Unique Surface Chemistry for Enhanced Cell CultureConsistent cell culture conditions arerequired for reproducible researchresults. In the manufacture of all cellcultureware, hydrophobic polystyreneis permanently rendered hydrophilicto support cell attachment andspreading.1,2,3 The consistency of thissurface depends on the treatmentmethod used.

Many manufacturers have long usedatmospheric plasma treatments (i.e.,corona) to create hydrophilic surfaces.In corona treatment, a high-voltagedischarge creates a reactive gas plasmaabove the growth surface of the vessel.In this process, the highly interactivegas plasma mixture is exposed toambient air. The consistency of the treatment surface can therefore be compromised by day-to-dayenvironmental changes.

At BD Biosciences, molded polystyrenevessels are placed in a closed, highlycontrolled environment that preventscontamination and ensures a consistenttreatment surface.

BD Biosciences made a significantresearch investment to develop thisunique vacuum-gas plasma processused to produce both BD Primaria™

and traditional tissue culture (TC)surfaces on BD Falcon™ dishes,plates, flasks, and roller bottles.

The incorporation of nitrogen-containing cations has been correlatedto attachment and spreading of primaryendothelial cells in a clonal cell-growth assay.4 The complex surfaceon BD Primaria cultureware ishomogeneous and stable and has beenin use by researchers for over a decade toimprove attachment and differentiationof a variety of cell types. For example,cell biologists have used BD Primariafor cultivating hepatocytes,5,6 neuronalcells7, and other endothelial cells.8

The surface chemistry of BD Primariaproducts has been confirmed byElectron Scanning for ChemicalAnalysis (ESCA).

BD Biosciences’ cell culture productsmeet high quality control standardsand are manufactured under theregistered ISO 9000 series QualitySystems. A sensitive clonogenic assayis used to validate the specificconditions used to treat each new TCproduct. Routine, 72-hour confluencytesting ensures lot-to-lot quality. Our industry-standard plug-seal caps provide a liquid-tight sealwhen closed and an open incubationposition for reliable gas exchangewhen partially opened. Our ventedcaps allow consistent gas exchangewhen the caps are in the closed posi-tion. Gases required for cell growthand metabolism pass freely throughthe vent while microorganisms cannot.Lined, phenolic caps are available forlong-term cultures. Vacuum-gas plasma treatment permanently and consistently modifies the cellgrowth surface for uniform chemistryand attachment.

BD Falcon Dishes are exceptionallyflat for distortion-free optics. The dish lids are designed for optimal gasexchange. Stacking rings on the dishesallow for easier stacking and handling.Sterilized by gamma irradiation, thecrystal-grade polystyrene dishes arepackaged in peel-open, medical-style bags.

Our easy-grip dishes improve handling.The unique design and frosted rimfound on BD Falcon Easy-Grip Dishesimprove handling. The ability to pickup a small dish conveniently withoutaccidentally removing the lid allowsyou to work faster and improves aseptic manipulation.

A sensitive clonogenic assay usingMRC-5 cells, a diploid human fibro-blast line, is used to validate the man-ufacturing process for each BD FalconTC product. Routine testing of standardTC products is performed by testinggrowth to confluency at 72 hours withMRC-5 or WI-38 cells, a diploid humanlung fibroblast cell line. The surfacechemistry of each lot of BD Primariaproducts is confirmed by ESCA.

Roller BottlesThe BD Falcon™ TufRol™ RollerBottle has a one-piece design. Whilepolystyrene does not normally offerhigh-impact resistance, the TufRolbottle is manufactured using a propri-etary manufacturing process thatprovides excellent impact resistance.This inherent, high-strength featurereduces the risk of messy spills andloss of precious contents during acci-dental mishandling.

Roller bottles are used in both researchand manufacturing applicationsinvolving the scale-up of mammaliancells for purposes of virus propagationand bioproduct production. BD FalconRoller Bottles are tissue culture-treated,non-pyrogenic, and sterilized by gammairradiation. As with most BD Falconproducts, our Roller Bottles aremanufactured under a registered ISO 9002 Quality System.

Custom ProductsSince many industrial customers have special requirements relative to labeling, lot control, packagingconfigurations, and delivery schedules,we are able to develop custom servicesand products to suit individual needs.As with all BD Biosciences products,certification is available upon request.

Multiple Cap Styles andSurface TypesBD Falcon Roller Bottles are availablewith a smooth surface with a growtharea of 850 cm2 or a pleated surfacewith a growth area of 1450 cm2. Easy on/off or vented cap styles areavailable. The vented cap has anintegral hydrophobic 0.2 µm micro-porous membrane filter vent, allowingconsistent gas exchange when the capis in the closed position.

Automation-FriendlyOne-piece design of BD Falcon RollerBottles is compatible with manual orautomated filling systems. Our RollerBottles are supplied with easy on/offpolyethylene caps that are designed toprevent misthreading and to provide asecure seal.

BD Cell™ MAb Medium

BD Cell™ is a new basal medium formu-lated to produce high-yield monoclonalantibody (MAb) secretion using serumor serum-free supplementation. Thissystem enables the purification of 5 to25 times more antibody compared toconventional media with similar sup-plementation. BD Cell also reducesmedia consumption and labor cost.

