INTRODUCTION Preservation by Plastination The study of biological specimens is significantly impeded by decay. Thus for centuries, people have been looking for appropriate methods of preservation. Thanks to plastination, specimens can be preserved for research, teaching, and demonstration purposes in a lifelike and durable manner. Specimens are impregnated with special reactive polymers in a vacuum process. The mechanical (flexible or rigid) and optical (translucent or opaque) properties of the polymers used determine the characteristics of the preserved objects. Plastinated specimens are dry and odorless; they maintain their original surface relief and are identical to their pre-preservation state, down to the cellular level. Histological studies can even be performed on them. Plastination involves replacing the water and fat in tissues with a reactive polymer such as silicone rubber, epoxy, or a polyester resin: In a solvent bath, initially the tissue water is replaced by freeze-substitution, and later, at room temperature, the tissue fats are gradually replaced by acetone. The dehydrated and degreased specimen subsequently is placed into the polymer solution.

Plastinated specimens maintain their original surface relief.

Under vacuum conditions, the gaseous solvent is then continuously extracted from the specimen, creating a lower pressure that gradually drives the polymer into the tissue. After this “forced impregnation,“ the specimen is cured with special hardeners, UV radiation, or heat, depending on the polymer used.

Hardening of a silicone-impregnated specimen

in a gas curing box A special variation of plastination is so-called sheet plastination. With this method, specimens such as individual organs or entire bodies, mostly in a deep-frozen state, first are cut or sawn into slices that are 2 mm to 8 mm (about 1/12 in to 1/3 in) thick. These slices are placed between polymer nettings, dehydrated, degreased, and eventually impregnated with a polymer under vacuum conditions. To give the specimens a smooth surface, the impregnated slices either are cured between foil or have additional resin casted around them inside a flat chamber. The refractive index of the resin used determines the optical properties of the plastinated body slices: Epoxy resin yields translucency and good coloration of the various tissues. Polyester resin, which is used for plastination of brain slices, allows for particularly good discrimination between white and gray brain matter.

Plastinated slices of organs and bodies constitute excellent teaching materials in cross-sectional anatomy, a field of ever-increasing importance, and they correlate well with radiographic images. Serial translucent body slices are useful in various scientific research approaches. In addition, they are a suitable diagnostic aid in pathology because they allow for quick macroscopic-diagnostic screening of entire organs and organ specimens. Pathologically modified tissue areas can then be selectively analyzed with conventional histological methods.

The plastination process has been constantly refined since its invention in 1977 at the Institute of Anatomy at Heidelberg. Today, it is recognized as a valid method of preservation and is practiced at more than 400 institutions in 40 countries. The main reasons for this widespread popularity are the toughness, the durability, the lifelikeness, and high teaching value of plastinated specimens. Note:

Numerous customer interviews and trainings have made clear that it is best to begin your own endeavors in plastination with the BIODUR® S 10 standard technique. This will allow you to master the technology through hands-on experience so that you can produce appealing specimens. This approach also will provide you with the foundation for mastering one or more sheet plastination techniques. Using your acquired knowledge and upgrading your plastination equipment accordingly will enable you to produce high-quality sheet plastinates within a reasonably short time. Experience has taught us that highly specialized plastination tasks, such as the exclusive production of brain slices, are not for beginners and require specific, in-depth training.

II. POLYMERS & CHEMICALS Plastination with BIODUR® Silicones Silicone polymers S 10, S 15, S 49, and S 14 Red are available for different applications. The impregnation or processing mixture consists of the silicone polymer and the added hardener. Final curing of S 10, S 15, and S 49 is achieved with hardener S 6 in its gaseous state. (For equipment, see also the section on “Accessories for Polymer Curing,“ page 34). Silicone Polymers KSK01 BIODUR® S 10 – Suitable for all putrefiable macroscopic specimens. The S 10 standard technique is easy to handle, not susceptible to mistakes, and requires only a relatively small amount of technical equipment. Thus, it is especially suitable for beginners.

BIODUR® S 10/BIODUR® S 3 impregnation bath can be continuously used until completely depleted or can be frozen at –70°C (–94°F).

KSK03 BIODUR® S 15 – Silicone of low viscosity and reactivity, allowing for impregnation at room temperature. Recommended for the plastination of archeological wooden objects of any size, especially because wood can be impregnated with BIODUR® S 15 using ethanol as the final dehydration intermedium. BIODUR® S 15 is also recommended for the plastination of organs and organ packages (not suitable for skeletal muscle specimens).

KSK05 BIODUR® S 49 – High-viscosity silicone for the surface treatment of specimens that in their final state are supposed to be shiny and of high-impact strength. Examples of recommended use include joint planes, placentas, and cut surfaces of organs. Before gas curing, thinly coat specimens with BIODUR® S 49/BIODUR® S 3. Afterward, cure with BIODUR® S 6. For use on already-cured silicone specimens, the surface must first be treated with BIODUR® Adhesion Mediator AS 70 (page 16).

Silicone Hardeners KSH01 BIODUR® S 1 – Hardener (infiltration hardener) for BIODUR® S 10-impregnated and BIODUR® S 15-impregnated specimens for fast curing of deep or sealed-off areas of specimens (e.g., deep gluteus muscle region, prostate gland, erector spinae muscle). Infiltration hardening is done in parallel to gas hardening. Not recommended for general use, owing to possible loss of optical and/or mechanical quality. Also use hardener BIODUR® S 1 for curing injection polymer BIODUR® S 14 Red.

