What is Gluten detection?
Different gluten detection kits use different antibodies to identify components of gluten.However, most components of gluten are actually safe for people with celiac disease!Research indicates that a only very small fraction is toxic for celiacs: the 33-mer peptide
of the a-gliadin molecule.
The G12 antibody in all GlutenTox kits was specifically developed to detect this toxicpeptide. G12 searches for the QPQLPY sequence, found only in the 33-mer peptide.
Because of this specificity, G12 is a superior choice for fermented, hydrolyzed, or heat-treated foods with complicated molecular structures.

“Gluten” refers to the storageproteins in certain grains. Gluten iscomposed of several different typesof protein, for example wheatcontains several different types of
gliadins and glutenins.
G12, Celiac Disease and OatsQ: Are oats gluten-free?
A: Sometimes.
Oats are often subject to cross contamination (in the field and/or during processing).
However, just because oats have been grown andprocessed carefully does not mean they are gluten-free.A 2010 study showed that not all oats are created equal.Different strains of oat have different avenin structures, andonly some are toxic to celiacs.
The study used the G12 antibody to review several strains ofoat:
• 3 distinct groups of oat were identified: no, low andhigh toxicity strains.
• The strains were introduced to cells from celiacpatients, and the cells responded appropriately: theyonly reacted to the toxic oat strains.

Q: Are test results valid sooner than 10 minutes / after more than 10 minutes?
A: If the sample has a high concentration of gluten, a pink line may appear in
only a few minutes. This is a valid result. However, if no pink line appears you
must wait the full 10 minutes to declare a truly gluten-free sample.
Test results may shift if left out for too long (positive results fade in intensity and
negative results sometimes develop a faint pink positive line), so it’s important
to check promptly at 10 minutes.

FAQs
FAQsQ: Why is the kit not recommended for chocolate?
A: Gluten from chocolate or cocoa is very hard to extract because of the
phenols present in this matrix: they can chelate gluten. For this reason the level of
gluten can be slightly underestimated. However, if you get a positive result with
chocolate, it definitely does contain gluten and should not be consumed by
celiac disease patients.

A: Yes, the G12 antibody specifically recognizes the
fragment that is toxic to celiac patients and can
therefore assess toxicity even if the food is
fermented, hydrolyzed or heat processed.
Q: Can the kit be used with fermented, hydrolyzed or baked goods ?
FAQsQ: Why is it important to shake the sample vigorously in the extraction stage?
A: This stage is crucial because it breaks apart the sample so that the gluten
molecules can be isolated / detected. The first step is to grind and homogenize
the sample as much as possible. The second step is to shake it, which continues
the grinding/breaking-down process.
Q: What do I do with the test once I’ve used it?
A: All elements of all GlutenTox tests can be disposed of in
ordinary trash, or recycled where appropriate.
If you need to stop briefly during the 2 minutes of
shaking, it’s OK. Just make sure the total amount of
shaking time is at least 2 minutes.
FAQsQ: Do I need to purchase separate swabs for surface testing?
A: Nope! The test strip is all you need. You’ll notice that when the strip is face-
up, one end has a fuzzy, cotton texture. This end is what gets rubbed directly
onto the surface you are testing, and it is also what gets inserted into the
dilution solution.
Q: Can I use GlutenTox Home to surface test?
A: We recommend using GlutenTox Pro or GlutenTox Sticks, however in a
pinch you can modify GlutenTox Home to surface test. Simply pry open the
plastic casing with a thin blade and remove the test strip. You can then treat
it like any other surface test.
One thing to be careful of: always surface test
using clean hands (or a glove), to ensure thatyou do not accidentally contaminate the strip.
