Welcome

Dear Colleagues:

Welcome to the 17th International Congress on Animal Reproduction (ICAR 2012). An

icon of animal reproductive biology, the ICAR is like the Olympics - it has emerged every

4 years since 1948 in some of the most remarkable cities in the world. This is the first

time Canada has ever hosted ICAR and we are excited to have you in Vancouver.

For the Local Organizing Committee, ICAR 2012 began 5 years ago. The LOC has spent

countless hours developing the scientific and social programs, identifying and inviting

world-class speakers, reviewing manuscripts, coordinating poster sessions, satellite

symposia, contracting services, venues and hotels, and promoting what we hope will be

an outstanding Congress. Give them a pat on the back when you get a chance!

The 4-day Congress includes Plenary Sessions, concurrent Symposia and Workshops, and Poster sessions (both traditional

and electronic posters). Invited speakers will present on topics that span the breadth of reproduction from the sperm

to the egg, from genomic conservation to cryopreservation, and from global food systems to reproductive education

systems. The peer-reviewed proceedings include 60 full-length review articles and more than 550 abstracts. Opening and

closing keynote addresses by distinguished authors/scientists encapsulate the theme of ICAR 2012 - Celebrating the past

and embracing the future.

We gratefully acknowledge the financial support of several organizations and institutions, as well as the livestock and bio-

pharmaceutical industries. Furthermore, we congratulate Dr. John Aitken, recipient of the prestigious ICAR Simmet Prize for

Assisted Reproduction.

Welcome to Vancouver! Enjoy the meeting, make new friends and meet old friends, and enjoy the hospitality and breath-

taking scenery of Canada.

ICAR Affairs

The International Congress on Animal Reproduction (ICAR) is a non-profit international organization that conducts conferences on Animal Reproduction. Founded in 1948, ICAR has held a major conference every 4 years in countries throughout the world. The theme of the congress is animal reproduction, including animal physiology, animal pathology and reproductive technology. Each conference has invited plenary and symposia papers, other contributed papers, workshops, and a trade exhibition. ICAR has no

members as such, but is governed by a Standing Committee of representatives of more than 40 countries. An Executive Committee oversees general business according to the Constitution and By-Laws adopted in 1992 and modified in 2002. The Secretary General is responsible for routine administration. The Executive Committee of ICAR is elected from and by members of the Standing Committee, with due reference to maintaining an appropriate geographical representation.

Prizes and Awards

SIMMET PRIZE FOR ASSISTED REPRODUCTION

The ICAR Simmet Prize recognizes an active scientist for outstanding basic and applied research in the area of Assisted Reproduction in Animals. The Prize is awarded every four years at the ICAR meeting and comes with a prize of 50,000 Euro. The Simmet Prize is sponsored by the Simmet Family in honor of Dr. Ludwig Simmet, founder of Minitube.

Dr. R. John Aitken is the 2012 recipient of the Simmet Prize.

Dr. Aitken’s numerous landmark studies in male infertility in domestic livestock and humans have influenced laboratories and allied industries around the world.

His research career began with a PhD in reproductive biology from the University of Cambridge under

the supervision of RV Short. Following post-doctoral positions at the Universities of Edinburgh and Bordeaux, he joined the World Health Organization in Geneva. In 1977, he was appointed to the prestigious MRC Reproductive Biology Unit at the University of Edinburgh to establish a research group in developmental biology with clinical outreach into male infertility. In 1992, he was awarded an Honorary Professorship within the Faculty of Medicine and in 1998 received a ScD from the University of Cambridge in recognition of his research contributions to gamete biology. He then moved to the University of Newcastle, NSW, Australia, as Chair of Biological Sciences, and later, Director of the ARC Centre of Excellence in Biotechnology and Development.

Dr. Aitken has published more than 450 research articles, given more than 300 invited lectures and filed 12 patents. His work has been cited more than 13,000 times (h-index of 60). He has been elected a Fellow of both the Royal Society of Edinburgh and the Australian Academy of Science. Dr. Aitken has supervised 11 MSc students and 23 PhD students.

