Protein and Nucleic Acid Degradation

A set of enzymes targets a protein for degradation by tagging it with an ~70 amino acid protein called ubiquitin. The ubiquitin tag is recognized by a large protein machine called a proteasome. Upon entering the proteasome, the protein is unfolded and hydrolysed (degraded). Ubiquitin molecules are highly resistant to the proteasome and can be recycled.

Facts:

1. Cln1,2,3 bind Cdk1 in G1 and form an active kinase.

Cells without cln1,2,3 are inviable

2. Clb 5,6 bind Cdk1 in G1 but as long as they are bound

by Sic1 they don’t form an active kinase.

3. Clb5,6-cdk1 activity is essential for DNA replication
