Results: R-(+)-1-phenylethanol reduced obviously, while the S-(-)-1-phenylethanol barely reacted.

The conversion of the substrate was 17.2% and the eep>99%.

Further work

To verify the hypothesis of bio-imprinting process in accordance with compulsory-order mechanism, the following work should be done.

Imprinted with R-(+)-1-phenylethanol

Imprinted with S-(-)-1-phenylethanol

Imprinted with Vinyl Acetate and R-(+)-1-phenylethanol / S-(-)-1-phenylethanol

Highlight

The E values didn’t depend on the catalytic constants involved in the acylation step.

The sensitivity of the enantioselectivity was related to the enantiomeric form of the leaving alcohol, indicate that the imprinting effect involves modifications near the active site.

The ENZYME ENGINEERING

Asp and Glu with acidic residues were modified with EDC.

Chemical modification did not have a direct effect on chemical groups related to the enzyme activity, but may be altering the percentage or the shape of the open form of the lipase.

Combining the immobilization and amination seems to be a very useful tool in the combinatorial biocatalysis, which can change the characteristics of the enzyme like specific activity and enantioselectivity.