Dr. Aitken pioneered the development of objective methods for measuring sperm movement, beginning with time-lapse photography and culminating in CASA systems that are now commonplace in modern-day theriogenology. He also developed a novel technique for monitoring the acrosome reaction in live cells, and was instrumental in developing heterologous in vitro fertilization assays of sperm function that are used to monitor male fertility in large domestic animals. He subsequently developed biochemical parameters to predict male infertility. In the course of these studies he pioneered the fundamental concept that oxidative stress is a major determinant of sperm function and DNA damage and developed fluorometry/ flow cytometry based assays that are now widely used in diagnosing male infertility in a range of mammalian species.

Dr. Aitken is among the first reproductive biologists with the vision to implement advanced mass spectrometry platforms dedicated to proteomics and metabolomics in gametes and embryos. Dr. Aitken is an exceptional scientist who has developed a legacy of well-trained graduate students and post doctoral fellows. Dr. Aitken truly personifies the intent of the Simmet Prize in Animal Reproduction.

Keynote and Plenary Sessions

Plenary Session 1: Opening Session & Simmet Prize Monday, July 30 201208:15 – 10:30East Ballroom ABCChair: William C. Wagner

08:50 – 09:40Matthew Cobb, University of Manchester United KingdomAn amazing 10 years: The discovery of egg and sperm in the 17th century.

Matthew Cobb is a professor of zoology and Associate Dean for Social Responsibility, University of Manchester, UK. Author of the THE

EGG AND SPERM RACE (Simon & Schuster / Bloomsbury, 2006) and THE RESISTANCE (Simon & Schuster, 2009), he is a frequent contributor to the Times Literary Supplement and Journal of Experimental Biology.

Abstract: “Before reproduction: How we discovered the secrets of sex.”  The scientific identification of the key components of sexual reproduction – eggs and sperm – took place during an amazing explosion of discovery in the 1660s and 1670s. And yet despite this, their respective roles remained unclear for another 170 years. Why did this take so long? And what did people think before these discoveries? By tracing the history of this major milestone in human knowledge, we can also gain insight into our current knowledge, and the boundaries we may be unwittingly trapped by.

09:40 – 10:30 R. John Aitken, University of Newcastle, AustraliaThe 2012 recipient of the Simmet Prize for Assisted ReproductionNew horizons on an old landscape - oxidative stress, DNA damage and apoptosis in the male germ line.

R. John Aitken is Laureate Professor of Biological Sciences and Director of the Priority Research Centre in Reproductive Science at the University of Newcastle. He is a Fellow of the Royal Society of Edinburgh and the Australian Academy of Science. He has published more than 450 research articles in gamete and developmental biology, which have been cited more than 14,000 times.

Abstract: Practical applications in assisted reproduction ultimately depend upon a knowledge of how the reproductive system works at the cellular level. Recent insights in this area have helped engineer improved methods for the diagnosis and management of infertility and generate innovative methods for selection and preservation of mammalian gametes.

Plenary Session 2: Post natal consequences of prenatal environment Tuesday, July 31 201208:15 – 10:30East Ballroom ABCChair: Kevin Sinclair

08:15 – 09:00Paul Fowler, University of Aberdeen, UKEffects of exposure to environmental chemicals during pregnancy on the development of the male and female reproductive axes

Paul A. Fowler (B.Sc. Hons Zoology, Ph.D) is professor of Translational

Medical Sciences and lead for the Cell, Developmental and Cancer Biology research programme at the University of Aberdeen, UK. Since the late 1990s his research has focused on the effects of environmental chemicals on fetal development and subsequent reproduction in both human pregnancy and in animal models (see: http://www.abdn.ac.uk/ims/staff/details.php?id=p.a.fowler ).

Abstract: The developing fetus is routinely exposed to a wide range of ubiquitous chemicals, both natural and anthropogenic,

09:00 – 09:45Cheryl Rosenfeld, University of Missouri, USAEffects of maternal diet on sexually dimorphic responses in conceptuses and offspring

Cheryl S. Rosenfeld, DVM, PhD is a graduate of the University of Illinois at Urbana-Champaign and the University

of Missouri. Her research emphasis is on how maternal diet and in utero exposure to endocrine disrupting compounds epigenetically alters conceptus development and offspring outcomes, including neurobehavioral abnormalities.