Epoxy Resins KEP01 BIODUR® E 12 – Standard epoxy resin of medium viscosity for sheet plastination; translucent, firm, and slightly flexible when cured. Choose from different hardeners to vary the properties of the specimens: BIODUR® E 12 in combination with hardener BIODUR® E 1: Standard epoxy resin for sheet plastination. The properties of the hardener produce especially good staining of specimens. BIODUR® E 12 in combination with hardener BIODUR® E 6 and accelerator BIODUR® E 600: Suited for plastinated tissue slices to be examined histologically. The cured polymer can be removed from the specimen with a saturated solution of sodium methylate in anhydrous methanol. BIODUR® E 12 in combination with hardener BIODUR® E 7 and accelerator BIODUR® E 700: For producing thin-ground section specimens. The cured polymer becomes very firm and cannot be separated from the glass.

BIODUR® E 12 will partially crystallize after only a few days, especially when stored cool. Heat to +50°C to +70°C (+120°F to +160°F) before use. It is advisable to order BIODUR® E 12 in buckets of a size compatible with the available heating cabinets.

KEP03-KEP05 BIODUR® E 20 Plus – Capillary-passable epoxy resin for color-contrast presentation of vessels in plastination and for the manufacturing of corrosion specimens. BIODUR® E 20 Plus is characterized by very low viscosity, great firmness and a low (<2%) tendency to shrink, is resistant to acetone, and can withstand methylene chloride for a short period of time (<24 h). The processing properties and the flexibility of the cured resin can be widely varied by using appropriate additives (plasticizers, organic solvents; see page 16). Available in clear, red, or blue. BIODUR® E 20 Plus Clear can be pigmented with dye pastes (see page 16) as required.

Formaldehyde content: 16%–18%. The fixing, aqueous liquid part of the solution penetrates gradually after injection into the surrounding tissue, while the plastic component remains inside the vessels and hardens there. Specimens injected with BIODUR® Solution U are suitable for subsequent silicone plastination.

Special Notes on BIODUR® Polymers

Because of the complex nature of polymer systems required for plastination, we always recommend purchasing entire polymer kits (i.e., the polymers together with the matching hardeners and additives in the quantities required). This is especially true for beginners in plastination.

Experience has shown that the use of unsuitable polymer combinations may cause significant complications, such as these:

(a) Insufficient impregnation or curing of the specimens due to demixing of the polymer components under vacuum conditions or the incomplete chemical binding of monomers and hardeners due to stoichiometric imbalances of the reactive mixture. Both chemical effects will lead to stickiness and unpleasant odors of the specimens, and in duromers may also cause tension cracks.

(b) Over the years, polymers may release compounds that have not completely hardened, and these may in turn cause skin irritations.

(c) Depolymerization due to unintended secondary reactions of chemical compounds of the plastinated tissues with compounds of the polymer mixtures. These may manifest only years after the specimen has been produced.

The required displacement of a plastination vacuum pump depends on the volume of the impregnation bath and the volume of the immersed specimen. As a rule of thumb, we recommend a displacement of 1 m3/h per 10 L (2.6 gal) of impregnation bath. For the small Heidelberg Plastination Kettle (HI02A1.0), for example, with its average filling volume of 15 L (3.9 gal) of impregnation bath, we recommend a pump with a displacement of 1.5 m3/h. When in doubt, opt for the lower of the two speeds you are considering.

Silicone Gaskets – Spacers for flat chamber construction in sheet plastination. The required thickness of the gasket depends on the thickness of the specimens. The gasket should be 2 mm to 3 mm thicker than the section to be plastinated, because the binder clips (see below) used for the construction of the flat chamber will compress the gasket. Silicone gaskets are reusable after removal of polymerized resin residues by immersion in acetone. BIODUR® Gas Curing Units are used for curing of silicone-impregnated specimens with hardener BIODUR® S 6 (page 11). Its modular structure (unit consists of a supply unit and at least one gas curing box) meets requirements for explosion protection yet allows for operation of several different boxes with one supply unit (see diagram), making refitting with small or medium gas curing boxes possible. Plastination Deep-Freezers HK05A1.0, HK06A1.0 Special Freezers for Plastination – With glass sliding lid, –23°C (–9°F), stationary, suitable for freeze-substitution with acetone and for forced impregnation. Outer paneling: brushed stainless steel; interior paneling: stainless steel. See table on page 39 for technical specifications. Unless otherwise specified in your order, the freezer will have four holes drilled into the short side facing the wall, so that vacuum tubing can be inserted (HI07A1.0, page 27). BIODUR® freezers are adapted to the special needs of plastination: The bottom has been reinforced by a metal plate, and the refrigeration unit has been removed and placed together with the temperature control unit in a separate case.

For protection from explosion, the freezer and the compressor housing should be set up in neighboring rooms. Thus, potential ignition sources are kept away from the actual working area in which flammable solvents will be used. The operator is responsible for complying with all national, state, local, and institutional safety regulations!

Freezer and refrigeration unit are connected by two refrigerant pipes and the cable for the temperature sensor.