Benefits of BD Cell™ in Roller Bottle ProductionFirst evaluated in roller bottles, BD Cellhas proven itself a worthy alternativeto traditional basal media. While astandard method of culturing anti-body, roller bottle production hassome limitations: extensive labor andfrequent feedings. The initial rollerbottle evaluation compared BD CellMAb Medium (basal) with Iscove’sModified Dulbecco’s Medium (IMDM)both supplemented with 10 percentfetal bovine serum (FBS). The cultureswere inoculated at a density of 2 x 105

cells/ml. The IMDM was run conven-tionally with harvesting and feedingperformed every two to three days. By contrast, the BD Cell culture wasrun batch-fed and incubated untouchedfor 21 days. Now evaluated in a pro-duction environment on more than300 hybridoma lines, BD Cell’s averageincrease of antibody yield is 8 to 10times that of classic IMDM media.

In the biopharmaceutical industry,Bovine Serum Extract (BSE) concernshave resulted in the need to replacemedia that contain animal-derivedcomponents with a non-animal alter-native. The challenge has been tomake substitutions that do not adverselyaffect the performance of the cell lines.A selection of non-animal peptones was

evaluated in BD Cell MAb Medium tooptimize a cell culture medium thatwas free of animal components. Thesoy hydrolysate Select Soytone waschosen based on optimal performanceand was used to formulate a BD CellMAb Medium free of animal-basedcomponents.

CELLine™ System: CultivationFlask for Secreted ProductThe CELLine™ 1000 Device, based onmembrane technology, is a new devel-opment in cell cultivation. The systemguarantees high cell densities and iseasy to use for recombinant proteinexpression and high yield monoclonalantibody production.

Monoclonal antibodies (MAbs) arebecoming increasingly important asresearch tools. The primary methodsavailable to generate research quanti-ties of MAbs (10 mg to 500 mg) arestatic tissue culture, spinner, or rollersystems and ascites fluid from mice.As demand for MAbs has increased,so has the pressure to develop alterna-tive in vitro production methods thatwill reduce animal use, streamlinedownstream processing, and reducevariability in production runs. Theeffort to develop a production methodthat meets these emerging require-ments has culminated in the CELLineSystem, a novel, membrane-based dis-posable cell cultivation system.

The CELLine System yields antibodyconcentrations comparable to that ofascites. One CELLine flask can be usedto produce as much antibody as thatof 12 mice. The harvest volumes alsoresult in antibody concentrations thatare 50 to 100 times higher than bothroller bottles and tissue culture flasks.Average yields of monoclonal antibod-ies range from 1 to 5 mg/ml. Using BDCell MAb media in a CELLine device,antibody yields average between 30 to150 mg of antibody every two weeks.

CELLine Membrane Technology

Cells are cultured in a 15 ml cultivation chamber separated from the 1000 ml nutrientsupply compartment by an upper semi-permeable membrane. Nutrients and other smallmolecules pass across the semi-permeable, cellulose acetate membrane of the cellcompartment. Any cell-secreted product with a molecular weight of over 10,000 Dalton is retained in the growth chamber of the device. The molded silicone membrane on thebottom of the device allows oxygen to reach the cells from underneath. The cells settleupon the bottom of the cell compartment atop of this gas exchange surface, across whichoxygen and carbon dioxide rapidly diffuse. This approach leads to high cell concentrationswithin the small cell compartment volume.

Separate ports provide selective access to the nutrient supply chamber and the cultivationchamber. The solid white cap identifies the cultivation chamber port while a blue vent capidentifies the nutrient supply chamber. The cell chamber is accessed via the cultivationchamber port via a serological pipet. The total system is transparent to allow visibility.

Pre-tested BD Falcon™ In VitroFertilization (IVF) ProductsBD Falcon™ In Vitro Fertilization (IVF)products are the first plasticwareavailable that is certified sterile, non-pyrogenic and non-embryotoxic. Thesepre-tested products save you time andexpense in complying with the Collegeof American Pathologists (CAP) andAmerican Fertility Society (AFS) recommended standards for IVF labs.

Mouse embryo testing

Test articles are prepared in compli-ance with cGMP standards. Each lotof BD Falcon IVF product is tested forembryo toxicity using the 1-cell mouseembryo assay. Mouse embryos are iso-lated at the 1-cell stage from B6C3F1females following super ovulation withgonadotropin and mating with B6D2F1males. BD Falcon IVF products aretested using both a direct and indirecttest method. In the direct assay, theembryos are cultured directly in the

IVF labware. For the indirect assay,culture media is incubated in the IVFlabware for 24 hours at 37ºC. Theincubated media is then transferred toa culture plate and the 1-cell mouseembryo assay is performed. Productsare deemed non-embryotoxic if theysupport the growth of more than 75percent of the embryos to the expandedand/or hatched blastocyst stage.

To improve manipulation of ova andembryos, BD Biosciences designed, inconjunction with embryologists, aninnovative 4-well plate. A unique lidreduces the risk of contamination andminimizes evaporation by providingaccess to two wells at a time, whiletwo remain covered. The wells arenumbered and a large writing patchallows clear sample identification.Plates are packaged in individual peel-open trays for sterile presentation.

BD Falcon IVF Dishes and 4-well Platesare manufactured from virgin crys-talline polystyrene tested for USP Class

VI cytotoxicity. They have perfectly flat,optically clear surfaces for optimummanipulation and observation of ovaand embryos. Lids were designed foraseptic manipulation and consistentventing to maintain humidification.