Abstract: While sexual differentiation is considered as the onset of differentiation of the male or female gonads, mounting evidence indicates that sex differences in developmental programming are established as early as the zygotic stage. Genetic and epigenetic differences between the sexes might govern how each responds to shifts in their early environment, including in the uterus or culture dish in the case of in vitro cultured pre-implantational embryos. Even if no differences are evident between the sexes at birth, divergent conceptus responses to surrounding changes, such as maternal diet and exposure to endocrine disrupting compounds might predispose one sex over the other to later adult onset diseases, otherwise termed developmental origin of health and disease (DOHaD). This review examines the evidence to date that male and female zygotes and conceptuses diverge in their responses to shifting environmental conditions and whether these contrasting sexually dimorphic responses underpin later DOHaD outcomes, namely neurobehavioral changes.

09:45 – 10:30 Alex Evans, University College, IrelandMaternal dietary restriction on ovarian folliculogenesis and fertility in bovine offspring

Abstract: Mammals such as cattle, swine, sheep and humans are born with a highly variable number of ovarian follicles

and oocytes that dwindle during ageing. This variation in the ovarian reserve is reflected in the numbers of antral follicles in the ovaries at all ages after birth. Since numbers of follicles in ovaries are determined during gestation, the role of maternal nutrition and health during gestation (at time of ovarian development in their fetuses) has been investigated as factors that may impact oogonia proliferation and thus follicle numbers postnatally. Both nutrition and health were shown to impact numbers of follicles in offspring and cows with a relatively high number of antral follicles were shown have higher pregnancy rates, shorter calving to conception intervals and fewer artificial inseminations compared with cows with a lower number of follicles; heifers with many follicles had higher pregnancy rates than those with fewer follicles. This review will highlight the importance of the maternal environment during gestation in determining the size of the ovarian reserve in offspring and the contribution of the ovarian reserve to subsequent fertility in cattle.

08:15 – 09:00 Phil Senger, Washington State University, USAExploiting multimedia in reproductive science education-research findings

P.L. Senger is Professor Emeritus at Washington State University and President of Current Conceptions, Inc. Dr. Senger has a BS from North Carolina

State University and a MS and PhD from Virginia Tech. He is the author of “Pathways to Pregnancy and Parturition, 2nd Rev. Ed.” Current Conceptions, Inc. is the publisher of the textbook and is currently developing multimedia content to explain complex mechanisms in reproductive science. This content is not only useful in academic venues but is intended as an educational medium to serve a wide variety of people within the general public.

Abstract: The technology exists to explain complex topics using multimedia presentations in which digital animation and 3-dimensional anatomical reconstruction greatly reduce the time required for delivery while at the same time improving learning. We have designed and produced multimedia programs to teach complex mechanisms in various topics within reproductive physiology. Demonstrations will be presented in the session. The programs have been tested in controlled experiments involving over 1,000 university students and 100 OB/GYN patients. Data from these experiments indicate that these multimedia presentations can result in “Higher Learning @ Higher Speeds”.

09:00 - 09:45James Knight Virginia Tech, USAEthics, politics and protests: Using contentious issues in reproductive sciences as educational opportunities

James W. Knight, M.S., Ph.D., is a Professor of Reproductive Physiology in the Department of Animal and Poultry Sciences at Virginia Tech, Blacksburg,

VA, U.S.A. Dr. Knight has been heavily involved in teaching and advising both undergraduate and graduate students throughout his career. Dr. Knight had been the recipient of numerous teaching awards at the university and national levels.

Abstract: Contentious issues can be utilized in the classroom to create reflective dialogue among students, leading to both a greater understanding and enhanced appreciation of alternative viewpoints. Exploring the scientific, ethical, moral, political, legal, and societal aspects of contentious reproductive issues provides ideal subject matter for developing critical thinking skills.

09:45 – 10:30Peter Chenoweth Charles Sturt University, AustraliaReproductive sciences in the Global Village

Peter Chenoweth is Adjunct Professor of Veterinary Reproduction at Charles Sturt University, Australia and current President of the Association for Applied

Animal Andrology. Books include Beef Practice: Cow-Calf Production Medicine (Blackwell 2005), and 2 volumes of Applied Animal Andrology (Nottingham University Press)

Abstract: Livestock are vitally important for developing countries, and efficient and viable animal production depends upon effective animal reproduction. Attention to areas such as health, nutrition and genetics in addition to applying principles of good reproductive management are required. Although resources are limited, progress can be made via relatively minor managerial changes at the local level of small, mixed farms.

