Successful experiences in creating technology parks/ incubators

providing a basic infrastructure to transform ideas into products and

services can serve as model for developing Countries.

For example, given the comparative advantage of diversified

ethanol production in the world, particular emphasis could be

focussed upon research and transformation of agricultural products

in such technology parks.

It would also be advisable to set up technology parks near existing

institutions to attract foreign contract research funds to perform

applied research in situ.

La linea pilota sarˆ basata sulle apparecchiature per la macinazione criogenica, la separazione della lignina, il s istema di fermentazione per la preparazione dellÕ inoculo, la produzione e distillazione dellÕ etanolo. Il processo che si sperimenterˆ sarˆ del tipo SSCF ( Simultaneous Saccharification and Co- Fermentation) con la variante di: i) non avere enzimi in soluzione, ii) avere un unico agente microbico, il lievito S. cerevisiae ingegnerizzato con enzimi cellulasi ed emicellulasi espressi sulla superficie cellulare e iii) in grado di trasformare zuccheri in una pi alta percentuale di alcol. Questo processo innovativo, derivante dallÕevoluzione del precedente SSCF, verrˆ indicato con la nuova sigla CEDY (Cellulose to Ethanol Directly by Yeast). La fermentazione un processo complesso ma noto nelle sue variabili e caratteristiche mentre pi necessaria la comprensione dei meccanismi di adesione e catalisi degli enzimi endo- ed eso-glucanasi, cellobioidrolasi ed i corrispondenti per lÕ emicellulosa, ai siti di legame e di catalisi sulla cellulosa ed emicellulosa . Questa dinamica regola i tempi e lÕ efficacia del processo. Il glucosio e lo xilosio saranno metabolizzati dal lievito nello stesso momento della sua produzione per cui mancherˆ lÕ inibizione da metaboliti. La concentrazione del lievito aumenterˆ fino a livelli stazionari in cui sarˆ rimosso dal bioreattore per mantenere il processo nella fase esponenziale. Il tasso alcolico massimo attuale del 13 %, nella fase di ingegneria genetica si cercherˆ di portarlo al 20 Ğ 26% con un notevole guadagno economico nel processo.

3.4 Ingegnerizzazione genetica del lievito 3.4.1 Identificazione dei geni eterologhi degli enzimi necessari Il lievito Saccharomyces cerevisiae, oggi universalmente riconosciuto come il pi importante microrganismo per le biotecnologie, e di grado GRAS (Generally Recognized As Safe), non possiede la capacitˆ enzimatica per metabolizzare la cellulosa, ma bens“ estremamente efficiente nella produzione di etanolo partendo da zuccheri semplici quali il glucosio. Di c onseguenza, occorre introdurre nel lievito i g eni codificanti per gli enzimi che degradano la s truttura fibrillare della celulosa fino a ridurla a l ivello del monosaccaride glucosio che la cellula pu˜ assimilare. Di tali geni esistono diversi esempi, alcuni dei quali giˆ studiati ed isolati da laboratori di r icerca, in particolare in Giappone e Finlandia. Verranno quindi isolati i geni codificanti per i seguenti enzimi: i) endoglucanasi II (EGII) dal fungo Trichoderma reesei, ceppo QM9414, in grado di digerire le catene di cellulosa in modo casuale; ii) cellobioidrolasi II (CBHII) dal fungo Trichoderma reesei, ceppo QM9414, in grado di staccare cellobiosio, cio molecole dimeriche di gl ucosio dallÕestremitˆ della catena di cellulosa; iii) §-glucosidasi I (BGL1) dal fungo Aspergillus aculeatus, ceppo F50, in grado di scindere una molecola dimerica di cellobiosio in due molecole di glucosio. Come miglioramento innovativo dellÕattuale stato dellÕarte, i ceppi transgenici di l ievito cos“ costruiti verranno sottoposti ad u n nuovo sistema di mutagenesi globale per traslocazione cromosomica. In pratica, il ba ckground genetico di questi ceppi verrˆ riarrangiato casualmente su l arga scala sfruttando una rivoluzionaria tecnologia genetica messa a p unto nei laboratori dellÕICGEB dellÕ AREA di Ricerca di Trieste.

3.4.5 Ottimizzazione / Fermentazione La ricerca di condizioni ottimali per la produzione di etanolo verrˆ effettuata sia su piastra, misurando la diminuzione della fluorescenza emessa dalla cellulosa presente. Successivamente i ceppi che daranno i valori pi alti saranno oggetto di mutagenesi globale tramite induzione di traslocazioni cromosomiche per mezzo del sistema BIT (Bridge-Induced Translocation). La selezione avverrˆ per rivelazione automatica di concentrazione di etanolo per mezzo di appositi sensori enzimatici. I ceppi selezionati verranno testati in mini-fermentatori con diverse composizioni di MALIC ed eventualmente riassoggettati a ulteriori cicli di mutagenesi globale fino a dare risultati soddisfacenti, da testare su scale pilota. Per ultimo, utilizzando le recentissime scoperte del gruppo di Gerald Fink negli USA, sul Òquorum sensingÓ del lievito, si utilizzeranno le due molecole-segnale individuate, il feniletanolo e triptofolo, per aumentare la densitˆ massima delle cellule in terreno liquido nonch il g rado alcolico prodotto dai ceppi transgenici.

3.1.1.1 Struttura chimica della lignina La lignina u n polimero amorfo, eterogeneo e c himicamente complesso. LÕeterogeneitaÔ della lignina dovuta al suo elevato peso molecolare derivante dallÕunione di differenti acidi ed alcoli fenilpropilici (cumarilico, coniferilico e sinapilico). LÕaccoppiamento casuale di questi alcoli dˆ luogo alla struttura tridimensionale complessa della lignina. Nelle figure seguenti sono riportate le strutture chimiche degli alcooli (A) e acidi cinnamici (B), noncheÔ la formula strutturale della lignina (C) proposta da Adler. A
