Myalgias and arthralgias that can be very severe;
Nausea and vomiting;
A rash that may present at different stages of the
illness, and whose appearance can be variableit maybe maculopapular, petechial, orerythematous. And
Hemorrhagic manifestations

NS1
a glycoprotein is detected in high titers in
patients with primary or secondary dengue
infections but its function is unknown
found associated with intracellular organelles
or is transported through the cellular
secretory pathway to the cell surface
found circulating during the acute phase of
the disease in sera from patients

IgM Spesific dengue
dengue virus-specic IgM as a antibodies
response of dengue virus infection
Produce after 3 days of fever onset usuallyappears on Days 45 of the illness

Benefit NS1
NS1 AG test kit gives an overall higher sensitivity
rate than the current three established diagnostic
test methods for laboratory diagnosis of acute
dengue infection
NS1 antigen-capture ELISA gave a higher positive
detection within the rst four days of illness
NS1 antigen-capture ELISA has the addedadvantage of continuing to give good detection
rates up to seven days of the illness

NS1
Still limited by the need for sophisticated
instrumentation
higher technical skill which is normally onlyavailable in large hospital diagnostic
laboratories

IgM ELISA Captured
The sensitivity rate of IgM assay for early
diagnosis of dengue was poor in the rst three
days of the illness
specic IgM was merely indicative of recent
dengue infection, and not conrmative of
acute dengue infection