Keynote and Plenary Sessions (Continued) Such changes are a necessary prerequisite for the application of more sophisticate approaches. Both the dissemination and acceptance of information on what constitutes good reproductive management face many challenges. This review examines a number of the issues involved in meeting these challenges including the relevant roles of rural extension, biotechnology, biosecurity, research and institutional capacity. It is concluded that current programs aimed at raising livestock productivity in developing countries would benefit from increased focus on animal reproduction.

Plenary 4Thursday, August 2, 201208:15 – 10:30East Ballroom ABCChair: John Dobrinksy

08:15 – 09:00 Eckhard Wolf, Gene Center Munich, GermanyUsing domestic animals for biomedical research

Eckhard Wolf is Head of the Institute of Molecular Animal Breeding and Biotechnology at the Gene Center Munich. His research interests are

mechanisms of embryo maternal communication and the development of tailored pig models for human diseases. Abstract: The development of new therapies requires appropriate animal models for efficacy and safety studies. Although rodent models are widely used for biomedical research, they are not always predictive for clinical outcomes in human patients. Efficient and precise techniques for the genetic modification of pigs facilitate the generation of tailored large animal models which mimic human disease mechanisms at the molecular level. This will be exemplified by our genetically engineered pigs for diabetes research, which may help to translate therapeutic principles developed in rodents into clinical application.

09:00 – 09:45Steven Stice, Regenerative Bioscience Center, Greece Animal models and regenerative medicine

Steve Stice is a Georgia Research Alliance Eminent Scholar endowed chair and professor. Stice was a co-founder and chief scientific officer at

Advanced Cell Technology, Inc., a stem cell company that is currently in clinical trials for a rare disease affecting sight. His current research activities range from human to animal stem cell applications and he will present his research on induced pluripotent stem cells.

MONDAY JULY 30

Workshops 1 - 916:45 – 18:15

Workshop 1 - Reproductive physiology underlying low fertility in lactating dairy cows and current methods for fertility improvement. Milo Wiltbank, University of Wisconsin, USA ROOM: East Ballroom B Reproductive efficiency of lactating dairy cattle is suboptimal, leading to loss of profitability.  The reduction in fertility is multifactorial, potentially involving management, nutritional, and physiological factors.  This workshop will focus on physiology that underlies the reduction in conception rates in cows bred to estrus. New synchronized ovulation programs have been developed; the underlying physiology and shortcomings will be explored.

Workshop 2 - Equine reproduction Tom Stout, Utrecht University, The Netherlands ROOM: East Meeting Room 8 This workshop will focus on conceptus/endometrium interactions.• Conceptus signal(s) responsible for triggering maternal

recognition of pregnancy• Regulation of prostaglandin synthesis and oxytocin

receptor function• Does the blastocyst capsule contribute more than

physical protection?• Mechanisms or pathways required for successful

implantation• What information has systems biology (the -omics)

provided?

Workshop 3 - In vitro embryo production Osamu Dochi, Rakuno Gakuen University, JapanROOM: East Ballroom A

This workshop will focus on improving in vitro embryo production efficiency:• Fertilization rates differ among individual bulls

• Media composition affects embryo development and quality

• Cryopreservation of embryos (pregnancy rate)• Oocyte collection by OPU• In vitro embryo production using sexed semen

Workshop 4 - Buffalo (Bubulus) reproduction Nasim Ahmad, University of Veterinary & Animal Sciences, PakistanROOM: East Meeting Room 10

The following topics will be explored:• Delayed puberty (heifers and bulls, endocrinology,

nutrition, management, growth)• Long calving interval (suckling, summer anestrous,

estrus detection, low fertility, silent heat, early embryonic loss)

• Cryopreservation and semen quality; parameters for bull selection

• Synchronization of estrus & ovulation• Reproductive biotechnology (embryo transfer, AI, IVF,

IVEP, cloning)

Workshop 5 - Genomic and proteomic tools in reproduction research Claude Robert, University of Laval, Canada ROOM: East Ballroom C

Genomics – Microarrays and RNAseq• Sample processing profoundly impacts results making

comparisons difficult • Experimental design flaws with biological and technical

replication• Data normalization is extremely difficult• Proteomics: 2D gels and mass spectrometry• Is protein requirement a dead end?• Can proteins be efficiently identified?• How to handle post-translational modifications?

Workshops (Continued)

Workshop 6 - Emerging roles of seminal plasma Puttaswamy Manjunath, University of Montréal, Canada ROOM: East Meeting Room 12 Seminal plasma is a complex mixture of secretions from the testes, epididymides and accessory sex glands. While it is believed to play a primary role as the carrier of spermatozoa to the female reproductive tract, it also contains proteins which are beneficial (promote motility, capacitation, acrosome reaction, and play a role in sperm reservoir formation in the oviduct) or detrimental (particularly to sperm storage in liquid or frozen state). In recent years, several proteins have been characterized and their physiologic significance has been studied in depth. This workshop highlights emerging roles of major proteins of seminal plasma.

Workshop 7 - Biostatistics George Seidel Jr., Colorado State University, USAROOM: East Meeting Room 7

This Workshop will consider approaches for the following topics:• How to define and analyze pulse data, e.g. LH pulses.• How to choose methods of analyzing binomial data, e.g.

pregnant/not pregnant; fertilized / not fertilized.• Receiver Operator Characteristics/curves that help

define thresholds that minimize both false positives and false negatives (e.g. what concentration of progesterone is indicative of a functional CL).

• Characteristics of competitive fertilization data and analyzing such data.

Workshop 8 - Wild ungulate reproduction Rob McCorkell, University of Calgary, CanadaROOM: East Meeting Room 13

The wild ungulates of Canada live in a temperate climate where adapting their reproductive effort to the seasons is essential for their survival. Therefore, this workshop will focus on the effect of season on reproduction.• Species differences with some discussion about recent

studies involving reindeer• The different responses to season by males and females

of the same species• The effect of season on ovarian follicle development• The effect of season on spermatogenesis

• What sheep can tell us about the effect of season on wild ungulates

Workshop 9 - Tools to study reproductive behaviour Rodolfo Ungerfeld, Facultad de Veterinaria, Uruguay ROOM: East Meeting Room 14 The workshop aims to discuss different strategies to study reproductive behaviour, and the advantages of using reproductive behaviour as a tool to be included in other studies. As a non-invasive technique, the study of reproductive behaviour provides important information on physiological status of experimental units, homogeneity of the population used for research, and information on differences among populations of the same species. The study of sexual behaviour may also provide important clues to improve management, select animals, and improve welfare status. Examples are how social hierarchies may affect cyclic activity, pregnancy outcomes, or the response to biotechnologies. Conversely, socio-sexual stimuli may induce reproductive activity in inactive individuals. A well known example is the use of the male effect, widely studied in sheep, goats and swine, but also applicable in cattle.

WEDNESDAY AUGUST 1

Workshops 10 - 1816:45 – 18:15

Workshop 10 - Use of eCG in reproductive management of beef and dairy cattle Gabriel Bo, IRAC, Argentina ROOM: East Ballroom C In the cow, eCG may demonstrate either FSH or LH activity. eCG has been shown to improve follicular development and ovulation of a larger follicle resulting in a larger, more functional CL, which has resulted in improved pregnancy rates in anestrous beef cows, pasture managed dairy cows and embryo transfer recipients. More recently, eCG given after breeding reduced embryonic loss in dairy cows and in suckled beef cows in fair to poor body condition. With conception and pregnancy rates continuing to decrease globally, this workshop will explore strategies to utilize the potential benefits of eCG in improving bovine reproduction.

Workshops (Continued)

Workshop 11 - Small ruminant reproduction: Clean green and ethical (CGE) management in the developing world? Graeme Martin, University of Western Australia, Australia ROOM: East Meeting Room 8 The CGE concept is driven by consumer demand in high-profit markets for food that is: 1) Clean – food must be safe (no chemicals, pathogens) and healthy (eg, ‘healthy fats’); 2) Green – low environmental impact; 3) Ethical – attention on animal welfare. Importantly, CGE management practices are not expensive, difficult, or ‘high tech’; it simply involves application of basic knowledge of reproductive physiology and behaviour. We will explore the possibilities for CGE practices in the developing world. For example: the ‘male effect’ to control the timing of reproduction; ‘focus feeding’ to improve reproductive outcomes; forages that are anthelmintic and reduce methane production; the power of genetics.

Workshop 12 - Canine and feline reproductionStefano Romagnoli, University of PadovaROOM: East Meeting Room 10

Workshop 13 - Camelid reproduction Marcelo Ratto, University Austral de Chile, Chile ROOM: East Meeting Room 7

The following topics will be explored:• Functional and molecular characterization of an ovulation-

inducing factor present in the seminal plasma of llamas. • Recent advances in semen manipulation and

cryopreservation in South American camelids• Oocyte collection and in vitro maturation in alpacas• The most relevant reproductive disorders present in

alpaca farms in the USA

Workshop 14 - Sperm and embryo sexingSimon de Graaf, University of Sydney, AustraliaROOM: East Ballroom B

Sperm sexing by flow cytometry has produced millions of pre-sexed offspring and remains the only method available for the sex preselection of mammalian offspring. We will explore issues relevant to research and commercial applications, with particular emphasis on cattle.

• The current status of sperm sexing and reasons behind compromised fertility and differences between species

• Commercial aspects of sexed sperm production such as sperm quality and ‘reverse sorting’

• Ideas for improving current methods of sperm sexing, and• Recent technological developments and future research

goals in the field.

Workshop 15 - Imaging in reproductionHeinrich Bollwein, Clinic for Cattle, GermanyROOM: East Meeting Room 12

The introduction of sonography represents a technological breakthrough that has provided novel knowledge about reproductive biology, especially relating to ovarian, uterine and fetal function. To improve objectivity computer-assisted software has been used for analysis of B-Mode images. The development of Doppler processing extended the scope of sonographic imaging from an anatomic to a physiologic basis, and three-dimensional ultrasound imaging allows for the determination of the volume of corpus luteum and follicles and the evaluation of the fetal development. In this workshop these techniques will be reviewed and future developments, possibilities and limitations will be discussed.

Workshop 16 - Reproductive pathologyDon Schlafer, Cornell University, USAROOM: East Meeting Room 14

Endometritis - Overview and update• Role of endometritis in female subfertility  (by species)• Uterine pathogens (viral, bacterial, yeast) and their

identification• Comparison and characterization of host tissue (uterine)

responses - gross and histopathology• Endometritis with no demonstrable cause - possible

explanations• Endometrial biopsy:  Histopathology: interpretation;

formal “grading schemes”; prognostic value • Discussion of needed research, experimental

approaches, etc.

Workshops (Continued)

Workshop 17 - Swine reproductionNicoline Soede, Wageningen University, The Netherlands ROOM: East Meeting Room 13 Although sustained selection for larger litters has been quite successful, it has changed both the physiology and management of modern, highly prolific sows. This workshop will focus on these changes, during pregnancy (numbers of embryos, pre-natal mortality, pre-natal development), lactation (piglet survival and development, sow energy balance) and post-lactation (decreased subsequent fertility) and the associated changes in management (nutrient requirements, changed weaning management, etc.). The audience is asked to participate in defining the future challenges for on farm reproductive performance and, based on those challenges, discuss directions/challenges for reproductive research.

Workshop 18 - Diagnostic tools for gametes and embryosDon Rieger, LifeGlobal, CanadaROOM: East Ballroom A Gametes and embryos are often evaluated morphologically; however, this is unreliable to determine if a particular oocyte or embryo will result in a successful pregnancy. Measurement of metabolic activity of single embryos to evaluate viability has had contradictory results, but metabolomics and amino acid metabolism have been promising. Increasing evidence suggests that the developmental potential of the embryo is largely determined by the health of the oocyte, including ability to form gap junctional complexes between cumulus cells and expression of specific genes by follicular cells. Two approaches to sperm selection - hyaluronan binding and morphological selection under high magnification are useful for production of viable human embryos using ICSI.
